Chronic infection with Helicobacter pylori increases risk of gastric diseases including gastric cancer. Despite development of a robust immune response, H. pylori persists in the gastric niche. Progression of gastric inflammation to serious disease outcomes is associated with infection with H. pylori strains which encode the cag Type IV Secretion System (cag T4SS). The cag T4SS is responsible for translocating the oncogenic protein CagA into host cells and inducing pro-inflammatory and carcinogenic signaling cascades. Our previous work demonstrated that nutrient iron modulates the activity of the T4SS and biogenesis of T4SS pili. In response to H. pylori infection, the host produces a variety of antimicrobial molecules, including the iron-binding glycoprotein, lactoferrin. Our work shows that apo-lactoferrin exerts antimicrobial activity against H. pylori under iron-limited conditions, while holo-lactoferrin enhances bacterial growth. Culturing H. pylori in the presence of holo-lactoferrin prior to co-culture with gastric epithelial cells, results in repression of the cag T4SS activity. Concomitantly, a decrease in biogenesis of cag T4SS pili at the host-pathogen interface was observed under these culture conditions by high-resolution electron microscopy analyses. Taken together, these results indicate that acquisition of alternate sources of nutrient iron plays a role in regulating the pro-inflammatory activity of a bacterial secretion system and present novel therapeutic targets for the treatment of H. pylori-related disease.  相似文献

Cable‐shaped supercapacitors (SCs) have recently aroused significant attention due to their attractive properties such as small size, lightweight, and bendability. Current cable‐shaped SCs have symmetric device configuration. However, if an asymmetric design is used in cable‐shaped supercapacitors, they would become more attractive due to broader cell operation voltages, which results in higher energy densities. Here, a novel coil‐type asymmetric supercapacitor electrical cable (CASEC) is reported with enhanced cell operation voltage and extraordinary mechanical‐electrochemical stability. The CASECs show excellent charge–discharge profiles, extraordinary rate capability (95.4%), high energy density (0.85 mWh cm−3), remarkable flexibility and bendability, and superior bending cycle stability (≈93.0% after 4000 cycles at different bending states). In addition, the CASECs not only exhibit the capability to store energy but also to transmit electricity simultaneously and independently. The integrated electrical conduction and storage capability of CASECS offer many potential applications in solar energy storage and electronic gadgets.  相似文献

Phase behaviour modelling of reservoir fluid is a fundamental step for reservoir simulation. Furthermore, as the complexity of the recovery process increases, the fluid model plays a more important role in the reliability of the simulation outputs. Although the in situ combustion enhanced oil recovery method (ISC) is one of the most complex recovery techniques available in the petroleum engineering literature, for most of the simulation jobs related to this elaborate process only simple and rudimentary fluid characterization layouts are considered. In this work, the principal fluid properties of Athabasca bitumen with regard to the ISC process are recognized, extracted from the literature, validated for consistency, and used for the development of an inclusive and accurate fluid model. Then the fluid model is fully developed while considering the ISC reaction kinetics so that the model has both accuracy, indispensable for phase behaviour modelling, and consistency, essential for the reactions definitions.  相似文献

A review was undertaken of (a) the levels and prevalence of Salmonella spp., Campylobacter spp., Listeria monocytogenes, Escherichia coli 0157. Cryptosporidium parvum and Giardia intestinalis in livestock manure, and (b) factors which affect their survival during storage and following land application. Pathogens are commonly present in livestock manures and can survive in soil for several months or years after spreading or excretion onto land. Temperature was identified as being the most important factor influencing pathogen survival, although pH, UV light and drying were also important. Various practical measures to minimise the risk of pathogen transfer into the food chain were identified, including treatment, extended storage and no/harvest periods following land spreading. Guidelines are being developed on managing farm manures to minimise the risks of microbiological contamination of ready-to-eat crops.  相似文献

