The Impact of the ‘Mis-Peptidome’ on HLA Class I-Mediated Diseases: Contribution of ERAP1 and ERAP2 and Effects on the Immune Response

The strong association with the Major Histocompatibility Complex (MHC) class I genes represents a shared trait for a group of autoimmune/autoinflammatory disorders having in common immunopathogenetic basis as well as clinical features. Accordingly, the main risk factors for Ankylosing Spondylitis (AS), prototype of the Spondyloarthropathies (SpA), the Behçet’s disease (BD), the Psoriasis (Ps) and the Birdshot Chorioretinopathy (BSCR) are HLA-B*27, HLA-B*51, HLA-C*06:02 and HLA-A*29:02, respectively. Despite the strength of the association, the HLA pathogenetic role in these diseases is far from being thoroughly understood. Furthermore, Genome-Wide Association Studies (GWAS) have highlighted other important susceptibility factors such as Endoplasmic Reticulum Aminopeptidase (ERAP) 1 and, less frequently, ERAP2 that refine the peptidome presented by HLA class I molecules to CD8⁺ T cells. Mass spectrometry analysis provided considerable knowledge of HLA-B*27, HLA-B*51, HLA-C*06:02 and HLA-A*29:02 immunopeptidome. However, the combined effect of several ERAP1 and ERAP2 allelic variants could generate an altered pool of peptides accounting for the “mis-immunopeptidome” that ranges from suboptimal to pathogenetic/harmful peptides able to induce non-canonical or autoreactive CD8⁺ T responses, activation of NK cells and/or garbling the classical functions of the HLA class I molecules. This review will focus on this class of epitopes as possible elicitors of atypical/harmful immune responses which can contribute to the pathogenesis of chronic inflammatory diseases.     

Plant crown traits and carbon sequestration capability by Platanus hybrida Brot. in Rome

Measurements of carbon dioxide (CO₂) concentration carried out in the city of Rome in the period January–December 2005 showed a mean yearly CO₂ concentration of 414 ± 45 ppm; polluted sites (P sites) had 21% higher CO₂ concentration than control sites (C sites). The significant (p < 0.01) correlation analysis between CO₂ concentration and traffic density measured during the study period at P sites showed that traffic density explained 51% of the CO₂ variation. The CO₂ trend during the traffic limitation days (provision imposed by ordinance of the City Council) did not show significant differences as regards regular traffic days, suggesting the ineffectiveness of this provision. Leaf area index (LAI) of Platanus hybrida Brot. was on an average 11% lower at P sites than at C ones associated with a lower total photosynthetic leaf surface area (SPT, 34% lower at P sites than at C ones). P. hybrida had a total carbon sequestration of 117 ± 13 kg year⁻¹ (mean value of P and C sites) playing an important role in sequestering CO₂. Thus, the choice of plant species for urban areas may be set out taking into account their own air amelioration capability. Plant traits of each species may be used for urban tree planting programs to ameliorate urban air pollution.     

Options and overbooking strategy in the management of wireless spectrum

Dynamic spectrum management makes it possible for the owner of usage rights on some frequency blocks to sublet each of them in real time and for a limited period of time. As a softer implementation with respect to the spot market a two stage assignment is here proposed through the use of options, which give buyers the right to purchase the usage right on a single block and for a timeslot. In the sale of options the primary owner may accomplish an overbooking strategy, which consists in selling more blocks than the available ones, and acts as a hedging tool against the risk of unsold blocks. A model for the overbooking strategy is described and evaluated, which takes into account both the value of the option, the correlated decisions taken by the prospective purchasers, and the penalty to be paid to the unsatisfied customers. The dependence of the economical convenience of the overbooking strategy on the relevant parameters (among which the penalty value and the overbooking ratio) is shown for a significant range of cases.

Growth,nutritional quality and safety of oysters (Crassostrea gigas) cultured in the lagoon of Venice (Italy)

