Computational fluid dynamics (CFD) models were employed to investigate flow conditions inside a model reactor in which yield stress non‐Newtonian liquid is mobilized using submerged recirculating jets. The simulation results agree well with the experimental results of active volume in the reactor obtained using flow visualization by the authors in a previous study. The models developed are capable of predicting a critical jet velocity (vc) that determines the extent of active volume obtained due to jet mixing. The vc values are influenced both by the rheological properties of the liquid and the nozzle orientation. The liquid with higher effective viscosity leads to higher vc for a downward facing injection nozzle. However, an upward facing injection nozzle along with a downward facing suction nozzle generates enhanced complementary flow fields which overcome the rheological constraints of the liquid and lead to lower vc.  相似文献

We investigate the challenges of building an end-to-end cloud pipeline for real-time intelligent visual inspection system for use in automotive manufacturing. Current methods of visual detection in automotive assembly are highly labor intensive, and thus prone to errors. An automated process is sought that can operate within the real-time constraints of the assembly line and can reduce errors. Components of the cloud pipeline include capture of a large set of high-definition images from a camera setup at the assembly location, transfer and storage of the images as needed, execution of object detection, and notification to a human operator when a fault is detected. The end-to-end execution must complete within a fixed time frame before the next car arrives in the assembly line. In this article, we report the design, development, and experimental evaluation of the tradeoffs of performance, accuracy, and scalability for a cloud system.  相似文献

The diagnosis and treatment of prostate cancer (PCa) is a major health-care concern worldwide. This cancer can manifest itself in many distinct forms and the transition from clinically indolent PCa to the more invasive aggressive form remains poorly understood. It is now universally accepted that glycan expression patterns change with the cellular modifications that accompany the onset of tumorigenesis. The aim of this study was to investigate if differential glycosylation patterns could distinguish between indolent, significant, and aggressive PCa. Whole serum N-glycan profiling was carried out on 117 prostate cancer patients’ serum using our automated, high-throughput analysis platform for glycan-profiling which utilizes ultra-performance liquid chromatography (UPLC) to obtain high resolution separation of N-linked glycans released from the serum glycoproteins. We observed increases in hybrid, oligomannose, and biantennary digalactosylated monosialylated glycans (M5A1G1S1, M8, and A2G2S1), bisecting glycans (A2B, A2(6)BG1) and monoantennary glycans (A1), and decreases in triantennary trigalactosylated trisialylated glycans with and without core fucose (A3G3S3 and FA3G3S3) with PCa progression from indolent through significant and aggressive disease. These changes give us an insight into the disease pathogenesis and identify potential biomarkers for monitoring the PCa progression, however these need further confirmation studies.  相似文献

Near-infrared (NIR) activatable upconversion nanoparticles (UCNPs) enable wireless-based phototherapies by converting deep-tissue-penetrating NIR to visible light. UCNPs are therefore ideal as wireless transducers for photodynamic therapy (PDT) of deep-sited tumors. However, the retention of unsequestered UCNPs in tissue with minimal options for removal limits their clinical translation. To address this shortcoming, biocompatible UCNPs implants are developed to deliver upconversion photonic properties in a flexible, optical guide design. To enhance its translatability, the UCNPs implant is constructed with an FDA-approved poly(ethylene glycol) diacrylate (PEGDA) core clad with fluorinated ethylene propylene (FEP). The emission spectrum of the UCNPs implant can be tuned to overlap with the absorption spectra of the clinically relevant photosensitizer, 5-aminolevulinic acid (5-ALA). The UCNPs implant can wirelessly transmit upconverted visible light till 8 cm in length and in a bendable manner even when implanted underneath the skin or scalp. With this system, it is demonstrated that NIR-based chronic PDT is achievable in an untethered and noninvasive manner in a mouse xenograft glioblastoma multiforme (GBM) model. It is postulated that such encapsulated UCNPs implants represent a translational shift for wireless deep-tissue phototherapy by enabling sequestration of UCNPs without compromising wireless deep-tissue light delivery.  相似文献

