Human liver fatty acid binding protein (hL‐FABP) has been reported to act as an intracellular shuttle of lipid molecules, thus playing a central role in systemic metabolic homeostasis. The involvement of hL‐FABP in the transport of bile salts has been postulated but scarcely investigated. Here we describe a thorough NMR investigation of glycocholate (GCA) binding to hL‐FABP. The protein molecule bound a single molecule of GCA, in contrast to the 1:2 stoichiometry observed with fatty acids. GCA was found to occupy the large internal cavity of hL‐FABP, without requiring major conformational rearrangement of the protein backbone; rather, this led to increased stability, similar to that estimated for the hL‐FABP:oleate complex. Fast‐timescale dynamics appeared not to be significantly perturbed in the presence of ligands. Slow motions (unlike for other proteins of the family) were retained or enhanced upon binding, consistent with a requirement for structural plasticity for promiscuous recognition.  相似文献

This paper analyzes the thermal aging embrittlement occurred in a cast stainless steel valve, which is part of the reactor water clean-up (RWCU) system of a Spanish boiling water reactor (BWR) nuclear power plant. The aim is to estimate the current and future state of the material and the corresponding structural integrity of the valve. Given that there is no data available for the experimental characterization of the material, the evolution of the mechanical properties (fracture toughness, yield stress, flow stress and Ramberg-Osgood parameters) has been estimated using the ANL procedure.With the obtained estimations, the critical crack size has been calculated using the European procedure FITNET FFS and the ASME Code.This analysis considers not only the evolution of the mechanical properties up to now but also its future evolution in case there is a life extension of the plant until year 2029.  相似文献

Reservoirs are the main water source in the Brazilian semiarid region, especially in the crystalline-geology watersheds, forming high-density reservoir networks (HdRN). However, in most cases, the construction of these reservoirs has been done without technical supervision. The objective of this work was to map and evaluate the spatial distribution of the 25,000 km2 Orós Reservoir Basin (ORB) HdRN, in semiarid Brazil, with the help of remote-sensing tools associated with geographic information systems (GIS). Using LANDSAT 5 images of the end of the 2011 rainy season of the ORB, the remote-sensing technique allowed the identification of 6002 polygons, which corresponded to only 4717 reservoirs (implying a misidentification of 21%). Between 2002 and 2011, a 17.5% increase (and 1.81% annual increase) in the number of reservoirs in the basin was observed, still lower than the annual increment from 1970 to 2002, when an average increase of 2.64% per year was observed, in other studies. The perimeter of the reservoirs ranged from 0.250 to 560 km and the individual water surface area ranged from 0.004 to 195.0 km2, resulting in a total surface of 465.0 km2. Analysing the surface area of the strategic reservoirs, results showed that the estimation of the surface area (from remote sensing with manual polygon adjustment) yielded values very close to those of the on-site monitored areas, with R2 = 0.99 and normalized difference index ranging from ?0.02 to +0.09. The reservoir density in the ORB in 2011 was 0.19 reservoirs km2, higher than the recommended optimum density of 0.15 reservoirs km2 basin. Analysis of reservoir density by municipality recorded values ranging from 0.02 to 0.40 reservoirs km2. The sedimentary-geology municipalities presented a reservoir density on average 80% lower than the that of the crystalline-geology municipalities, indicating a strong relationship between geology and reservoir density. Neither population density nor rainfall explained the spatial distribution of reservoirs within the basin, both yielding R2 lower than 0.1. This remote-sensing survey of reservoirs demonstrated two major flaws: the misidentification of shadows as reservoirs and the inability to identify the presence of macrophytes, which negatively affected the number and surface area of the target reservoirs. Despite these problems, remote sensing has been shown to be a technique of great potential in the planning and management of water resources in regions with dense reservoir networks.  相似文献

This paper reviews over 120 papers regarding the effect of heat treatment on the catalytic activity and stability of proton exchange membrane (PEM) fuel cell catalysts. These catalysts include primarily unsupported and carbon-supported platinum (Pt), Pt alloys, non-Pt alloys, and transition metal macrocycles. The heat treatment can induce changes in catalyst properties such as particle size, morphology, dispersion of the metal on the support, alloying degree, active site formation, catalytic activity, and catalytic stability. The optimum heat-treatment temperature and time period are strongly dependent on the individual catalyst. With respect to Pt-based catalysts, heat treatment can induce particle-size growth, better alloying degree, and changes in the catalyst surface morphology from amorphous to more ordered states, all of which have a remarkable effect on oxygen reduction reaction (ORR) activity and stability. However, heat treatment of the catalyst carbon supports can also significantly affect the ORR catalytic activity of the supported catalyst. Regarding non-noble catalysts, in particular transition metal macrocycles, heat treatment is also important in ORR activity and stability improvement. In fact, heat treatment is a necessary step for introducing more active catalytic sites. For metal chalcogenide catalysts, it seems that heat treatment may not be necessary for catalytic activity and stability improvement. More research is necessary to improve our fundamental understanding and to develop a new strategy that includes innovative heat-treatment processes for enhancing fuel cell catalyst activity and stability.  相似文献