Quality aspects of oysters (Crassostrea gigas) from a suspended culture in the lagoon of Venice (Valle Dogà) were examined in different seasons over a 1‐year period. Ecophysiological and commercial quality indicators (condition index, content of meat, shell and intervalvar fluid), nutritional quality parameters (proximate and mineral composition, glycogen content, fatty acid profile, cholesterol, plant sterols, fat‐soluble vitamins content) and levels of organic pollutants (polychlorinated biphenyls and organochlorine pesticides) were determined at different times of the year. Seasonal variations were observed in the nutrient content, with particular regard to moisture (ranging from 866.8 g kg^?1 in June to 938.8 g kg^?1 in September), protein (23.9 g kg^?1 in September to 76.6 g kg^?1 in June), ash (22.5 g kg^?1 in February to 29.5 g kg^?1 in July), lipid (3.0 g kg^?1 in September to 8.8 g kg^?1 in June) and glycogen (0.7 g kg^?1 in September to 11.5 g kg^?1 in February). In spite of this variability, the nutritional quality of the oysters was generally good, especially just before gamete release when the concentration of nutrients was at its maximum. Low levels of organochlorine chemicals were detected in the edible meat of oysters but, because only a limited number of samples were analysed, no general conclusion can be drawn on the safety of seafood from this area. Advances in cultivar choice,hazelnut orchard management,and nut storage to enhance product quality and safety: an overview

European hazelnut (Corylus avellana L.) is a major species of interest for nutritional use within the Betulaceae family and its nuts are widely used throughout the world in the chocolate, confectionery, and bakery industries. Recently its cultivation has been expanded in traditional producer countries and established in new places in the southern hemisphere, including Chile, South Africa, and Australia. Introducing hazelnut in new environments could reduce its productivity, lead the trees to experience eco‐physiological disorders, and expose the crop to high pressure from common and new pests and diseases. Thus, new approaches in cultivar choice guidance, in the sustainable orchard management and even in nut storage and kernel quality evaluation are urgently required to improve the hazelnut production and processing chain. The main objective of this study was to systematize the published information regarding recent findings about the cultural operations that directly influence nut and kernel quality, support varietal choice for new plantations, and list the recent advances in nut storage and in quality and safety evaluation. © 2020 Society of Chemical Industry

Forensic surface metrology: tool mark evidence

Over the last several decades, forensic examiners of impression evidence have come under scrutiny in the courtroom due to analysis methods that rely heavily on subjective morphological comparisons. Currently, there is no universally accepted system that generates numerical data to independently corroborate visual comparisons. Our research attempts to develop such a system for tool mark evidence, proposing a methodology that objectively evaluates the association of striated tool marks with the tools that generated them. In our study, 58 primer shear marks on 9?mm cartridge cases, fired from four Glock model 19 pistols, were collected using high-resolution white light confocal microscopy. The resulting three-dimensional surface topographies were filtered to extract all "waviness surfaces"-the essential "line" information that firearm and tool mark examiners view under a microscope. Extracted waviness profiles were processed with principal component analysis (PCA) for dimension reduction. Support vector machines (SVM) were used to make the profile-gun associations, and conformal prediction theory (CPT) for establishing confidence levels. At the 95% confidence level, CPT coupled with PCA-SVM yielded an empirical error rate of 3.5%. Complementary, bootstrap-based computations for estimated error rates were 0%, indicating that the error rate for the algorithmic procedure is likely to remain low on larger data sets. Finally, suggestions are made for practical courtroom application of CPT for assigning levels of confidence to SVM identifications of tool marks recorded with confocal microscopy.     

Morphometric analysis of cryofixed muscular tissue for intraoperative consultation

For diagnostic purposes, cryofixation of tissues is a daily routine technique to investigate rapidly about the presence of tumours during a surgical procedure in patients. We performed morphometric analysis of cryofixed muscular tissues according to different techniques. About 1,000 muscle fibers and 1,493 nuclei, were automatically examined. After freezing, ice tissue interfaces shrinkage of the cells were present. Liquid isopentane or liquid nitrogen produced a statistical increase of fractal dimension, D, of the ice‐tissue interfaces, P < 0.001 respect to the formalin‐fixed samples, cryofixation performed inside the cryostat chamber at t = ?20°C produced a D value close to the formalin‐fixed samples. Shrinkage of the muscle fibers was higher in the samples cryofixed inside the cryostat chamber (P < 0.001). Cryofixation inside cryostat or by liquid nitrogen caused decreases of the nuclei dimensions and altered nuclear morphology (P < 0.01), liquid isopentane appeared not affecting the nuclei of the fibers. Cryofixation inside the cryostat chamber produced the highest shrinkage but it was reduced performing cryofixation in liquid nitrogen or isopentane. Freezing damage inside the muscle cells was absent in the samples cryofixed inside the cryostat, it was present after cryofixation by liquid nitrogen or isopentane. Subcellular components like the nuclei were preserved by isopentane. This paper present, for the first time, an objective method able to quantify and characterize the damages produced by cryofixation in biological sample for intraoperative consultation. Microsc. Res.
