Calorie restriction (CR) is the most efficacious treatment to delay the onset of age-related changes such as mitochondrial dysfunction. However, the sensitivity of mitochondrial markers to CR and the age-related boundaries of CR efficacy are not fully elucidated. We used liver samples from ad libitum-fed (AL) rats divided in: 18-month-old (AL-18), 28-month-old (AL-28), and 32-month-old (AL-32) groups, and from CR-treated (CR) 28-month-old (CR-28) and 32-month-old (CR-32) counterparts to assay the effect of CR on several mitochondrial markers. The age-related decreases in citrate synthase activity, in TFAM, MFN2, and DRP1 protein amounts and in the mtDNA content in the AL-28 group were prevented in CR-28 counterparts. Accordingly, CR reduced oxidative mtDNA damage assessed through the incidence of oxidized purines at specific mtDNA regions in CR-28 animals. These findings support the anti-aging effect of CR up to 28 months. Conversely, the protein amounts of LonP1, Cyt c, OGG1, and APE1 and the 4.8 Kb mtDNA deletion content were not affected in CR-28 rats. The absence of significant differences between the AL-32 values and the CR-32 counterparts suggests an age-related boundary of CR efficacy at this age. However, this only partially curtails the CR benefits in counteracting the generalized aging decline and the related mitochondrial involvement. 相似文献

Multiple lines of evidence support the pathogenic role of maternal immune activation (MIA) in the occurrence of the schizophrenia-like disturbances in offspring. While in the brain the homeostatic role of neuron-microglia protein systems is well documented, the participation of the CX3CL1-CX3CR1 and CD200-CD200R dyads in the adverse impact of MIA often goes under-recognized. Therefore, in the present study, we examined the effect of MIA induced by polyinosinic:polycytidylic acid (Poly I:C) on the CX3CL1-CX3CR1 and CD200-CD200R axes, microglial trajectory (MhcII, Cd40, iNos, Il-1β, Tnf-α, Il-6, Arg1, Igf-1, Tgf-β and Il-4), and schizophrenia-like behaviour in adult male offspring of Sprague-Dawley rats. Additionally, according to the “two-hit” hypothesis of schizophrenia, we evaluated the influence of acute challenge with Poly I:C in adult prenatally MIA-exposed animals on the above parameters. In the present study, MIA evoked by Poly I:C injection in the late period of gestation led to the appearance of schizophrenia-like disturbances in adult offspring. Our results revealed the deficits manifested as a diminished number of aggressive interactions, presence of depressive-like episodes, and increase of exploratory activity, as well as a dichotomy in the sensorimotor gating in the prepulse inhibition (PPI) test expressed as two behavioural phenotypes (MIAPPI-low and MIAPPI-high). Furthermore, in the offspring rats subjected to a prenatal challenge (i.e., MIA) we noticed the lack of modulation of behavioural changes after the additional acute immune stimulus (Poly I:C) in adulthood. The important finding reported in this article is that MIA affects the expression and levels of the neuron-microglia proteins in the frontal cortex and hippocampus of adult offspring. We found that the changes in the CX3CL1-CX3CR1 axis could affect microglial trajectory, including decreased hippocampal mRNA level of MhcII and elevated cortical expression of Igf-1 in the MIAPPI-high animals and/or could cause the up-regulation of an inflammatory response (Il-6, Tnf-α, iNos) after the “second hit” in both examined brain regions and, at least in part, might differentiate behavioural disturbances in adult offspring. Consequently, the future effort to identify the biological background of these interactions in the Poly I:C-induced MIA model in Sprague-Dawley rats is desirable to unequivocally clarify this issue. 相似文献

The development of the IEEE 802.22 standard is aimed at providing broadband access in rural areas by effectively utilizing the unused TV band, provided no harmful interference is caused to the incumbent operation. The motivation behind TV band selection is of having lower frequencies compared to other licensed bands, which, therefore, results in lower propagation path loss. Due to this quality, the spectral power density of the radio signal reduces slowly, which results in a high coverage area. Further, it has been observed that many TV channels largely remain unoccupied, as most households and businesses rely on cable and satellite TV services. This is the first international standard for a wireless regional area network (WRAN) based on cognitive radio technologies. This standard provides both PHY and MAC layer functionalities in an infrastructure based network for communication between customer premise equipments (CPEs) through a base station (BS). The Spectrum Manager is the central part of the BS, which plays a significant role in maintaining spectrum availability information, channel selection, channel management, scheduling quiet periods for spectrum sensing, accessing to the database and implementing IEEE 802.22 policies. A WRAN can particularly accommodate up to 512 CPEs in a cell. Contention may occur during initial ranging, periodic ranging, bandwidth request and urgent coexistence situation notification. The medium access control (MAC) incorporates several schemes to control contention between CPEs within a cell and overlapping cells sharing the same channel. A CPE has to make decision to resolve collisions in the upstream direction. In the case of initial ranging and periodic ranging, code division multiple access (CDMA) is employed to resolve collisions. For bandwidth and UCS notification, either a CDMA or exponential time backoff approach can be applied for collision resolution. This paper presents the analytical framework to evaluate the number of active CPEs in a cognitive radio network. It is important to note that when the arrival rate becomes equal to the service rate, the active CPEs curve attains a constant value. Further, the active CPEs length is highly dependent on service rate. The different special cases have been addressed and the effectiveness of the proposed framework has been validated through various evaluation results. 相似文献

Microstructure, mechanical properties and electrical conductivity of industrial Cu-0.5% alloy subjected to equal channel angular pressing (ECAP) by route A and cold rolling with and without aging treatment were investigated. The lamellar grains in thickness of 100 nm were obtained after eight ECAP passes. They were not further pancake shaped, but fragmentary and obtained less sharp boundaries with more dislocations in addition to cold rolling. After aging at 450 °C for 1 h, high density of dislocations and some coarse grains were observable after ECAP and the additional cold rolling, respectively. The tensile tests show that tensile strength arrived at 460 MPa and 484 MPa after four and eight passes of ECAP, respectively, the corresponding tensile strength increased to 570 MPa and 579 MPa after the additional cold rolling. However, the electrical conductivity was not more than 35% IACS. It was proved that four passes of ECAP followed by 90% cold rolling and aging at 450 °C for 1 h offered a short process for Cu-0.5%Cr alloy to balance the paradox of high strength and electrical conductivity, under which the tensile strength 554 MPa, elongation to failure 22% and electrical conductivity 84% of IACS could be obtained. This paper presents about conceptual designs of Advanced Recycling Reactor (ARR) focusing on enhancement in transuranics (TRU) burning and americium (Am) transmutation. The design has been conducted in the context of the Global Nuclear Energy Partnership (GNEP) seeking to close nuclear fuel cycle in ways that reduce proliferation risks, reduce the nuclear waste in the US and further improve global energy security. This study strives to enhance the TRU burning and the Am transmutation, assuming the development of related technologies in this study, while the ARR based on mature technologies was designed in the previous study. It has followed that the provided TRU burning core is designed to burn TRU at 28 kg/TWthh, by adding moderator pins of B4C (Enriched B-11) and the Am transmutation core will be able to transmute Am at 34 kg/TWthh, by locating Am blanket of AmN around the TRU burning core. It indicates that these concepts improve TRU burning by 40-50% than the previous core and can transmute Am effectively, keeping the void reactivity acceptable. 相似文献
