The potential contribution of maternal age to tetrahydrocannabinol's (THC) in utero effects in rats was studied. Pregnant animals were intubated with 25, 10 or 0 mg/kg of THC from gestation day six to parturition. Animals in the 10 and 0 mg/kg groups were pair fed to those given the 25 mg/kg dose. Each series of doses was administered to females three, four or six months of age. THC lowered maternal weight gain and weights of offspring at birth and at 21 days of age, but did not affect litter size, spontaneous alternation or passive avoidance learning in offspring. Increased maternal age was associated with smaller litter size and lower birth weight and weight at 21 days, but did not interact significantly with THC.  相似文献

When the F1-ATPase from the thermophilic bacterium, PS3, was inactivated by 90% with 7-chloro-4-nitro[14C]benzofurazan ([14C]Nbf-Cl) at pH 7.3 and then gel-filtered, 1.25 mols of [14C]Nbf-O-Tyr and less than 0.1 mol of Nbf-N-Lys were formed per mol of enzyme. After adjusting the pH of the gel-filtered, modified enzyme to 9.0 and incubating it for 14 hrs. at 23 degrees C to promote O----N migration, 0.68 mol of Nbf-N-Lys were formed per mol of enzyme while about 16% of the original activity reappeared. Isolation of the subunits after the O----N migration showed that 90% of the incorporated 14C was present in the beta subunit, which contained 0.21 mols of [14C]Nbf-N-Lys per mol. A tryptic peptide which contained the majority of the 14C incorporated into the beta subunit was isolated and subjected to automatic amino acid sequence analysis contained 38 residues. The amino acid sequence immediately around the lysine residue labeled with [14C]Nbf-, K*, was found to be: ...I-G-L-F-G-G-A-G-V-G-K*-T-V-L-I-G... .  相似文献

Optimal muscular coordination strategies for jumping   总被引:5,自引:0,他引:5  
This paper presents a detailed analysis of an optimal control solution to a maximum height squat jump, based upon how muscles accelerate and contribute power to the body segments during the ground contact phase of jumping. Quantitative comparisons of model and experimental results expose a proximal-to-distal sequence of muscle activation (i.e. from hip to knee to ankle). We found that the contribution of muscles dominates both the angular acceleration and the instantaneous power of the segments. However, the contributions of gravity and segmental motion are insignificant, except the latter become important during the final 10% of the jump. Vasti and gluteus maximus muscles are the major energy producers of the lower extremity. These muscles are the prime movers of the lower extremity because they dominate the angular acceleration of the hip toward extension and the instantaneous power of the trunk. In contrast, the ankle plantarflexors (soleus, gastrocnemius, and the other plantarflexors) dominate the total energy of the thigh, though these muscles also contribute appreciably to trunk power during the final 20% of the jump. Therefore, the contribution of these muscles to overall jumping performance cannot be neglected. We found that the biarticular gastrocnemius increases jump height (i.e. the net vertical displacement of the center of mass of the body from standing) by as much as 25%. However, this increase is not due to any unique biarticular action (e.g. proximal-to-distal power transfer from the knee to the ankle), since jumping performance is similar when gastrocnemius is replaced with a uniarticular ankle plantarflexor.  相似文献

Using a battery of seven lectin-ferritin conjugates as probes for cell surface glycoconjugates, we have studied the pattern of plasmalemmal differentiation of cells in the embryonic rat pancreas from day 15 in utero to the early postpartum stage. Our results indicate that differentiation of plasmalemmal glycoconjugates on acinar, endocrine, and centroacinar cells is temporally correlated with development and is unique for each cell type, as indicated by lectin-ferritin binding. Specifically, (a) expression of adult cell surface saccharide phenotype can be detected on presumptive acinar cells as early as 15 d in utero, as indicated by soybean agglutinin binding, and precedes development of intracellular organelles characteristic of mature acinar cells; (b) maturation of the plasmalemma of acinar cells is reached after intracellular cytodifferentiation is completed, as indicated by appearance of Con A and fucoselectin binding sites only at day 19 of development; conversely, maturation of the endocrine cell plasmalemma is accompanied by "loss" (masking) of ricinus communis II agglutinin receptors; and (c) binding sites for fucose lectins and for soybean agglutinin are absent on endocrine and centroacinar cells at all stages examined. We conclude that acinar, centroacinar, and endocrine cells develop from a common progenitor cell(s) whose plasmalemmal carbohydrate composition resembles most closely that of the adult centroacinar cell. Finally, appearance of acinar lumina beginning at approximately 17 d in utero is accompanied by differenetiation of apical and basolateral plasmalemmal domains of epithelial cells, as indicated by enhanced binding of several lectin-ferritin conjugates to the apical plasmalemmal, a pattern that persists from this stage through adult life.  相似文献

