1. At a holding potential of -40 mV, carbachol (50 microM) produced a complex pattern of inward currents in single smooth muscle cells freshly isolated from the mouse anococcygeus. Membrane currents were monitored by the whole-cell configuration of the patch-clamp technique. Previous work has identified the first, transient component as a calcium-activated chloride current (ICl(Ca)) and the second sustained component as a store depletion-operated non-selective cation current (I(DOC)). The object of the present study was to examine the cellular mechanisms underlying the third component, a series of inward current oscillations (I(oscil)) superimposed on I(DOC). 2. Carbachol-induced I(oscil) (amplitude 97 +/- 11 pA; frequency 0.26 +/- 0.02 Hz) was inhibited by the chloride channel blocker anthracene-9-carboxylic acid (A-9-C; 1 mM), and by inclusion of 1 mM EGTA in the patch-pipette filling solution. 3. In calcium-free extracellular medium (plus 1 mM EGTA), carbachol produced an initial burst of oscillatory current which lasted 94 s before decaying to zero; I(oscil) could be restored by re-admission of calcium. The frequency, but not the amplitude, of I(oscil) increased with increasing concentrations of extracellular calcium (0.5-10 mM). 4. Inclusion of the inositol triphosphate (IP3) receptor antagonist heparin (5 mg ml(-1) in the patch-pipette filling solution, or pretreatment of cells with the sarcoplasmic reticulum (SR) calcium ATPase inhibitor cyclopiazonic acid (CPA; 10 microM), prevented the activation of I(oscil) by carbachol. Caffeine (10 mM) activated both ICl(Ca) and I(DOC) and prevented the induction of I(oscil) by carbachol. Caffeine and CPA also abolished I(oscil) in the presence of carbachol, as did both a low (3 microM) and a high (30 microM) concentration of ryanodine. 5. Carbachol-induced I(oscil) was abolished by the general calcium entry blocker SKF 96365 (10 MM) and by Cd2+ (100 microM), but was unaffected by La3+ (400 microM). As found previously, I(DOC) was also blocked by SKF 96365 and Cd2+, but not La3+; the inhibition of I(DOC) preceded the abolition of I(oscil) by 27 s with SKF 96365 and by 30 s with Cd2+. Nifedipine (1 microM) produced a partial inhibition of the carbachol-induced I(oscil) frequency at holding potentials of -20 mV and -60 mV and, in addition, reduced I(DOC) at -60 mV by 18%. 6. It is concluded that carbachol-induced inward current oscillations in mouse anococcygeus cells are due to a calcium-activated chloride current, and reflect oscillatory changes in cytoplasmic calcium ion concentration. These calcium oscillations are derived primarily from the SR stores, but entry of calcium into the cell is necessary for store replenishment and maintenance of the oscillations. Capacitative calcium entry (via I(DOC) appears to be important not only for sustained contraction of this tissue, but also as a route for re-filling of the SR and, therefore, represents an important target for the development of novel and selective drugs.  相似文献

BACKGROUND: Surgical training and experience are frequently claimed to influence early and late outcome measures. The aim of this study was to examine any improvement in an individual surgeon's performance in one operation over a period of 7 years from initial appointment to date. METHODS: Patients undergoing Ivor Lewis subtotal oesophagectomy performed by a single surgeon between April 1990 and December 1996 were identified from a prospectively compiled oesophageal cancer database. Operating time (abdominal, thoracic and 'one-lung time'), blood loss, transfusion requirements (intraoperative and total), extent of lymphadenectomy (number of lymph nodes sampled), intensive treatment unit (ITU) stay, hospital stay, postoperative morbidity and mortality, pathological stage, grade and survival were recorded. RESULTS: The records of 150 patients were identified for analysis. The cohort was split into five groups, each of 30 patients operated on consecutively. Each of the groups was comparable for age, sex, smoking history, preoperative haemoglobin and creatinine levels, weight loss, American Society of Anesthesiologists' grade, and histological stage and grade of disease. Analysis of the variables pertaining to operation revealed a significant improvement with time including reduced single-lung operating time (P=0.01), reduced blood loss (P=0.03), reduced transfusion requirement (P < 0.0001), reduced ITU stay (P< 0.0001), reduced inpatient stay (P< 0.0001) and an increased yield of lymph nodes (P < 0.0001). CONCLUSION: This study showed a continuing improvement in a surgeon's performance over a 7-year period. With the current trend to shorter training periods there is a case for continuing supervision of the 'fully trained' surgeon within highly specialist units.  相似文献

