The two-oscillator model of human circadian rhythmicity was analyzed when a zeitgeber relative intensity of 1, 0.5, or 0.1 was introduced into the equations. Fourier analysis was compared with dynamic analysis such as attractor reconstruction or Liapunov exponent calculation. After a 50 or 90% reduction in zeitgeber intensity, the dynamics of the system became equivalent and differed significantly from those of a system with maximal zeitgeber intensity. When 10% aleatory noise was added to the data, the analysis was still applicable, and the results obtained were essentially the same as in the absence of noise. Dynamic analysis could thus provide a distinct classification for periodic data, based on the type of analysis.  相似文献

In order to study glycerolipid metabolism during early embryonic development, Bufo arenarum Hensel female toads were administered [1-3-(14)C]glycerol together with hormonal stimulus. Triglycerides comprised 60% of the total label in all developmental stages analyzed. The highest specific activity was observed in phosphatidic acid which underwent a net decrease as a function of development. Phosphatidylcholine was the most actively synthesized phosphoglyceride. A significant decrease in the total lipid labeling, mainly accounted for by triglycerides, was detected at fertilization time concomitant with a net diminution in the content of these neutral lipids. Subcellular studies showed that yolk platelets contain about 99% of the total oocyte triglycerides suggesting that massive breakdown would be confined to this fraction. Present findings evidence that the de novo lipid biosynthesis is operating during the developmental analyzed period. In addition, they would suggest the active role of triglycerides in the energetic events taking place around fertilization.  相似文献

We have previously shown that a monoclonal antibody (MAb) recognizing the human growth hormone (hGH) antigenic domain left exposed after binding to lactogenic receptors enhanced hGH binding probably through allosteric effects on the hormone binding site. Since receptors displaying different specificities would not recognize exactly the same hGH region, we explored whether some of our MAb could affect hGH binding to somatogenic receptors from rabbit liver and to human liver hGH-specific receptors.The effect of MAbAE5, AC8 and F11 on hGH binding was measured by determining the formation of125I-MAb:hGH:receptor complexes using two different experimental approaches. Results from procedure A, which involved the previous binding of the hormone to microsomes before adding125I-MAb, indicated that the hGH domain defined by epitopes AE5, AC8 and F11 is uncovered in the various hormone:receptor complexes.Procedure B was devised to reveal any alteration in the hGH molecule induced by the MAb. In this case preformed125I-MAb:hGH complexes were added to microsomes. Data showed that125I-MAb AE5:hGH complexes bound better to the various receptors than125I-MAb AE5 to hGH:receptor complexes. On the contrary, hGH previously bound to125I-MAb AC8 or125I-MAb F11 was less recognized by the receptors than the free hormone. Furthermore, binding of MAb AE5 or MAb F11 to hGH 20 K (a natural hGH variant lacking residues 32–46) also enhanced its affinity to the various receptors whereas MAb AC8 did not inhibit hGH 20 K binding.Results indicated that MAb recognizing the hGH antigenic area that remains unmasked after binding to different membrane-bound receptors are able to affect hormone binding site. MAb would induce either positive or negative allosteric changes in the hormone region involved in its binding to lactogenic, somatogenic and hGH-specific receptors.  相似文献

Recent ethnobotanical exploration of henequen (Agave fourcroydes) in the Peninsula of Yucatan, Mexico, finds that inflorescence peduncles are used as emergency food and in the preparation of a fermented drink. Bromatological analysis and determination of total carbohydrates were made for the two length classes (ca. 3.30 m and ca. 0.60 m) which are consumed. The analysis of both the cultivated plant and its putative wild ancestor (Agave angustifolia) suggests that utilization of the inflorescence peduncles as food may have been involved in the initial stages of the history of its evolution under artificial selection, because the wild and the cultivated plants have similar palatability. The subsequent agricultural prevalence of annual crop species in the region was possibly responsible for the abandonment of henequen in the local diet. No significant differences are observed between the bromatological and total carbohydrate values of domesticated and wild plants. The preference for small inflorescence peduncles as a vegetable is a consequence of its significantly minor content of raw fiber and its larger content of total carbohydrates. As a fermented drink, longer peduncles are preferred because they provide more substrate material and because fiber can be eliminated by filtering. This agricultural byproduct, almost totally wasted, has potential value as a source of carbohydrates and raw fiber.  相似文献

