We caught dunnocks at a wintering site near Pisa/Italy prior to their departure for breeding territories and held them indoors north of this site (Andechs/Germany) on a simulated photoperiod of 52.5 °N. After birds had gone through a reproductive cycle and postnuptial moult they developed migratory restlessness in autumn. At this time one group was transferred back to the previous wintering site (Pisa) where birds were held in individual activity cages in an outdoor aviary, allowing them to perceive as much environmental information as possible. A second group was transferred to a control site near Tour du Valat/France of approximately the same latitude and climate, but different longitude and held in an identical aviary. The diurnal activity pattern changed after transfer back to the previous wintering site, but not after transfer to the control site. Specifically, the amount of morning activity was reduced while afternoon activity was increased. This effect was restricted to those individuals that had been developing nocturnal migratory restlessness the previous spring. It was absent in individuals without migratory restlessness in spring, indicating that the different patterns were not due to unspecific effects from the testing sites. These results are consistent with the hypothesis that birds were able to derive information about their locality and to recognize their previous wintering site, resulting in suppression of migratory state by experience. The results are not definitely conclusive, however, because of several difficulties in the interpretation of perch-hopping activity, which are discussed in detail.  相似文献

Peroxidation of membrane lipids has been hypothesized to play a key role in various types of tissue degeneration and pathology. Lipid peroxides are formed when oxygen reacts with an unsaturated fatty acid chain. Virtually all of the unsaturated fatty acids in biological systems are bound by ester linkages in phospholipids or triglycerides. Phospholipid and triglyceride peroxides are primary products of lipid peroxidation and have rarely been measured. Most of the commonly used methods for detection of lipid peroxidation are based on detection of malondialdehyde or other chemical species that are derived from oxidized fatty acids. This review presents an overview of recently developed methods aimed at identifying and measuring oxidized phospholipids and triglycerides which are direct evidence of the occurrence of lipid peroxidation in vivo.  相似文献

Pyrophosphate:D-fructose-6-phosphate 1-phosphotransferase waspurified over 700-fold from germinating cucumber (Cucumis sativuscv. Fletcher) seeds. The purified enzyme has a specific activityof 5.2 µmol.min–1.mg protein–1 in the presenceof 1 µM fru-2,6-P2. The pH optima is similar for boththe forward and reverse reactions (pH 7.5–7.8). Magnesium,manganese and cobalt activate the enzyme, with the highest affinitybeing for magnesium. The enzyme exhibits normal Michaelis-Mentenkinetics in both the presence and absence of fru-2,6-P2. Half-maximumactivation of the enzyme was obtained with 35 nM fru-2,6-P2.Fru-2,6-P2 stimulates activity by increasing Vmax and increasingthe affinity for fru-6-P, fru-1,6-P2 and PPi. Phosphate causesnoncompetitive inhibition with respect to both fru-6-P and PPi.On the basis of the steadystate substrate interaction and Piinhibition data a sequential ternary complex mechanism is proposed. Methods for measuring 3H-SCH 23390 binding and dopamine (DA) stimulated adenylate cyclase (AC) were established in identical tissue preparations and under similar experimental conditions. Pharmacological characterization revealed that both assays involved interaction with the D1 receptor or closely associated sites. In order to investigate whether the binding sites for 3H-SCH 23390 and DA in fact are identical, the antagonistic effects of a variety of pharmacologically active compounds were examined. Surprisingly, the Ki-values obtained from Schild-plot analysis of the antagonism of DA-stimulated AC, were 80-240 times higher than the Ki-values obtained from competition curves of 3H-SCH 23390 binding. Since both assays were performed under identical conditions, the differences in Ki-values indicate the possibility of different binding sites for DA and 3H-SCH 23390 or, that DA and 3H-SCH 23390 label different states of the same receptor.  相似文献

