Barrier dysfunction of airway epithelium contributes to the development of allergies, airway hyper-responsiveness and immunological respiratory diseases. Short-chain fatty acids (SCFA) enhance and restore the barrier function of the intestinal epithelium. This study investigated whether acetate, propionate and butyrate enhance the integrity of bronchial epithelial cells. Differentiating human bronchial epithelial cells (16HBE) grown on transwells were exposed to butyrate, propionate and acetate while trans-epithelial electrical resistance was monitored over time. Restorative effects of SCFA were investigated by subsequent incubation of cells with IL-4, IL-13 or house dust mite extract and SCFA. SCFA effects on IL-4-induced cytokine production and the expression of zonula occludens-1 (ZO-1) and Mitogen-activated protein kinases (MAPK) signalling pathways were investigated by ELISA and Western blot assays. Propionate and butyrate enhanced the barrier function of differentiating 16HBE cells and induced complete recovery of the barrier function after exposure to the above-mentioned stimuli. Butyrate decreased IL-4-induced IL-6 production. IL-4 decreased ZO-1 protein expression and induced phosphorylation of extracellular signal-regulated protein kinases 1/2 (ERK1/2) and c-Jun N-terminal kinases (JNK) in 16HBE cells, both of which could be restored by SCFA. SCFA showed prophylactic and restorative effects on airway epithelial barrier function, which might be induced by increased ZO-1 expression.  相似文献

Since solid, non-metallic inclusions influence considerably the quality of casted steel products, carbon-bonded alumina foam filters are used in secondary metallurgical treatments to remove these particles from steel melts. In order to attain a significant improvement of the filtration process, five different carbonaceous spinel compounds from the Fe-/Mg-/Mn-Al-O systems are applied on carbon-bonded alumina filters in this study and investigated with regard of their filtration efficiency. However, these spinel compounds decompose partially during sintering at 1400?°C under reducing atmosphere, wherefore the resulting coatings contain not only spinel compounds, but also oxidic and metallic components. The subsequent interaction with molten steel leads to the development of multicrystal structures on the filter surface, which stem from interfacial reactions between coating materials, molten steel, and inclusions. As a result of this procedure, a reduction of almost 60% alumina inclusions is measured with the aid of an automatic SEM, whereby spinel compounds from the Fe-Mn-Al-O system achieve highest filtration efficiencies.  相似文献

Many therapeutic peptides can still be improved with respect to target specificity, target affinity, resistance to peptidases/proteases, physical stability, and capacity to pass through membranes required for oral delivery. Several modifications can improve the peptides’ properties, in particular those that impose (a) conformational constraint(s). Screening of constrained peptides and the identification of hits is greatly facilitated by the generation of genetically encoded libraries. Recent breakthrough bacterial, phage, and yeast display screening systems of ribosomally synthesized post-translationally constrained peptides, particularly those of lanthipeptides, are earning special attention. Here we provide an overview of display systems for constrained, genetically encoded peptides and indicate prospects of constrained peptide-displaying phage and bacterial systems as such in vivo.  相似文献

