Weissella cibaria short-fermented liquid sourdoughs based on quinoa or amaranth flours as fat replacer in focaccia bread formulation

Production of the traditional Italian Focaccia bread (FcBread) requires reformulation to meet the new rules for healthier foods. We tested the applicability of a short-fermented (15 h) liquid sourdough (LS) (dough yield, DY, 250), obtained using quinoa (Q) or amaranth (Am) flour fermented with a Weissella cibaria strain (C43-11) producing exopolysaccharides (EPS) as fat replacer in yeast-leavened FcBread. The LSs were applied during the FcBread-making process at industrial pilot plant, reducing the added fat amount by 20% (FcBread-QLS and FcBread-AmLS) and products were compared with reference samples not containing EPS. All the products were analysed for physico-chemical properties (pH, TTA, organic acids, protein content and profile, instrumental colour), textural properties, sensory quality and glycemic index. The application of both pseudo-cereal-based W. cibaria LSs in FcBread allowed obtaining products with a reduced amount of fat, increased protein amount, mainly due to the albumin and glutenin/glutelin fractions, reduced glycemic index, improved texture and preserved in the traditional sensory profile of ‘focaccia’.

Three-level designs: Evaluation and comparison for screening purposes

Since their introduction by Box and Hunter, resolution criteria have been widely used when comparing regular fractional factorials designs. In this article, we investigate how a generalized resolution criterion can be used to assess some recently developed three-level screening designs, such as definitive screening designs (DSDs) and screening designs from weighing matrices. The aim of this paper is to capture the projection properties of those three-level screening designs, complementing the work of Deng and Tang, who used generalized resolution and minimum aberration criteria for ranking different two-level designs, particularly Plackett-Burman and other nonregular factorial designs. An advantage of generalized resolution, extended here to work on three-level designs, is that it offers a useful criterion for ranking three-level screening designs, whereas the Deng and Tang resolution is used mainly for the assessment of two-level designs. In addition, we applied a projection estimation capacity (PEC) criterion to select three-level screening designs with desirable properties. Practical examples and the best projections of the designs are presented in tables.     

Biodegradation of urea in river waters under controlled laboratory conditions

The biodegradation of urea in river waters has been evaluated under laboratory conditions. Urea will degrade to ammonia at a rate depending on the bacterial state of the river water and on the water temperature. Under normal conditions no breakdown may be expected to occur at temperatures below 8°C for 14 days contact. In river waters with a high suspended solids content, simulating extreme winter river conditions, a maximum breakdown of 3–6 per cent daily of the original urea levels was found for temperatures not exceeding 8°C during the first 7 days contact.     

Assessment of DNA damage in coal open-cast mining workers using the cytokinesis-blocked micronucleus test and the comet assay

Coal mining is one of the most important causes of environmental pollution, as large quantities of coal dust particles are emitted. Colombia-South America has large natural coal reserves and “El Cerrejón” is the world's largest open-cast mine located in the northern department of Guajira. The aim of the present study was to evaluate genotoxic effects in a population exposed to coal residues from the open-cast mine “El Cerrejón”. 100 exposed workers and 100 non-exposed control individuals were included in this study. The exposed group was divided according to different mining area activities: (i). Transport of extracted coal, (ii). Equipment field maintenance, (iii). Coal stripping and, (iv). Coal embarking. Blood samples were taken to investigate biomarkers of genotoxicity, specifically, primary DNA damage as damage index (DI), tail length and% of tail DNA using the Comet assay (alkaline version) and chromosome damage as micronucleus (MN) frequency in lymphocytes. Both biomarkers showed statistically significantly higher values in the exposed group compared to the non-exposed control group. No difference was observed between the exposed groups executing different mining activities. These results indicate that exposure to coal mining residues may result in an increased genotoxic exposure in coal mining workers. We did not find a correlation between age, alcohol consumption and service time with the biomarkers of genotoxicity. Our results are the first data of genotoxic effects induced by coal mining exposure in Colombia, and thus, contribute to the exploration of test batteries use for monitoring of exposed populations and may stimulate designing control, hygiene and prevention strategies for occupational health risk assessment in developing countries.     

The Association of Il28b Genotype with the Histological Features of Chronic Hepatitis C Is HCV Genotype Dependent

The interleukin 28B (IL28B) rs12979860 polymorphism is associated with treatment outcome in hepatitis C virus (HCV) genotype 1 and 4 patients. Its association with the histological features of chronic hepatitis C and disease severity needs further clarifications. To assess the correlation between IL28B genotype, HCV genotype and liver biopsy findings in untreated patients.

Materials and Methods

Pre-treatment liver biopsies from 335 HCV Caucasian patients (59% males, age 50 years) enrolled in the MIST study were staged for fibrosis and inflammation according to the METAVIR and the Ishak scoring systems; steatosis was dichotomized as <5% or ≥5%. IL28B was typed by Taqman Single Nucleotide Polymorphism (SNP) genotyping assay. HCV genotype was 1 in 151 (45%), 2 in 99 (30%), 3 in 50 (15%) and 4 in 35 (10%) patients. IL28B genotype was CC in 117 (34%), CT in 166 (49%) and TT in 52 (15%). At univariate analysis, the IL28B CC genotype was associated with severe portal inflammation in HCV-1 patients (CC vs. CT/TT: 86% vs. 63%, p = 0.005), severe lobular inflammation in HCV-2 patients (CC vs. CT/TT: 44% vs. 23%, p = 0.03), and less fatty infiltration in HCV-1 patients (CC vs. CT/TT: 72% vs. 51%, p = 0.02). Despite the lack of any association between IL28B and fibrosis stage, in HCV-3 patients IL28B CC correlated with METAVIR F3–F4 (CC vs. CT/TT: 74% vs. 26%, p = 0.05). At multivariate analysis, the genotype CC remained associated with severe portal inflammation in HCV-1, only (Odds Ratio (OR): 95% Confidence Interval (CI): 3.24 (1.23–8.51)). IL28B genotype is associated with the histological features of chronic hepatitis C in a HCV genotype dependent manner, with CC genotype being independently associated with severe portal inflammation.     

Influence of road markings,lane widths and driver behaviour on proximity and speed of vehicles overtaking cyclists

The proximity and speed of motor traffic passing cyclists in non-separated conditions may be so close and so great as to cause discomfort. A variety of road design and driver behaviour factors may affect overtaking speeds and distances. The investigation presented in this paper builds on previous research and fills gaps in that research by considering the presence of cycle lanes on 20 mph and 30 mph roads, different lane widths, different lane markings, vehicle type, vehicle platooning and oncoming traffic. Data were collected from a bicycle ridden a distance of one metre from the kerb fitted with an ultrasonic distance detector and forward and sideways facing cameras.
