In toto staining and preservation of peripheral nervous tissue   总被引:2,自引:0,他引:2  
A simple quantitative modification of the in toto staining technique for nervous networks of Sihler is described. The results are demonstrated on the innervation pattern of the hard palate of the rat. Formalin fixed hard palates of rat were macerated and bleached in an aqueous solution of 3% potassium hydroxide with a few drops of 3% hydrogen peroxide added. Thereafter, the specimens were decalcified in Sihler's solution I (1 part glacial acetic acid, 1 part pure glycerin and 6 parts 1% chloral hydrate), the process being controlled by radiography. The specimens were next stained in Sihler's solution II (1 part Ehrlich's hematoxylin, 1 part pure glycerin and 6 parts 1% chloral hydrate). After staining, the non-nervous tissues were destained in Sihler's solution I. Destaining was checked microscopically and was stopped before the finest branches of the nerves began to fade. The specimens were then washed in a weak aqueous solution of lithium carbonate and cleared in increasing concentrations of glycerin. Good visualization of nervous structures and a deep field of observation resulted; orientation of the peripheral nerves with respect to surrounding structures was readily seen and a three-dimensional image of the nervous networks was obtained.  相似文献

In onion ( Allium cepa L.) postponement of sprouting is necessary to achieve long term storage. We studied the factors determining sprouting during dry storage at 16°C. The period to visible sprouting depends on the length of the dormancy period, if present, and on the growth rate of the sprout. In the three cultivars tested, sprouts were initiated within 2 weeks after harvest indicating the absence of a real dormancy period. Sprout length increased linearly during storage. The mitotic activity of the apex decreased before harvest, was low at the transition from scale to leaf formation, and increased again when the sprout was initiated. From a few weeks before harvest, the initially high fructan content of the scales decreased, leading to a large increase in fructose. The sprout always contained enough carbohydrates for growth (between 50 and 60 mg g−1 dry weight, of which 30% was fructan). The activity of sucrose synthase (EC 2.4.1.13) increased as the sprout grew, indicating an increase in sink strength. Invertase (EC 3.2.1.26) was absent in all bulb organs, during the various developmental stages. Although carbohydrates and enzymes were available for fast sprouting, sprout growth was still linear instead of exponential during dry storage at temperatures favorable for growth (16°C). Type II DNA methyltransferases (MTases) are enzymes found ubiquitously in the prokaryotic world, where they play important roles in several cellular processes, such as host protection and epigenetic regulation. Three classes of type II MTases have been identified thus far in bacteria which function in transferring a methyl group from S-adenosyl-l-methionine (SAM) to a target nucleotide base, forming N-6-methyladenine (class I), N-4-methylcytosine (class II), or C-5-methylcytosine (class III). Often, these MTases are associated with a cognate restriction endonuclease (REase) to form a restriction-modification (R-M) system protecting bacterial cells from invasion by foreign DNA. When MTases exist alone, which are then termed orphan MTases, they are believed to be mainly involved in regulatory activities in the bacterial cell. Genomes of various lytic and lysogenic phages have been shown to encode multi- and mono-specific orphan MTases that have the ability to confer protection from restriction endonucleases of their bacterial host(s). The ability of a phage to overcome R-M and other phage-targeting resistance systems can be detrimental to particular biotechnological processes such as dairy fermentations. Conversely, as phages may also be beneficial in certain areas such as phage therapy, phages with additional resistance to host defenses may prolong the effectiveness of the therapy. This minireview will focus on bacteriophage-encoded MTases, their prevalence and diversity, as well as their potential origin and function.  相似文献

Assessing the distribution of 16S rRNA gene sequences within a biological sample represents the current state-of-the-art for determination of human gut microbiota composition. Advances in dissecting the microbial biodiversity of this ecosystem have very much been dependent on the development of novel high-throughput DNA sequencing technologies, like the Ion Torrent. However, the precise representation of this bacterial community may be affected by the protocols used for DNA extraction as well as by the PCR primers employed in the amplification reaction.