This article reports the preparation and characterization of multiwalled carbon nanotubes (MWCNTs)‐filled thermoplastic polyurethane–urea (TPUU) and carboxylated acrylonitrile butadiene rubber (XNBR) blend nanocomposites. The dispersion of the MWCNTs was carried out using a laboratory two roll mill. Three different loadings, that is, 1, 3, and 5 wt % of the MWCNTs were used. The electron microscopy image analysis proves that the MWCNTs are evenly dispersed along the shear flow direction. Through incorporation of the nanotubes in the blend, the tensile modulus was increased from 9.90 ± 0.5 to 45.30 ± 0.3 MPa, and the tensile strength at break was increased from 25.4 ± 2.5 to 33.0 ± 1.5 MPa. The wide angle X‐ray scattering result showed that the TPUU:XNBR blends were arranged in layered structures. These structures are formed through chemical reactions of ? NH group from urethane and urea with the carboxylic group on XNBR. Furthermore, even at a very low loading, the high degree of nanotubes dispersion results in a significant increase in the electrical percolation threshold. © 2014 Wiley Periodicals, Inc. J. Appl. The thermoelectric properties of melt-processed nanocomposites consisting of a polycarbonate (PC) thermoplastic matrix filled with commercially available carboxyl (–COOH) functionalized multi-walled carbon nanotubes (MWCNTs) were evaluated. MWCNTs carrying carboxylic acid moieties (MWCNT-COOH) were used due the p-doping that the carboxyl groups facilitate, via electron withdrawing from the electron-rich π-conjugated system. Preliminary thermogravimetric analysis (TGA) of MWCNT-COOH revealed that the melt-mixing was limited at low temperatures due to thermal decomposition of the MWCNT functional groups. Therefore, PC was mixed with 2.5 wt% MWCNT-COOH (PC/MWCNT-COOH) at 240 °C and 270 °C. In order to reduce the polymer melt viscosity, a cyclic butylene terephthalate (CBT) oligomer was utilized as an additive, improving additionally the electrical conductivity of the nanocomposites. The melt rheological characterization of neat PC and PC/CBT blends demonstrated a significant decrease of the complex viscosity by the addition of CBT (10 wt%). Optical and transmission electron microscopy (OM, TEM) depicted an improved MWCNT dispersion in the PC/CBT polymer blend. The electrical conductivity was remarkably higher for the PC/MWCNT-COOH/CBT composites compared to the PC/MWCNT-COOH ones. Namely, the PC/MWCNT-COOH/CBT processed at 270 °C exhibited the best values with electrical conductivity; σ = 0.05 S/m, Seebeck coefficient; S = 13.55 μV/K, power factor; PF = 7.60 × 10−6μW/m K−2, and thermoelectric figure of merit; ZT = 7.94 × 10−9. The PC/MWCNT-COOH/CBT nanocomposites could be ideal candidates for large-scale thermal energy harvesting, even though the presently obtained ZT values are still too low for commercial applications.  相似文献

Visco-elastic and dielectric spectra of multiwalled carbon-nanotube reinforced silicon elastomer nanocomposites were used to study relaxation behavior. SEM photomicrographs shows well dispersion of MWCNT in elastomer matrix. In visco-elastic analysis primary relaxation was studied as a function of temperature (?100 to 100 °C) at frequency 1Hz and strain 1 %. The effect of MWCNT loadings on storage modulus, loss modulus, and loss tangent has been studied. The non-linearity in loss tangent, storage modulus and loss modulus was explained on the basis of MWCNT-elastomer interaction and the inter-aggregate attraction of MWCNT. The secondary β relaxation was studied using dielectric relaxation spectra in the frequency range of 0.1 Hz to 106 Hz. The effect of MWCNT loadings on the complex and real parts of impedance was distinctly visible which has been explained on the basis of interfacial polarization of fillers in a heterogeneous medium and relaxation dynamics of polymer chains in the vicinity of fillers. The dielectric formalism has been utilized to further investigate the conductivity and relaxation phenomenon. The ‘percolation limit’ of the MWCNT in the silicon elastomer was found to be in the range of 4 phr loading.  相似文献

Organic-rich sediments are known to be effective accumulators for uranium and arsenic. Much is known about the capacity for metal or metalloid fixation by microbes and organic compounds as well as inorganic sediment particles. Experiments investigating the effect of microbes on the process of metal fixation in sediments require sterilized sediments as control treatment which is often realized by gamma-sterilization. Only few studies show that gamma-sterilization has an effect on the remobilization of metal and metalloids and on their physico-chemical properties. These studies deal with sediments with negligible organic content whereas almost nothing is known about organic-rich sediments including a probably high microbial activity. In view of this, we investigated the effect of gamma-sterilization of organic-rich sediments on uranium and arsenic fixation and release. After ten days within an exposure experiment we found a significant higher remobilization of uranium and arsenic in sterile compared to unsterile treatments. In line with these findings the content of dissolved organic carbon (DOC), manganese, and iron increased to even significantly higher concentration in the sterile compared to unsterile treatment. Gamma-sterilization seems to change the physico-chemical properties of organic-rich sediments. Microbial activity is effectively eliminated. From increased DOC concentrations in overlaying water it is concluded that microbes are eventually killed with leaching of cellular compounds in the overlaying water. This decreases the adsorption capacity of the sediment and leads to enhanced uranium and arsenic remobilization.  相似文献

