mPEG-SPA对红细胞A、B抗原的遮蔽及稳定性初步研究

摘    要:目的探讨mPEG-SPA修饰对A、B、AB三种血型红细胞上A、B抗原的遮蔽效果及稳定性。方法采用合适浓度的mPEG-SPA修饰不同血型红细胞,并分别保存在O型血浆中以观察红细胞凝集情况、保存在原血浆和MAP液中以测定游离血红蛋白。结果连续保存45d,显微镜下观察mPEG-SPA修饰的红细胞在O型血浆中未见凝集,在原血浆中保存21d时血浆游离血红蛋白≤0.29g/L;在MAP液中保存35d时游离血红蛋白≤1g/L。结论在保存期内,经mPEG-SPA化学修饰的A、B、AB三种血型红细胞的抗原遮蔽效果稳定、游离血红蛋白指标均符合临床输血要求。

Preliminary Study on Camouflage and Stability of A/B Antigens on mPEG-SPA Modified Red Blood Cells

Abstract:Objective To observe the camouflage effect and storage stability of A/B antigens on the mPEG-SPA modified red blood cells (RBCs) of blood type A,B or AB. Methods To modify the RBCs of blood type A,B or AB with mPEG-SPA. To store the modified RBCs in blood type O plasma,original plasma and MAP liquids respectively to check the RBC condensation and storage stability. Results In 45 days,no RBCs condensation was observed under microscope in O-type plasma; the free haemoglobin in the original plasma was no more than 0.29 g/L after 21-day preservation; the free haemoglobin in MAP liquid was no more than 1 g/L after 35-day preservation. Conclusion The measured free haemoglobin contents fully meet the requirements of clinic transfusion,the A/B antigens on the modified RBCs are stably camouflaged during the storage.