Chemical models have been used to examine the adsorption behaviour of Cu, Mn and Zn onto freshly precipitated iron oxyhydroxide as a function of pH and salinity. Solutions of varying salinity (S = 0, 5, 10, 15‰) containing 0.05 mM Fe and 1.5 μM of each metal were allowed to attain equilibrium under carefully controlled conditions. The solutions were analysed for the dissolved metals (passing a 0.45 μm filter) over the pH range 3.5–10. The pH of the onset of adsorption followed the order Cu < Zn < Mn and it occurred at pHs well below the hydrolysis of the cations in the bulk solution. Hypotheses on the metal uptake involving coulombic interactions between the precipitate and the metal species in solution did not fit the observations and it was concluded that a detailed knowledge of surface complexation reactions was required, as was shown for Cu. The adsorption isotherms for Cu were independent of salinity, on the other hand, those for Zn and Mn showed an increase in the pH of the adsorption edge with increases in salinity. It was suggested that the major cations, Ca2+ and Mg2+, were probably co-adsorbed and competition from these species for adsorption sites increased with increasing salinity. The results reported here are useful for the development of predictive models for the fate of trace metals injected into estuarine systems where iron compounds may be freshly precipitated.  相似文献

Treatment of atrazine in nursery irrigation runoff by a constructed wetland   总被引:6,自引:0,他引:6  
To investigate the treatment capability of a surface flow wetland at a container nursery near Portland, Oregon, atrazine was introduced during simulated runoff events. Treatment efficiency was evaluated as the percent atrazine recovered (as percent of applied) in the water column at the wetland's outlet. Atrazine treatment efficiency at the outlet of the constructed wetland during a 7-d period ranged from 18-24% in 1998 (experiments 1-3) and 16-17% in 1999 (experiments 4 and 5). Changes in total flow, or frequency and intensity of runoff events did not affect treatment. For experiment 6 in 1999, where the amount, frequency, and duration of runoff events exceeded all other experiments, treatment was compromised. For all experiments, deethylatrazine (DEA) and deisopropylatrazine (DIA) accounted for 13-21% of the initial application. Hydroxyatrazine (HA) was rarely detected in the water. Organic carbon adsorption coefficients (Koc) were determined from batch equilibrium sorption isotherms with wetland sediment, and they decreased in the order of HA > DIA > atrazine > DEA. Static water-sediment column experiments indicated that sorption is an important mechanism for atrazine loss from water passing through the constructed wetland. The results of the MPN assay indicated the existence in the wetland of a low-density population of microorganisms with the potential to mineralize atrazine's ethyl side chain.  相似文献

The concentrations of total mercury in the sediments, water and fish of Giauque Lake and Thompson Lake in northern Canada were determined during 1977 and 1978. Both lakes had formerly received mercury-laden industrial discharges, which were terminated in 1968 (Giauque Lake) and 1949 (Thompson Lake). In Giaque Lake, lake trout (Salvelinus namaycusy) contained an average of 3.79 mg kg−1 in their muscle while the average concentrations in northern pike (Esox lucius) and round whitefish (Prosopium cylindraceum) were 1.75 and 1.22 mg kg−1 respectively. About 7% of the lake's bottom contained mercury levels of > 500 μg kg−1 (dry weight) but, in the water, concentrations were always below detectable limits (0.2 μg l−1). In Thompson Lake, mercury levels in northern pike averaged 1.69 mg kg−1, whereas in lake whitefish (Coregonus clupeaformis), the highest recorded value was 0.6 mg kg−1. Although mercury was usually not detectable in the water column, there was heavy contamination of the sediments, with values reaching 1300 μg kg−1. In addition, approximately 15% of the lake bottom contained mercury in excess of 500 μg kg−1. Based on these data, it is concluded that: (1) northern pike are still accumulating mercury from exposed tailings which were deposited in Thompson Lake 30 years ago, and (2) contamination of only a small part of a lake may result in high levels in fish throughout the lake: this is probably due to the movement of fish from contaminated to noncontaminated areas.  相似文献

The success of the design and build (D&B) procurement route could be undermined by issues arising from the rigid professional cultures of individual participants within project workgroups. These have the potential to inhibit the achievement of a key espoused benefit of D&B procurement, i.e. that it promotes the integration of the design and construction processes for improved project performance. Cultural non-interoperability is identified as a significant potential barrier to effective change management, and to the achievement of innovation within the design and construction processes. This note argues that project responsibilities, which currently are delineated along professional identity lines, produce design and construction solutions that fail to fulfil the potential of D&B procurement. It is suggested that addressing cultural interoperability will require a fundamental and long term reshaping of the industry's structure, beginning with the professional bodies and the higher education system that underpins their future membership.  相似文献