Blood samples were collected via jugular catheters from ovariectomized rats at 10-minute intervals for one hour before and two hours after microinjection of 0.5 μl of either saline vehicle or morphine sulfate (10 μg) into the dorsal raphe nucleus (DNR) or adjacent peri-aqueductal gray by means of chronically-implanted guide cannulae. LH was measured by radioimmunoassay and mean pre-injec post-injection values were compared for each rat (t test) as well as for each treatment group (paired t test). Neither saline in DRN nor morphine at other sites significantly altered circulating LH. A significant decrease in LH was observed following injection of morphine into DRN. This effect of morphine was prevented by pre treatment of the animals with the narcotic antagonist naltrexone (10 mg/kg i.v.), indicating the involvement of opiate receptors. These results indicate that DRN is one site at which systemically-administered morphine might act, and suggest the possibility of participation of this mechanism in modulation of LH release by endogenous opioids.  相似文献

A previous report (P. Mavromara-Nazos and B. Roizman, Proc. Natl. Acad. Sci. USA 86:4071-4075, 1989) demonstrated that substitution of sequences of the thymidine kinase (tk) gene, a beta gene, extending from -16 to +51 with sequences extending from -12 to +104 of the gamma 2 UL 49.5 gene in viral recombinant R3820 conferred upon the chimeric gene gamma 2 attributes in the context of the viral genome in a productive infection. The UL49.5 gene sequences extending from -179 to +104 contain four DNA binding sites for the major regulatory protein ICP4. Of these sites, two map between nucleotides +20 and +80 within the sequence which confers gamma 2 regulation upon the chimeric gene. To determine the role of these ICP4 binding sites in conferring the gamma 2 gene attributes, sequences comprising the two ICP4 binding sites were mutagenized and used to reconstruct the R3820 recombinant virus. In addition, a new recombinant virus (R8023) was constructed in which tk sequences extending from -240 to +51 were replaced with wild-type or mutated sequences contained between nucleotides -179 to +104 of the UL 49.5 gene. Vero cells infected with the recombinant viruses in the presence or absence of phosphonoacetate, a specific inhibitor of viral DNA synthesis, were then tested for accumulation of tk RNA by using an RNase protection assay. The results indicate that in the recombinant R3820, a mutation which destroyed one of the two UL49.5 ICP4 DNA binding sites significantly reduced the accumulation of tk RNA at both early and late times after infection. The effect of this mutation was less pronounced in cells infected with the R8023 virus, whose chimeric tk gene contains the two upstream UL49.5 ICP4 binding sites. None of the mutations affected the sensitivity of the chimeric genes to phosphonoacetate. The mutated site appears to be involved in the accumulation of RNA.  相似文献

The fatty acid composition and content of phosphatidylinositol, phosphatidylserine and phosphatidic acid have been studied during the early development of toad embryos. Acidic phospholipids have been analyzed in whole oocytes and embryos and in the following subcellular fractions: yolk platelets, mitochondria and microsomes. Also cardiolipin, a mitochondrial phospholipid, has been analyzed. Gastrula stage embryos have shown, mainly in the mitochondrial fraction, an increase in the content of phosphatidic acid, phosphatidylserine and phosphatidylinositol with respect to unfertilized oocytes. Changes in the distribution of acyl groups of phosphatidic acid have been detected when different subcellular fractions are compared. On the other hand, the phosphatidylserine composition remains unmodified. Arachidonate and stearate are the principal components of phosphatidylinositol. Cardiolipin shows the same composition up to gastrulation and linoleate comprises about 50% of the total acyl groups.  相似文献

Three thermophilic cellulolytic fungi, Chaetomium thermophile var. coprophile, Sporotrichum thermophile, and Thermoascus aurantiacus were studied to determine the conditions for a high rate of cellulose degradation. The range of temperature over which good growth occurred was determined first in a temperature gradient incubator; the optimum temperature was then established in shake flask cultures. T. aurantiacus had the highest optimum growth temperature range (46 to 51 C), whereas S. thermophile had the broadest range over which good growth occurred (36 to 43 C). Optimum temperatures for the three organisms, T. aurantiacus, S. Thermophile, and C. thermophile were 48, 40, and 40 C, respectively. It was found that the addition of an organic carbon and nitrogen source to a cellulose mineral solution medium markedly increased the rate of cellulose degradation. The surfactant, Tween 80, which has been reported to be of value in the production and recovery of the enzyme, cellulase, was shown to be detrimental to the degradation of cellulose in culture. In the medium used, S. thermophile gave the highest rate of substrate utilization; 56% of the cellulose was hydrolyzed in 72 h. The average degree of polymerization of cellulose decreased from 745 to 575.  相似文献