In the Arctic, the traditional diet exposes its people to a very high intake of cadmium because it is highly concentrated in the liver and kidneys of commonly eaten marine mammals. In one study in Greenland, the cadmium intake was estimated to 182 microg/day/person in the fall and 346 in the spring. To determine whether the cadmium is accumulated in humans, we analyzed autopsy samples of liver and kidneys from 95 ethnic Greenlanders (aged 19-89) who died from a wide range of causes. The cadmium concentration in liver (overall mean 1.97 microg/g wet wt) appeared to be unrelated to any particular age group, whereas the concentrations in the kidneys peaked in Greenlanders between 40 and 50 years of age (peak concentration 22.3 microg/g wet wt). Despite the high cadmium levels in the typical Greenlander diet, we found that the cadmium concentrations in livers and kidneys were comparable to those reported from Denmark, Sweden, Australia and Great Britain. Furthermore, even though the mean cadmium intake from the diet was estimated to be 13-25 times higher in Greenlanders than in Danes, we found similar cadmium levels in the kidneys of both. Seal livers and kidneys are the main source of cadmium in the diet of Greenlanders, but these tissues are not eaten in Denmark. Thus, our results suggest that the accumulation of cadmium from Greenlander's marine diet is very low.  相似文献

OBJECTIVES: The purpose of this study is to monitor and assess human exposure to pollution in the Arctic which presents a potential future health risk for the local populations. Epidemiological studies in Greenland have shown that human blood levels of several organic contaminants are very high, especially in the North where people depend on local food. In East Greenland (Ittoqqortoormiit (Scoresbysund)) the population shows the highest blood levels of several persistent organic pollutants found in Arctic countries, especially PCB, the levels of which exceed Canadian guideline levels. As in other Arctic countries, the predominant source of these contaminants is the local diet. However, other factors such as smoking may influence the metabolism and thereby the accumulation of toxic substances. STUDY DESIGN: This project is part of the human health program of the ongoing circumpolar "Artic Monitoring and Assessment Programme". Lifestyle, anthropometric factors, and dietary survey results covering 500 men and women from 6 Greenlandic districts were analyzed along with other factors blood lipids, fatty acid profiles, heavy metals and concentrations of persistent organic pollutants (POPs). RESULTS: The dietary survey showed that the traditional food on the average provided 20-30% of the total energy intake. However, the relative monthly meal intake of seal, whale, polar bear, fish and game, and the composition of imported food, varied between districts. Seal and polar bear intake, and, in particular all the plasma n-3 fatty acids, were significantly correlated with organic contaminant concentrations, (betaHCH, chlordanes, DDTs, Hexachlorobenzene, Mirex, PCBs, and toxaphenes) p<0.01(). After adjusting for age, district, dietary factors or plasma n-3 fatty acids, smoking was significantly correlated with high levels of all the above mentioned POPs. CONCLUSION: The main predictors of high contaminant levels in Greenland were age, district, male gender, smoking and high plasma n-3/n-6 ratio as a marker of high dietary intake of local marine mammals.  相似文献

Accumulation of arsenic in Lemna gibba L. was investigated in tailing waters of abandoned uranium mine sites, following the hypothesis that arsenic poses contamination risks in post uranium mining in Saxony, Germany. Consequently, macrophytes growing in mine tailing waters accumulate high amounts of arsenic, which might be advantageous for biomonitoring arsenic transfer to higher trophic levels, and for phytoremediation. Water and L. gibba sample collected from pond on tailing dumps of abandoned mine sites at Lengenfeld and Neuensalz-Mechelgrun were analysed for arsenic. Laboratory cultures in nutrient solutions modified with six arsenic and three PO(4)(3-) concentrations were conducted to gain insight into the arsenic-L. gibba interaction. Arsenic accumulation coefficients in L. gibba were 10 times as much as the background concentrations in both tailing waters and nutrient solutions. Arsenic accumulations in L. gibba increased with arsenic concentration in the milieu but they decreased with phosphorus concentration. Significant reductions in arsenic accumulation in L. gibba were observed with the addition of PO(4)(3-) at all six arsenic test concentrations in laboratory experiments. Plant samples from laboratory trials had on average twofold higher bioaccumulation coefficients than tailing water at similar arsenic concentrations. This would be attributed to strong interaction among chemical components, and competition among ions in natural aquatic environment. The results of the study indicate that L. gibba can be a preliminary bioindicator for arsenic transfer from substrate to plants and might be used to monitor the transfer of arsenic from lower to higher trophic levels in the abandoned mine sites. There is also the potential of using L. gibba L. for arsenic phytoremediation of mine tailing waters because of its high accumulation capacity as demonstrated in this study. Transfer of arsenic contamination transported by accumulations in L. gibba carried with flowing waters, remobilisation through decay, possible methylisation and volatilisation by L. gibba need to be considered.  相似文献