The effects of a single injection of morphine (20 mg/kg) on serum testosterone levels were examined in the male rat. Within 2 hours after the morphine injection, testosterone levels were significantly lower than control levels. The decline in testosterone levels reached a maximum 4 hours after the administration of morphine, at which time testosterone levels were reduced by more than 85% with respect to controls. The ability of a large number of narcotics to depress serum testosterone levels, 4 hours after their administration, was also examined. All narcotics depressed testosterone levels significantly and their potency relative to morphine was comparable to that observed in several other preparations, such as the guinea-pig ileum and mouse vas deferens. The testosterone-depleting effects of the narcotics appear to represent specific narcotic effects since the (-)-isomers of the narcotics were considerably more potent than the (+)-isomers, naloxone competitively inhibited the effects of morphine on testosterone levels and tolerance developed to the testosterone-depleting effects of these drugs. Acute treatment with morphine also lowered serum luteinizing hormone levels, and this reduction preceded the fall in testosterone levels by 1 to 2 hours.  相似文献

Although it is recognized that drugs ingested by pregnant females produce marked cognitive and physiological deficits in their offspring, the possibility that paternal exposure to drugs prior to mating may have adverse effects on fertility and fetal outcome has not received much attention. The purpose of the present studies was to examine whether a single, acute exposure to alcohol influences the subsequent ability of adult male rats to mate and produce healthy and viable litters. Our results showed that a relatively large dose of alcohol 24 hours prior to breeding had little effect on the mating behavior of male rats, but there were markedly fewer pregnancies in females mated with alcohol-exposed male rats than in controls. Of equal importance, we found that, even when conception occurred and live births were produced, there were striking differences in fetal outcome. Alcohol-treated males sired many fewer pups than control males and there was a markedly enhanced mortality rate in their offspring. Collectively, these data suggest that acute paternal alcohol administration 24 hours prior to breeding does not affect mating behavior, but results in a greatly diminished fertility rate and fewer and less viable offspring. These studies suggest that paternal alcohol use may be as important as maternal alcohol abuse as a negative variable in pregnancy and fetal outcome.  相似文献

BACKGROUND: Clinical criteria to select patients with headache in whom structural diagnostic studies (computed tomography) have a high yield disclosing intracranial pathologic findings, independent of abnormal findings on neurologic examination, have not been defined. OBJECTIVE: To determine which clinical characteristics predict the presence of intracranial pathologic findings, independently of neurologic examination, in patients with headache. DESIGN: Case-control, consecutive sample. SETTING: Major metropolitan trauma center emergency department. PATIENTS AND MATERIALS: Hospital records of 139 hospitalized and 329 randomly selected patients from 1720 nonhospitalized adult patients, consecutively evaluated for headache in the emergency department, were reviewed. Demographic data, clinical characteristics of the headache, results of neurologic and physical examinations, and diagnostic radiologic and laboratory results were correlated with final diagnosis and outcome at 6 months after emergency department visit. DATA ANALYSIS: Nonparametric statistical analysis. RESULTS: Intracranial pathologic findings were found in 18 (3.8%) of 468 patients. Acute onset and occipitonuchal location of headache, presence of associated symptoms, and patient age of 55 years or older were significantly associated with the finding of intracranial pathology, independently of the findings from neurologic examination. Abnormal findings on neurologic examination alone, whether focal or nonfocal, had a highly significant association and a positive predictive value for intracranial pathology of 39%. CONCLUSIONS: Abnormal results from neurologic examination are the best clinical parameters to predict structural intracranial pathology; however, in patients 55 years or older with headache of acute onset located in the occipitonuchal region that has associated symptoms, computed tomographic scan of the head is justified as part of their clinical evaluation independently of the findings of the neurologic examination.  相似文献

The fracture assessment of notched components based on cracked components approaches leads to over‐conservative failure predictions. In the research literature, several approaches are proposed to overcome this problem using an apparent fracture toughness, . Nevertheless, most of these approaches are based on deterministic assumptions despite the large and variable scatter exhibited by for different notch radii (ρ) or temperatures (T). This paper proposes a methodology for deriving a probabilistic field including the effect of temperature on the failure of notched components. First, the theory of critical distances is applied to transform each apparent fracture toughness into the equivalent fracture toughness for ρ = 0. Then, the temperature is supposed to act as a scale effect in the Weibull cumulative distribution function of the equivalent fracture toughness, and the corresponding scale effect function is derived. Finally, the applicability of the proposed methodology is illustrated by an example using two ferritic‐pearlitic steels: S275JR and S355J2.  相似文献

Food security and nutrition is a multidimensional theme consisting of three pillars: food access, availability, and sufficiency. Given food security and nutrition's extent, its measures are usually limited. To address these limitations, this paper proposes the creation of a disaggregated food insecurity index for Brazilian states, with the three dimensions in mind. The results show that while food availability elevates the average of the index, the sufficiency still is a problem. Even though the distribution of and access to food in Brazil has increased in the last years, it was not combined with policies that guarantee its safety and quality, reinforcing the need for specific interventions considering regional particularities.  相似文献
