HPLC法同时测定消炎片中绿原酸和秦皮乙素的含量

目的：建立反相高效液相色谱法同时测定消炎片中绿原酸和秦皮乙素的含量。方法：采用Agilent EclipseXDB-C18（250 mm×4.6 mm,5μm）色谱柱,以乙腈-0.04%磷酸溶液为流动相,梯度洗脱;检测波长为328 nm;流速1.0 ml/min;进样量10μl;柱温35℃。结果：绿原酸和秦皮乙素的线性范围分别为2.515～30.18μg/ml（r=0.999 9,n=5）和6.388～76.65μg/ml（r=1.000 0,n=5）;提取回收率分别为98.99%（RSD为0.2%,n=6）和98.93%（RSD为0.5%,n=6）。结论：本法操作简便、快速、结果准确,可用于消炎片的质量控制。

Simultaneous determination of chlorogenic acid and esculetin in Xiaoyan Tablets by HPLC

Objective： To establish RP-HPLC method for simultaneous determination of chlorogenic acid and esculetin in Xiaoyan Tablets.Methods： Column packed with Agilent Eclipse XDB-C18（250 mm×4.6 mm,5 μm） was used;the mobile phase was acetonitrile-0.04%H3PO4 by gradient elution and the detection wavelength was 328 nm;the flow rate was 1.0 ml/min,the injection volume was 10 μl,the column temperature was 35℃.Results： The linear response of chlorogenic acid ranged from 2.515 to 30.18 μg/ml,r=0.999 9（n=5）;the linear response of esculetin ranged from 6.388 to 76.65 μg/ml,r=1.000 0（n=5）;the average recovery of chlorogenic acid was 98.99%,with RSD of 0.2%（n=6）.The average recovery of esculetin was 98.93% with RSD of 0.5%（n=6）.Conclusion： The method is convenient,quick and accurate,and can be used for the quality control of Xiaoyan Tablets.
