Oligo(p-phenyleneethynylene)s (OPEs) end-capped with (alkynyl)bis(diphosphine)ruthenium and thiol/thiolate groups stabilize ca. 2 nm diameter gold nanoparticles (AuNPs). The morphology, elemental composition and stability of the resultant organometallic OPE/AuNP hybrid materials have been defined using a combination of molecular- and nano-material chacterization techniques. The hybrids display long-term stability in solution (more than a month), good solubility in organic solvents, reversible ruthenium-centered oxidation, and transparency beyond 800 nm, and possess very strong nonlinear absorption activity at the first biological window, and unprecedented two-photon absorption activity in the second biological window (σ2 up to 38,000 GM at 1,050 nm).

Efforts to engineer recombinant antibodies for specific diagnostic and therapy applications are time consuming and expensive, as each new recombinant antibody needs to be optimized for expression, stability, bio-distribution, and pharmacokinetics. We have developed a new way to construct recombinant antibody-like “devices” by using a bottom-up approach to build them from well-behaved discrete recombinant antibody domains or “parts”. Studies on antibody structure and function have identified antibody constant and variable domains with specific functions that can be expressed in isolation. We used the SpyTag/SpyCatcher protein ligase to join these parts together, thereby creating devices with desired properties based on summed properties of parts and in configurations that cannot be obtained by using genetic engineering. This strategy will create optimized recombinant antibody devices at reduced costs and with shortened development times.  相似文献

Complementary advances in medical imaging, vascular biology and biomechanics promise to enable computational modelling of abdominal aortic aneurysms to play increasingly important roles in clinical decision processes. Using a finite-element-based growth and remodelling model of evolving aneurysm geometry and material properties, we show that regional variations in material anisotropy, stiffness and wall thickness should be expected to arise naturally and thus should be included in analyses of aneurysmal enlargement or wall stress. In addition, by initiating the model from best-fit material parameters estimated for non-aneurysmal aortas from different subjects, we show that the initial state of the aorta may influence strongly the subsequent rate of enlargement, wall thickness, mechanical behaviour and thus stress in the lesion. We submit, therefore, that clinically reliable modelling of the enlargement and overall rupture-potential of aneurysms may require both a better understanding of the mechanobiological processes that govern the evolution of these lesions and new methods of determining the patient-specific state of the pre-aneurysmal aorta (or correlation to currently unaffected portions thereof) through knowledge of demographics, comorbidities, lifestyle, genetics and future non-invasive or minimally invasive tests.  相似文献

Experimental studies were performed with a 4.0 in. (10.2 cm.) diameter extraction column containing ten dualflow type trays and operated in a controlled cycling mode. The systems methyl isobutyl ketone/acetic acid/water and toluene/acetone/water were used. The overall stage efficiency was found to be a function of the ratio of the volume of a phase transferred during a cycle to the volume of that phase held up on the tray, of the feed-to-solvent ratio, and of the phase throughput rates. A non-equilibrium model was developed to represent the experimental data. For the HIBK/acetic acid/water system, overall stage efficiencies were in the range of 30 to 38 percent, corresponding to HETS values of 0.46 to 0.55 meters. For the toluene/acetone/water system, overall stage efficiencies were in the range of 20 to 31 percent, corresponding to HETS values of 0.55 to 0.95 meters.  相似文献

The assembly of multifunctional nanocomposite materials is demonstrated by exploiting the molecular sieving property of SBA‐16 nanoporous silica and using it as a template material. The cages of the pore networks are used to host iron oxide magnetic nanoparticles, leaving a pore volume of 0.29 cm3 g?1 accessible for drug storage. This iron oxide–silica nanocomposite is then functionalized with amine groups. Finally the outside of the particle is decorated with antibodies. Since the size of many protein molecules, including that of antibodies, is too large to enter the pore system of SBA‐16, the amine groups inside the pores are preserved for drug binding. This is proven using a fluorescent protein, fluorescein‐isothiocyanate‐labeled bovine serum albumin (FITC‐BSA), with the unreacted amine groups inside the pores dyed with rhodamine B isothiocyanate (RITC). The resulting nanocomposite material offers a dual‐targeting drug delivery mechanism, i.e., magnetic and antibody‐targeting, while the functionalization approach is extendable to other applications, e.g., fluorescence–magnetic dual‐imaging diagnosis.  相似文献