Zusammenfassung An Hand von 229 Brutbeginn-Daten von freilebenden Gänsen, die während der Jahre 1956–1966 in Seewiesen (Obb.) (48°N, 11°11E) brüteten, wurden die mittleren Brutbeginn-Daten von 5 Gänsearten und von Artbastarden bestimmt. Es zeigte sich, daß die untersuchten Arten unter diesen Bedingungen in derselben Reihenfolge brüteten, wie ihre Artgenossen in freier Wildbahn. Die mittleren Brutbeginn-Termine wurden allerdings um so mehr vorverlegt, je später die Art normalerweise brütet (Abb. 1). , die mit artfremden verpaart waren, brüteten zur selben Zeit wie ihre Artgenossen, die mit artgleichen verpaart waren (Abb. 1). GraugansxSchneegans-Bastard-, die mit Schneegantern verpaart waren, begannen meist nach den Graugänsen, aber stets vor den Schneegänsen zu brüten (Abb. 1, 2). Das intermediäre Brüten dieser wird als starkes Argument für die Richtigkeit der Hypothese gewertet, nach welcher die artspezifisch verschiedenen Brutzeiten wenigstens zum Teil genetisch bedingt sind. In der Diskussion wird die Frage kritisch erörtert, wie weit schon allein die Tatsache, daß die verschiedenen Arten über Generationen hinweg in derselben Reihenfolge wie ihre wildlebenden Artgenossen zu brüten beginnen, als Beweis für derartige genetische Unterschiede angesehen werden kann.
Summary In 229 cases onset of breeding was recorded from free-living geese of 5 species and of some hybrids of these species, kept in Seewiesen/Obb. (48° N, 11° 11E) from 1956 to 1966. It was found that the species under these conditions bred in the same seasonal sequence as did wild birds. The mean breeding times, however, were found to be advanced in relation to the onset of breeding in the wild (Fig. 1). This was especially evident in the case of late-breeding species. paired with of another species came into breeding condition at the same time as paired with of the same species (Fig. 1). GraylegxSnowgoose hybrid paired with Snowgoose in most cases started to breed later than Greyleg geese but always earlier than the mean breeding time for Snowgeese (Fig. 1, 2). This intermediate breeding time is taken as a strong argument for the hypothesis that the species specific differences in breeding times are, at least in part, genetic in origin. The question as to the extent to which the differences in breeding times alone, persisting for generations in the same sequence as those of wild birds, can be attributed to genetic differences between the species, is critically discussed.

Summary In the European starling,Sturnus vulgaris, circannual rhythms in gonadal size, molt and other related functions persist only in photoperiods close to 12 h, but are absent in longer or shorter daylengths. To find out whether the arrhythmia seen in long photoperiods results from an arrest of the underlying clock system, three groups of male starlings were held for 10, 14, or 20 months in a 13 h photoperiod and then transferred to a 12 h photoperiod. A control group was held in the 13 h photoperiod throughout the experiment for 28 months. During the initial exposure to the 13 h photoperiod, all birds went through a gonadal cycle, followed by a complete molt. Subsequently, the control birds retained small testes to the end of the experiment and there was no further molt. In contrast, most of the experimental birds re-initiated a testicular cycle, following transfer to the 12 h photoperiod and molted after its completion. The latency between the transfer to the 12 h photoperiod and the onset of testicular growth was not significantly different among the three groups, indicating that the underlying circannual clock had been arrested in the 13 h photoperiod and restarted in the 12 h photoperiod. The pattern of the second testicular cycle did, however, differ among groups. Particularly its amplitude decreased from group 1 to group 3, suggesting that the capacity of the birds to respond to a 12 h photoperiod decreased with increasing duration of exposure to the 13 h photoperiod.Dedicated to Prof. Dr. C.S. Tannic acid induces aggregation and formation of multilamellar vesicles when added to preparations of small unilamellar vesicles, specifically those containing phosphatidylcholine. Aggregation and clustering of vesicles was demonstrated by cryo-electron microscopy of thin films and by freeze-fracture technique. Turbidity measurements revealed an approximately one-to-one molar ratio between tannic acid and phosphatidylcholine necessary for a fast and massive aggregation of the small unilamellar vesicles. When tannic acid-induced aggregates were dehydrated and embedded for conventional thin-section electron microscopy, multilamellar vesicles were retrieved in thin sections. It is concluded from morphological studies, as well as previous tracer studies, that tannic acid, at least to a great extent, prevents the extraction of phosphatidylcholine. Multilamellar vesicles were also observed in tannic acid-treated vesicles prepared from total lipid extracts from either rabbit or rat hearts. Substantially more multilamellar vesicles were retrieved in the rabbit vesicle preparation. This difference can probably be explained by the difference in the proportion of the plasmalogen phosphatidylcholine, and possibly the content of sphingomyelin, in lipid extracts of rabbit and rat hearts. It is concluded that the dual effect (reduced extraction and aggregation) of tannic acid on phosphatidylcholines should be taken into consideration when tannic acid is used in tissue preparation.  相似文献
