耳针改善血管性痴呆大鼠记忆障碍及其与bcl-2表达的关系

摘    要:目的 :探讨耳针对血管性痴呆大鼠 (VD)学习记忆障碍的改善与bcl 2凋亡蛋白表达的关系。方法 :采用 4 血管阻断制备VD大鼠模型 ,针刺脑、肾耳穴后 ,作免疫组化、Nissl染色、行为学测试及图像分析。结果 :与正常对照比较 ,VD大鼠海马CA1 区神经元严重丢失 ,而bcl 2蛋白表达增加 ,大鼠学习记忆产生明显障碍 ;耳针治疗后海马CA1 区神经元丢失减轻 ,但bcl 2增加更为明显 ,此时大鼠学习记忆障碍得到明显改善。结论 :耳针改善VD大鼠学习记忆 ,可能是通过针刺调控细胞凋亡 ,达到对VD大鼠海马神经元的保护作用

Abstract:Objective: To study the relationship between auricular acupuncture improving dysmnesia and expression of bcl 2 protein in vascular dementia (VD) rats. Methods: 40 Wistar rats were randomly and evenly divided into control group, VD group, acupuncture+VD group and pseudo operation group. Rat VD model was established by using 4 vessel occlusion method. Otopoint “Nao” point and “Shen” (MA SC) were punctured, once daily continuously for 15 days. Rats' memory capability was tested with Y maze method and bcl 2 expression in the brain tissues displayed with immunohistochemical method and detected with MIAS 2000 Image Analyzer. Results: After auricular acupuncture treatment, the VD rats' memory was raised significantly in comparison with VD group (P<0.01). In control, VD and auricular acupuncture groups, bcl expression positive neurons in CA 1 region of the hippocampus were 14.31±4.87, 28.67±5.63 and 65.74±8.19 respectively, suggesting that the improvement of memory caused by acupuncture may be via up regulation of bcl 2 expression (an inhibitory gene of apoptosis) to lighten neuron lesion of hippocampus. Conclusion: Auricular acupuncture can raise the learning and memory capability, which may be realized by regulation of apoptosis and protecting hippocampal neurons.
