Connexin43 (Cx43), the main gap junction and hemichannel forming protein in the urinary bladder, participates in the regulation of bladder motor and sensory functions and has been reported as an important modulator of day–night variations in functional bladder capacity. However, because Cx43 is expressed throughout the bladder, the actual role played by the detrusor and the urothelial Cx43 is still unknown. For this purpose, we generated urothelium-specific Cx43 knockout (uCx43KO) mice using Cre-LoxP system. We evaluated the day–night micturition pattern and the urothelial Cx43 hemichannel function of the uCx43KO mice by measuring luminal ATP release after bladder distention. In wild-type (WT) mice, distention-induced ATP release was elevated, and functional bladder capacity was decreased in the animals’ active phase (nighttime) when Cx43 expression was also high compared to levels measured in the sleep phase (daytime). These day–night differences in urothelial ATP release and functional bladder capacity were attenuated in uCx43KO mice that, in the active phase, displayed lower ATP release and higher functional bladder capacity than WT mice. These findings indicate that urothelial Cx43 mediated ATP signaling and coordination of urothelial activity are essential for proper perception and regulation of responses to bladder distension in the animals’ awake, active phase.  相似文献

Spectroscopic characterization of AgI-ion-mediated C-AgI-A and C-AgI-T base pairs found in primer extension reactions catalyzed by DNA polymerases was conducted. UV melting experiments revealed that C-A and C-T mismatched base pairs in oligodeoxynucleotide duplexes are specifically stabilized by AgI ions in 1:1 stoichiometry in the same manner as a C-C mismatched base pair. Although the stability of the mismatched base pairs in the absence of AgI ions is in the order C-A≈C-T>C-C, the stabilizing effect of AgI ions follows the order C-C>C-A≈C-T. However, the comparative susceptibility of dNTPs to AgI-mediated enzymatic incorporation into the site opposite templating C is dATP>dTTP≫dCTP, as reported. The net charge, as well as the size and/or shape complementarity of the metal-mediated base pairs, or the stabilities of mismatched base pairs in the absence of metal ions, would be more important than the stability of the metallo-base pairs in the replicating reaction catalyzed by DNA polymerases.  相似文献

ABSTRACT

The thermal-neutron capture cross-section (σ0) and resonance integral (I0) were measured for the 135Cs(n,γ)136Cs reaction by an activation method and mass spectrometry. Because of difficulty in the preparation of pure 135Cs samples, we used 135Cs contained as an impurity in a normally available 137Cs standard solution. An isotope ratio of 135Cs and 137Cs in a standard 137Cs solution was measured by mass spectrometry to quantify 135Cs. Cesium-135 impurity along with the 137Cs standard solution was irradiated at the hydraulic conveyer of the research reactor in the Institute for Integral Radiation and Nuclear Science, Kyoto University. Wires of Co/Al and Au/Al alloys were used as neutron monitors to measure thermal-neutron fluxes and epi-thermal Westcott’s indices at an irradiation position. A gadolinium filter was used to measure the σ0, and a value of 0.133 eV was taken as the cut-off energy. Gamma-ray spectroscopy was used to measure induced activities of 137Cs, 136Cs and monitor wires. On the basis of Westcott’s convention, the σ0 and I0 values were derived as 8.57 ± 0.25 barn, and 45.3 ± 3.2 barn, respectively. The value of σ0 obtained in the present study agreed within the limits of uncertainties with the past-reported value of 8.3 ± 0.3 barn.  相似文献

In a plant consisting of parallelized microreactors (MRs), the product quality is lowered because of a lack of flow uniformity among them when blockage occurs. It is not practical to install sensors in every MR from the viewpoint of cost when detecting the blocked MRs. In the previous study, the multiple blockage detection (MBD) method using a small number of sensors was proposed, but its performance became low when the number of sensors decreased. Here, the conventional algorithm for MBD is improved by considering the process behavior on blockage occurrence, and the effectiveness of the improved algorithm is demonstrated through a numerical case study. The effects of flow distributor types and sensor types on the MBD performance are numerically investigated.  相似文献

This research demonstrated that polymerization of aniline on cellulose produces chiroptically active composites. Polymerization of aniline in the presence of cotton fibers consisting of chiral cellulose are performed to prepare a polyaniline (PANI)/cotton composite. The polymerization is conducted at the cotton interface. The resultant PANI/cotton composite shows chiroptical activity elucidated with diffuse reflectance circular dichroism. In this reaction, textile‐surface interfacial asymmetric polymerization is performed with imprinting of chiral structure from the cotton as a natural chiroptically active polymer to the PANI. © 2014 Wiley Periodicals, Inc. Poly(3-hexylthiophene-2,5-diyl) is among the most widely used conjugated polymers for opto-electronic applications. To enhance its properties, researchers have attempted to nanostructure this polymer using various processes including breath figure arrays, nanolithography and elaborated organic synthesis. We here demonstrate a simple process to nanostructure the conjugated polymer using self-assembly with polystyrene and selective removal of one of the phases. The influence of the molecular weight of each polymer on the thin film morphology was systematically studied by atomic force microscopy. Using this approach, we observe two types of nanostructure, namely, nanoporous and nanoisland structures, of which the dimensions can be tuned by modifying the molecular weight of each polymer in the blend. This simple process introduces a cost-effective alternative to produce thin films of conjugated polymer with average nano-features from 100 nm up to 500 nm which could be used in a wide range of applications.  相似文献

SiC powder was coated with SiO2 layer by chemical vapor deposition, and the SiC(core)/SiO2(shell) composite powder was consolidated to a SiC/SiO2 composite with a mosaic microstructure by spark plasma sintering (SPS) at 1923 K for 1.8 ks. The SiC(core)/SiO2(shell) powder with a 80–100 nm thick SiO2 layer resulted in a SiC/SiO2 composite with a relative density of 97% and hardness and fracture toughness of 17.1 GPa and 8.4 MPa m1/2, respectively.  相似文献