Almost 100% of 7- to 10-day-old Hymenolepis diminuta became established when surgically transplanted from donor mice into the duodenum of naive recipient mice. Transplanted tapeworms survived 8-12 days, by which time they had survived much longer in total than they would have done in the donor. Mice previously immunized by a primary infection rejected transplants within 4 days. Sub-lethal irradiation (550 rad) delayed rejection by immune mice but such mice still rejected worms much more quickly than did naive mice. Surgery was shown to delay by 2-3 days the rejection of worms by naive mice, and the importance of circumventing surgery by administering the worms per os is emphasized. Prospects for reconstituting irradiated immune mice are considered vis-à-vis work with nematodes, and the differences which, on present knowledge, appear to exist between nematode and cestode rejection are briefly discussed.  相似文献

The worldwide spread of a novel influenza A (H1N1) virus in 2009 showed that influenza remains a significant health threat, even for individuals in the prime of life. This paper focuses on the unusually high young adult mortality observed during the Spanish flu pandemic of 1918. Using historical records from Canada and the U.S., we report a peak of mortality at the exact age of 28 during the pandemic and argue that this increased mortality resulted from an early life exposure to influenza during the previous Russian flu pandemic of 1889–90. We posit that in specific instances, development of immunological memory to an influenza virus strain in early life may lead to a dysregulated immune response to antigenically novel strains encountered in later life, thereby increasing the risk of death. Exposure during critical periods of development could also create holes in the T cell repertoire and impair fetal maturation in general, thereby increasing mortality from infectious diseases later in life. Knowledge of the age-pattern of susceptibility to mortality from influenza could improve crisis management during future influenza pandemics.

Low-intensity (<100 mW/cm(2)) pulsed ultrasound (US) is an established therapy for fracture repair. In both animal and human trials, such US has been shown to facilitate fresh fracture repair and initiate healing in fractures with repair defects. However, the mechanism by which US achieves these outcomes is not clear. One possible mechanism is the direct stimulation of bone formation. To investigate this hypothesis, the current study investigated the mRNA response of isolated bone-forming cells (UMR-106 cells) to a single 20-min dose of low-intensity pulsed US. Using a novel US-cell coupling method, US was found to stimulate expression of the immediate-early response genes c-fos and COX-2 and elevate mRNA levels for the bone matrix proteins ALP and OC. These findings suggest that low-intensity pulsed US has a direct effect on bone formation. This may contribute to the beneficial effect of low-intensity pulsed US on fracture repair.  相似文献

Testing hypotheses related to the effect of gravitational orientation on neural control mechanisms is difficult for most locomotor tasks, like walking, because body orientation with respect to gravity affects both sensorimotor control and task mechanics. To examine the mechanical effect of body orientation independently from changes in workload and posture, Brown et al. (J. Biomech. 29 p. 1349, 1996) studied pedaling at altered body orientations. They found that subjects pedaling at different orientations changed needlessly their muscle excitations, putatively to preserve body-upright pedaling kinematics. We tested the feasibility of this hypothesis using simulations based on a three biomechanical-function pair organization for control of lower limb muscles (limb extension/flexion pair, extension/flexion transition pair, and foot plantarflexion/dorsiflexion pair), where each pair consists of alternating agonistic/antagonistic muscles. Adjustment of only three parameters, one to scale the muscle excitations of each pair, was sufficient to preserve pedaling kinematics to altered body orientation. Because these adjustments produced changes in muscle excitation and net joint moments similar to those observed in pedaling subjects, the hypothesis is supported. Moreover, the effectiveness of a decoupled gain adjustment procedure where each parameter was adjusted by error in only one aspect of the pedaling trajectory during each iteration (i.e., cadence adjusted the Ext/Flex parameter; peak-to-peak variation in crank velocity over the cycle adjusted the transition parameter; average ankle angle over the cycle adjusted the foot parameter) further supports the distinct function of each muscle pair.  相似文献