In a previous article the authors have described changes in molecular weight which occur when aspen milled wood lignin is subjecte to autocatalyzed hydrolysis (autohydrolysis) in the presence of three aromatic additives: 2-naphthol, resorcinol, and p-hydroxybenzoic acid. In the present work, the reactions of these additives have been studied from the viewpoint of their effect on the structure of the lignin. The use of milled wood lignin instead of aspen wood served to distinguish those reactions between the additives and the lignin, without involving the other wood components. The reaction products have been charactrized by elemental analysis and UV and IR spectroscopy. It was concluded that keto groups, generated on the lignin side-chain during autohydrolysis, condense with the additives, which become incorporated into the lignin. This occurs most probably through an aromatic eletrophilic substitution reaction. While 2-naphthol condensed primarily with the β-keto carbonyl groups, resorcinol attacked also the α-keto carbonyl groups. When p-hydroxybenzoic acid was the additive, its incorporation was accompanied by an increase in lignin carboxylic acid groups.  相似文献

This paper presents methods of modeling and predicting face recognition (FR) system performance based on analysis of similarity scores. We define the performance of an FR system as its recognition accuracy, and consider the intrinsic and extrinsic factors affecting its performance. The intrinsic factors of an FR system include the gallery images, the FR algorithm, and the tuning parameters. The extrinsic factors include mainly query image conditions. For performance modeling, we propose the concept of "perfect recognition", based on which a performance metric is extracted from perfect recognition similarity scores (PRSS) to relate the performance of an FR system to its intrinsic factors. The PRSS performance metric allows tuning FR algorithm parameters offline for near optimal performance. In addition, the performance metric extracted from query images is used to adjust face alignment parameters online for improved performance. For online prediction of the performance of an FR system on query images, features are extracted from the actual recognition similarity scores and their corresponding PRSS. Using such features, we can predict online if an individual query image can be correctly matched by the FR system, based on which we can reduce the incorrect match rates. Engine applications of the shape memory ("Marmem" ) effect associated with thermoelastic transformations are discussed. Such devices potentially enable the direct conversion of heat(i.e., solar energy or waste heat) into mechanical work. It is shown that efficiencies on the order of 20 pct may be expected and can be improved with alloys having certain transformation hysteresis loop characteristics. As part of a study on newly developed Fe-based shape memory alloys associated with face-centered cubic-hexagonal close-packed (fcc-hcp) martensitic transformations, the shape memory behavior is reported and various physical factors influencing it discussed, based on experimental results from dilatometry and bending tests. It is shown that the alloys studied exhibit a good shape memory effect (SME) without special training. The SME is significantly improved by prestraining at low temperatures. A higher net reversible strain is made available by over-prestraining, although the fully reversible prestrain usually does not exceed 2 pct. Super-elasticity, the two-way shape memory, and a softening effect of the matrix at low temperatures are briefly discussed.  相似文献

A transmission electron microscopy and diffraction study of martensite plates in an Fe-3 pct Mn-3 pct Cr-1 pct C alloy was carried out with particular attention to details of the martensite substructure. A corresponding optical metallographic study of plate morphology was made. A variability in martensite substructure was observed from plate to plate, although a (252)F * plate was generally associated with (112)B transformation twins and {111}F stacking faults. The particular (111)F fault variant gave rise to a wedge-shaped plate morphology. Planar {101}B inhomogeneities were frequently observed in the martensite, and most of these appear to be derived from austenite stacking faults. In general, more than one type of inhomogeneity was observed in a single martensite plate and the “typical” plate substructure was rather difficult to characterize, although the habit plane was found invariably to be (252)F.  相似文献

This investigation reports on an optical and electron microscope study of bct martensite formed in Fe-7 pct Al-1.5 pct C and Fe-7 pct Al-2.0 pct C alloys. In each case the martensite is plate-like containing \((112)[\bar 1\bar 11]\) transformation twins 100 to 200Å in width. The particular twin plane variant \((112)[\bar 1\bar 11]\) corresponds to the martensite habit plane variant (3, 15, 10)F, which is predicted by the crystallography theory. The twins are uniformly spaced and extend completely from one martensite-austenite interface to the other as would be theoretically expected. The martensite plates are ideally lenticular in the 2 pct C alloy but those in the 1.5 pct C alloy frequently exhibit irregular interfaces which are attributed to impingement effects. All observations are in accordance with the phenomenological crystallography theory as applied to ferrous martensites with a {3, 15, 10}F habit plane.  相似文献

The applicability of the phenomenological crystallographic theory to martensitic transformations in ferrous alloys is discussed, and it is concluded that only for the {3, 15, 10} F and {252} F transformations are experimental data sufficiently complete for detailed comparisons with predictions of the theory. Since crystallographic measurements have proved more difficult to obtain in the {252}F case a detailed assessment of electron microscopy observations and of reported measurements of the shape strain and orientation relationship is given for this transformation. The basic theory introduced, in the 1950's accounts satisfactorily for the {3, 15, 10} F transformation but not for the {252} F , case. Recent modifications of the theory based on multiple lattice invariant shears are also reviewed and are examined in the light of the crystallographic measurements. The use of experimental information to infer possible inhomogeneous deformation modes is emphasized.  相似文献
