Transient receptor potential ankyrin member 1 (TRPA1) belongs to the family of thermo TRP cation channels that detect harmful temperatures, acids and numerous chemical pollutants. TRPA1 is expressed in nervous tissue, where it participates in the genesis of nociceptive signals in response to noxious stimuli and mediates mechanical hyperalgesia and allodynia associated with different neuropathies. The glutamate N-methyl-d-aspartate receptor (NMDAR), which plays a relevant role in allodynia to mechanical stimuli, is connected via histidine triad nucleotide-binding protein 1 (HINT1) and type 1 sigma receptor (σ1R) to mu-opioid receptors (MORs), which mediate the most potent pain relief. Notably, neuropathic pain causes a reduction in MOR antinociceptive efficacy, which can be reversed by blocking spinal NMDARs and TRPA1 channels. Thus, we studied whether TRPA1 channels form complexes with MORs and NMDARs that may be implicated in the aforementioned nociceptive signals. Our data suggest that TRPA1 channels functionally associate with MORs, delta opioid receptors and NMDARs in the dorsal root ganglia, the spinal cord and brain areas. These associations were altered in response to pharmacological interventions and the induction of inflammatory and also neuropathic pain. The MOR-TRPA1 and NMDAR-TRPA1 associations do not require HINT1 or σ1R but appear to be mediated by calcium-activated calmodulin. The world is on the verge of a major antibiotic crisis as the emergence of resistant bacteria is increasing, and very few novel molecules have been discovered since the 1960s. In this context, scientists have been exploring alternatives to conventional antibiotics, such as ribosomally synthesized and post-translationally modified peptides (RiPPs). Interestingly, the highly potent in vitro antibacterial activity and safety of ruminococcin C1, a recently discovered RiPP belonging to the sactipeptide subclass, has been demonstrated. The present results show that ruminococcin C1 is efficient at curing infection and at protecting challenged mice from Clostridium perfringens with a lower dose than the conventional antibiotic vancomycin. Moreover, antimicrobial peptide (AMP) is also effective against this pathogen in the complex microbial community of the gut environment, with a selective impact on a few bacterial genera, while maintaining a global homeostasis of the microbiome. In addition, ruminococcin C1 exhibits other biological activities that could be beneficial for human health, as well as other fields of applications. Overall, this study, by using an in vivo infection approach, confirms the antimicrobial clinical potential and highlights the multiple functional properties of ruminococcin C1, thus extending its therapeutic interest. 相似文献

Pure and Mn-doped lithium tantalate nanofibers, with Mn concentrations of 1%, 2.5%, and 5%, were synthesized by the electrospinning method. The morphology, microstructure, and crystal structure of as-spun and annealed composite nanofibers were characterized by scanning electron microscopy, X-ray diffraction, and transmission electron microscopy. Raman spectroscopy has shown to be a powerful tool to detect either local variations or changes of the whole structure. Position and width of one Raman line can be used as markers of a structural change. Some vibrational modes are especially associated with the site of Li or Ta ions and so, they can be affected by the introduction of dopant ions. Any damages or local changes in the microstructure can be detected by a line broadening. With the use of Raman spectroscopy, the sites where Mn ions enter the doped structures were established by recording the shift and broadening of peaks in Mn-doped structures with respect to pure lithium tantalate. Thus it was proven that Mn ions enter the Li sites for low Mn concentration and, on the other hand, for higher concentrations, the dopant substitutes Li and Ta sites. First-principles calculations were performed within the density functional theory, including lattice-dynamic calculations of the phonon modes at the zone center (Γ point), for the pure structure, to find the irreducible representation of the modes. 相似文献

The aggregation of α-synuclein into small soluble aggregates and then fibrils is important in the development and spreading of aggregates through the brain in Parkinson's disease. Fibrillar aggregates can grow by monomer addition and then break into fragments that could spread into neighboring cells. The rate constants for fibril elongation and fragmentation have been measured but it is not known how large an aggregate needs to be before fibril formation is thermodynamically favorable. This critical size is an important parameter controlling at what stage in an aggregation reaction fibrils can form and replicate. We determined this value to be approximately 70 monomers using super-resolution and atomic force microscopy imaging of individual α-synuclein aggregates formed in solution over long time periods. This represents the minimum size for a stable α-synuclein fibril and we hypothesis the formation of aggregates of this size in a cell represents a tipping point at which rapid replication occurs. 相似文献

Recently released, BS 8571:2014 standard offers an alternative for the experimental determination of fracture toughness and resistance curves of metallic materials from SE(T) specimens. Similar in stress conditions and constraint to cracked tubes, specimens in SE(T) geometry should yield less conservative toughness values than conventional high‐constraint specimens, such as SE(B) geometry. However, the convention to determine the J‐integral fracture toughness proposed in the new BS standard is different from ASTM standards. In this work, SE(T) and SE(B) specimens of similar dimensions of a tough high‐strength seamless pipe steel were tested following the BS 8571:2014 and ASTM E1820‐16 standards, respectively. Because of the different standardized definitions, SE(T) specimens yield lower fracture toughness than SE(B) specimens, which could lead to more conservative results in structural integrity analysis. This investigation also suggests the introduction of the blunting line concept in the BS 8571:2014 standard in order to minimize this problem. 相似文献

This article details the analysis of two conducting shafts operating on blowers of a tunnel dryer. A deductive analysis was applied looking toward a successful recognition of root cause(s), where based on knowledge of background, and failure characteristics, multiple hypothesis are exposed that are further confirmed or rejected according to different tests, material analysis or maintenance history. In both shafts, failure arises from inadequate repairing. In order to confirm this, the failure analysis methodology included the following steps: technical review, visual inspection, fractography, penetrating inks, metallography, optic microscopy and stress analysis. Evidence obtained through these techniques indicates a fatigue failure induced by inadequate repairing procedures; therefore, main recommendation is to take the necessary precautions in order to guarantee minimal alterations on shaft geometry or material microstructure. 相似文献
