目的 建立蒙药绿松石的质量标准。方法 收集不同产地绿松石,共10批。观察绿松石样品和粉末的性状并进行理化鉴别;按2020年版《中国药典》(四部)通则方法测定绿松石样品中水分、浸出物含量;采用原子吸收光谱法测定绿松石样品铜元素含量。结果 绿松石为不规则、周围带有黑石的块状物,表面蓝绿色,体重,质硬脆,难砸碎,断面呈贝壳状,蜡样光泽,粉末呈灰绿色,无臭,味淡;理化鉴别结果显示,呈铜盐反应;10批次样品中水分含量为0.41%-3.94%(SD=1.37%),浸出物含量为0.21%-0.81%(SD=0.21%),铜元素含量为3.03%-4.63%(SD=0.63%)。结论 初步拟定绿松石中水分含量不得超多5.0%、浸出物含量不得低于0.10%,铜元素含量应为2.60%-4.84%,制定的标准可用于蒙药材绿松石的质量控制。 相似文献

Tamoxifen prevents recurrence of breast cancer and is suggested for preventive risk-reducing therapy. Tamoxifen reduces mammographic density, a proxy for therapy response, but little is known about its effects in remodelling normal breast tissue. Our study, a substudy within the double-blinded dose-determination trial KARISMA, investigated tamoxifen-specific changes in breast tissue composition and histological markers in healthy women. We included 83 healthy women randomised to 6 months daily intake of 20, 10, 5, 2.5, 1 mg of tamoxifen or placebo. The groups were combined to “no dose” (0-1 mg), “low-dose” (2.5-5 mg) or “high-dose” (10-20 mg) of tamoxifen. Ultrasound-guided biopsies were collected before and after tamoxifen exposure. In each biopsy, epithelial, stromal and adipose tissues was quantified, and expression of epithelial and stromal Ki67, oestrogen receptor (ER) and progesterone receptor (PR) analysed. Mammographic density using STRATUS was measured at baseline and end-of-tamoxifen-exposure. We found that different doses of tamoxifen reduced mammographic density and glandular-epithelial area in premenopausal women and associated with reduced epithelium and increased adipose tissue. High-dose tamoxifen also decreased epithelial ER and PR expressions in premenopausal women. Premenopausal women with the greatest reduction in proliferation also had the greatest epithelial reduction. In postmenopausal women, high-dose tamoxifen decreased the epithelial area with no measurable density decrease. Tamoxifen at both low and high doses influences breast tissue composition and expression of histological markers in the normal breast. Our findings connect epithelial proliferation with tissue remodelling in premenopausal women and provide novel insights to understanding biological mechanisms of primary prevention with tamoxifen. 相似文献

目的 运用Q-Exactive高分辨质谱结合指纹图谱评价不同批次广枣药材质量,为朱日亨滴丸源头把控提供参考。方法 采用Agilent ZORBAX ECLIPSE XDB-C18色谱柱,以0.1%甲酸水-甲醇梯度洗脱30min,柱温为30℃,流速为1.0ml.min-1。采用电喷雾(ESI)质谱离子源,在正离子与负离子模式下采集数据,检测范围为m/z 100-1500。依据精确质量数以及二级碎片离子,结合标准物质及相关数据库信息,对蒙药广枣药材主要化学成分进行定性研究。同时测定10批广枣药材,应用“中药色谱指纹图谱相似度评价系统(2004A)”建立广枣指纹图谱,并对各共有峰进行化学成分归属,对鞣花酸进行定量,评价各批次药材的质量。结果 鉴定出广枣中的37种化合物,并以鞣花酸为参照物,建立广枣药材指纹图谱,10批次广枣药材相似均在0.90以上。不同批次广枣药材鞣花酸含量差异明显。结论 建立基于Q-Exactive高分辨质谱结合指纹图谱技术的蒙药广枣药材质量评价研究方法为广枣药材质量控制提供参考,也为主日亨滴丸源头保证提供科学依据。 相似文献

Mutations in SH2D1A, encoding the intracellular adaptor signaling lymphocyte activation molecule associated protein (SAP), are associated with X‐linked lymphoproliferative disease type 1 (XLP1). We identified a novel hemizygous SH2D1A c.49G > A (p.E17K) variant in a 21‐year‐old patient with fatal Epstein‐Barr virus infection–associated hemophagocytic lymphohistiocytosis. Cellular and biochemical assays revealed normal expression of the SAP variant protein, yet binding to phosphorylated CD244 receptor was reduced by >95%. Three healthy brothers carried the SH2D1A c.49G > A variant. Thus, data suggest that this variant represents a pathogenic mutation, but with variable expressivity. Importantly, our results highlight challenges in the clinical interpretation of SH2D1A variants and caution in using functional flow cytometry assays for the diagnosis of XLP1. 相似文献

Decorin is known to influence tissue tensile strength and cellular phenotype. Therefore, decorin is likely to have an impact on tissue repair, including cutaneous wound healing. In this study, cutaneous healing of both excisional and incisional full‐thickness dermal wounds was studied in decorin‐deficient (Dcn?/?) animals. A statistically significant delay in excisional wound healing in the Dcn?/? mice occurred at 4 and 10 days postwounding and, in incisional wounds at 4, 10, and 18 days when compared with wild‐type (Dcn?/?) controls. Fibrovascular invasion into polyvinylalcohol sponges was significantly increased by day 18 in Dcn?/? mice relative to Dcn+/+ mice. The 18‐day sponge implants in the Dcn?/? mice showed a marked accumulation of biglycan when compared with the corresponding implants in Dcn+/+ mice. Thus, regulated production of decorin may serve as an excellent therapeutic approach for modifying impaired wound healing and harmful foreign body reactions. 相似文献