Macrophage apoptosis, a key process in atherogenesis, is regulated by oxidation products, including hydroxyoctadecadienoic acids (HODEs). These stable oxidation products of linoleic acid (LA) are abundant in atherosclerotic plaque and activate PPARγ and GPR132. We investigated the mechanisms through which HODEs regulate apoptosis. The effect of HODEs on THP‐1 monocytes and adherent THP‐1 cells were compared with other C18 fatty acids, LA and α‐linolenic acid (ALA). The number of cells was reduced within 24 hours following treatment with 9‐HODE (p < 0.01, 30 μM) and 13 HODE (p < 0.01, 30 μM), and the equivalent cell viability was also decreased (p < 0.001). Both 9‐HODE and 13‐HODE (but not LA or ALA) markedly increased caspase‐3/7 activity (p < 0.001) in both monocytes and adherent THP‐1 cells, with 9‐HODE the more potent. In addition, 9‐HODE and 13‐HODE both increased Annexin‐V labelling of cells (p < 0.001). There was no effect of LA, ALA, or the PPARγ agonist rosiglitazone (1μM), but the effect of HODEs was replicated with apoptosis‐inducer camptothecin (10μM). Only 9‐HODE increased DNA fragmentation. The pro‐apoptotic effect of HODEs was blocked by the caspase inhibitor DEVD‐CHO. The PPARγ antagonist T0070907 further increased apoptosis, suggestive of the PPARγ‐regulated apoptotic effects induced by 9‐HODE. The use of siRNA for GPR132 showed no evidence that the effect of HODEs was mediated through this receptor. 9‐HODE and 13‐HODE are potent—and specific—regulators of apoptosis in THP‐1 cells. Their action is PPARγ‐dependent and independent of GPR132. Further studies to identify the signalling pathways through which HODEs increase apoptosis in macrophages may reveal novel therapeutic targets for atherosclerosis.  相似文献

In the present paper, nanocrystalline hierarchical ZSM-5 zeolites were successfully synthesized by the hydrothermal method in the presence of tetrapropylammonium hydroxide as a single template with the gel composition of 58SiO2:Al2O3:20TPAOH:1,500H2O. The prepared zeolite catalysts were characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, Nitrogen adsorption–desorption (BET), scanning electron microscopy (SEM) and high resolution transmission electron microscopy (HR-TEM) techniques. The formation of pure and highly crystalline ZSM-5 zeolite phase is confirmed by XRD. The IR vibration band at 550 cm?1 is assigned to the double 5-rings of MFI-type zeolites. N2 adsorption–desorption isotherms showed that the synthesized product had high BET surface area and possessed composite pore structures with both micro and mesopores. The catalytic performance of hierarchical ZSM-5 zeolite was investigated in the selective oxidation of benzyl alcohol (BzOH) with hydrogen peroxide (H2O2) under mild conditions. The results showed that the conversion of BzOH and the selectivity to benzaldehyde were about 94 and about 99 % respectively, when using 0.08 g ZSM-5 catalyst with acetonitrile as the solvent and H2O2 as the oxidant at 90 °C. This catalyst can be retrieved and reprocessed for five times without a significant loss in its activity and selectivity.  相似文献

A generic analysis/design method for prismatic plate assemblies, using exact plate stiffnesses which are transcendental functions of loading and/or vibration frequency, has long been validated by the established program . Hitherto vibrations were permitted only in analysis because design uses the Wittrick–Williams algorithm to modify the design predicted by a linear optimiser to be ‘just stable’ by scaling all layer thicknesses by F. This works because buckling loads, unlike the fundamental frequency f1, vary monotonically with F. The paper considers additionally scaling all plate breadths by Fα such that the buckling loads and f1 all vary monotonically with F. Theoretical consideration of simple limiting cases establishes bounds on the permitted values of α as 0.1≤α≤0.4. Then results for typical stiffened panels demonstrate the method to be correct and are used to investigate the value of α which is most effective in returning stabilised designs of least mass and with least computation.  相似文献