BACKGROUND: This research investigated the effect of platinum (Pt) on the reactivity of tungsten oxide (WO3) for the visible light photocatalytic oxidation of dyes. RESULTS: Nanocrystalline tungsten oxide (WO3) photocatalysts were synthesised by a sol‐gel process and employed for the photocatalytic degradation of Methyl Orange under visible light. For comparison commercial bulk WO3 materials were also studied for the same reaction. These materials were fully characterised using X‐ray diffraction (XRD), UV‐visible diffuse reflection spectroscopy and transmission electron microscopy (TEM). The photocatalytic oxidation of iso‐propanol was used as a model reaction to follow the concomitant reduction of molecular oxygen. No reactions occured in the absence of platinum, which is an essential co‐catalyst for the multi‐electron reduction of oxygen. The platinised WO3 catalysts were stable for multiple oxidation–reduction cycles. The results from the catalytic activity measurements showed that platinised nanocrystalline WO3 is a superior oxidation photocatalyst when compared with bulk WO3. Methyl Orange was completely decolourised in 4 h. CONCLUSIONS: The enhanced performance of nanocrystalline Pt‐WO3 is attributed to improved charge separation in the nanosized photocatalyst. Platinum is an essential co‐catalyst to reduce oxygen. This photocatalyst could be applied to the treatment of organic pollutants in wastewater, with the advantage of using visible light compared with the widely studied TiO2, which requires UV light. Digital dermatitis is an infectious disease of cattle and the leading cause of lameness. This disease is complicated by the reoccurrence of the lesions and the observation of lesions on more than one limb at different time points, indicating infection may not result in a protective immune response. The objective of this study was to characterize the peripheral blood cellular response in naturally infected and naïve cattle to bacterial antigens derived from pathogens associated with digital dermatitis lesions. Peripheral blood mononuclear cells were isolated from dairy cattle identified as having active or chronic lesions during routine hoof-trimming. Following bacterial antigen stimulation, cells were analyzed for proliferation and phenotype by flow cytometry, and culture supernatants were analyzed for IFN-γ secretion. Digital-dermatitis-infected animals had greater serum antibody titers to treponemal antigens, higher percentages of proliferating CD8+, γδ-T cells, and B cells, and increased IFN-γ secretion in vitro when compared with responses of naïve animals. No increase in proliferation of CD4+ T cells was detected in infected or naïve cattle. Although CD8+ and γδ-T cell responses may be antigen specific, the memory nature or long-lived response is yet unknown. The lack of responsiveness of CD4+ memory cells to treponemal antigens could explain the high rate of reoccurrence of digital dermatitis in infected animals.  相似文献

Preliminary experimental results are reported for the structure of the flow due to the collision of a pair of boundary layers driven towards each other by buoyant forces along a vertical wall. Of the possible parameters characterizing the flow, a Rayleigh number ratio, R, and a dimensionless temperature difference, n, emerge as important. When 0.6 < R 2.0: if n < 0.20, the approaching streams merge in a two-dimensional streamlined manner; if n > 0.20 “fingering” of fluid between the streams and cross-stream secondary flows occur.  相似文献

The authors report the first low-noise InP/InGaAs heterostructure bipolar transistor (HBT). Minimum noise figures of 0.46, 2.0, and 3.33 dB were measured at 2, 10, and 18 GHz, respectively. The noise performance of this InP/InGaAs HBT with an emitter size of 3.5×3.5 μm2 is compared to that for FETs having a 1-μm gate length. The measured minimum noise figures agree well with calculated data using a modified Hawkins model. Broadband low-noise operation is observed because of the short transit time for injected nonequilibrium electrons to transverse the base and collector depletion region  相似文献
