BACKGROUND: Cultures of epidermal cells are commonly used to study skin biology and differentiation. Recently a method to culture nail matrix cells has been established. OBJECTIVE: We report the biologic characteristics of nail matrix cells in vitro compared with those of epidermal keratinocytes. METHODS: Human nail matrix cells were isolated and cultured in defined medium. Electron-microscopic examination, growth rate, integrin expression and keratin synthesis pattern were evaluated. In addition, the cells were cultured in serum-containing medium. RESULTS: Nail matrix cells appear to be larger than human epidermal keratinocytes and, at the ultrastructural level, they contain a higher euchromatin/heterochromatin ratio and a lower nucleus/cytoplasm ratio and have a higher growth rate. The synthesis of "hard" keratins was detected at all calcium concentrations. Immunofluorescence analyses showed the expression of alpha 2, alpha 3, and alpha 6 integrin subunits. When cultured in serum-containing medium, nail matrix cells produced an outgrowth of epithelium and a spontaneous migration phenomenon associated with a tendency to stratify in a semilunar area that resembles the architecture of the nail matrix. The pluristratified epithelium showed characteristic markers of nail differentiation. CONCLUSION: Culture of nail matrix cells may represent a useful model to study the biologic properties of nail structure, alterations in some nail diseases and the effects of drugs.  相似文献

Several missions with an Unmanned Aerial Vehicle (UAV) in different realistic safety, security, and rescue field tests are presented. First, results from two safety and security missions at the 2009 European Land Robot Trials (ELROB) are presented. A UAV in form of an Airrobot AR100-B is used in a reconnaissance and in a camp security scenario. The UAV is capable of autonomous waypoint navigation using onboard GPS processing. A digital video stream from the vehicle is used to create photo maps—also known as mosaicking—in real time at the operator station. This mapping is done using an enhanced version of Fourier Mellin based registration, which turns out to be very fast and robust. Furthermore, results from a rescue oriented scenario at the 2010 Response Robot Evaluation Exercises (RREE) at Disaster City, Texas are presented. The registration for the aerial mosaicking is supplemented by an uncertainty metric and embedded into Simultaneous Localization and Mapping (SLAM), which further enhances the photo maps as main mission deliveries.  相似文献

The diamondoids (nanodiamonds) diamantane and triamantane were selectively converted into diorganophosphinic acid chlorides by reacting them with phosphorus trichloride under Friedel–Crafts‐like conditions. The di‐diamondoid phosphinic acid chlorides were subsequently reduced with trichlorosilane to give the hitherto unknown corresponding di‐diamondoid phosphines. These diamondoid phosphinic acid chlorides and phosphines are of great utility as starting materials in organo‐element and coordination chemistry due to their extraordinary rigidity and steric bulk.  相似文献

A hybrid master oscillator power amplifier (MOPA) laser source has been realized by coupling a single-mode three-section distributed Bragg reflector (DBR) laser as master oscillator and a tapered power amplifier with a single lens only. A maximum continuous-wave optical output power of 3.1 W was achieved. The emission spectrum was completely determined by the DBR laser. Single longitudinal mode operation at a wavelength of /spl lambda/=1061 nm was maintained over the whole power range. Up to an output power of 1.8 W, the beam propagation factor M/sup 2/was less than 1.6.  相似文献

The major capsid protein VP1 of JC Polyomavirus assembles into pentamers that serve as a model for studying viral entry of this potentially severe human pathogen. Previously, labeling of viral proteins utilized large fusion proteins or non-specific amine- or cysteine-functionalization with fluorescent dyes. Imaging of these sterically hindered fusion proteins or heterogeneously labeled virions limits reproducibility and could prevent the detection of subtle trafficking phenomena. Here we advance the π-clamp-mediated cysteine conjugation for site-selective fluorescent labeling of VP1-pentamers. We demonstrate a one-step synthesis of a probe consisting of a bio-orthogonal click chemistry handle bridged to a perfluoro-biphenyl π-clamp reactive electrophile by a polyethylene glycol linker. We expand the scope of the π-clamp conjugation by demonstrating selective labeling of an internal, surface exposed loop in VP1. Thus, the π-clamp conjugation offers a general method to selectively bioconjugate tags-of-interest to viral proteins without impeding their ability to bind and enter cells.  相似文献

The aim of our study was to characterize functionally prejunctional neuropeptide Y (NPY) receptors in human and rabbit renal cortex, as well as in human right atrium. Segments of human atrial appendages and of human and rabbit renal cortex were preincubated with [3H]noradrenaline, superfused with Krebs-Henseleit solution and stimulated electrically in superfusion chambers. The stimulation-induced outflow of radioactivity was taken as an index of endogenous noradrenaline release. The effects of subtype-selective NPY analogs on the stimulation-induced noradrenaline release were studied. NPY, its endogenous analog, peptide YY, and its C-terminal fragment, NPY13-36, but not its analog, [Leu31,Pro34]NPY, concentration dependently (1-100 nM) inhibited [3H]noradrenaline release in all tissues studied. NPY-induced inhibition of [3H]noradrenaline release in human and rabbit kidney was abolished by pretreatment with pertussis toxin. We conclude that prejunctional inhibition of noradrenaline release in human heart and human and rabbit kidney occurs through NPY receptors of the Y2 subtype, which appear to couple to a pertussis toxin-sensitive G protein.  相似文献

The present work introduces the laser sintering as a novel and cost-effective production process for ultrapure quartz glass crucibles. By employing CO2 laser systems, providing a power output of up to 12 kW, large SiO2 crucible preforms are sintered to graded structures with no contamination and without any change in geometry.  相似文献

Mapping is an important task for mobile robots. The assessment of the quality of maps in a simple, efficient and automated way is not trivial and an ongoing research topic. Here, a new approach for the evaluation of 2D grid maps is presented. This structure-based method makes use of a topology graph, i.e., a topological representation that includes abstracted local metric information. It is shown how the topology graph is constructed from a Voronoi diagram that is pruned and simplified such that only high level topological information remains to concentrate on larger, topologically distinctive places. Several methods for computing the similarity of vertices in two topology graphs, i.e., for performing a place-recognition, are presented. Based on the similarities, it is shown how subgraph-isomorphisms can be efficiently computed and two topology graphs can be matched. The match between the graphs is then used to calculate a number of standard map evaluation attributes like coverage, global accuracy, relative accuracy, consistency, and brokenness. Experiments with robot generated maps are used to highlight the capabilities of the proposed approach and to evaluate the performance of the underlying algorithms.  相似文献

The present work presents first results obtained with a new semi-solid freeform fabrication technique for ceramic products. It is a combination of three-dimensional (3D) surface milling and laser surface sintering (3D-LSS). By using the 3D-LSS technique, complex-shaped vitreous silica prototypes with high durability have been constructed.  相似文献
