All solid tumors must acquire a vascular stroma to grow beyond a minimal size. Vascular endothelial growth factor (VEGF) is a potent and specific angiogenic growth factor both in vitro and in vivo that may participate in the formation of the vascular tumor stroma. In the present study, we examined the expression of VEGF in the paraffin sections of 20 eyes harboring retinoblastoma or posterior uveal melanoma, but also in corresponding tumor cellines. By using in situ hybridization, we found that all but one of the retinoblastomas expressed VEGF mRNA. Particularly high expression was detected in areas of loosely packed tumor cells with prominent chromatin. By contrast, none of the posterior uveal melanomas expressed significant amounts of VEGF mRNA. Immunostaining with an antibody against VEGF confirmed that retinoblastomas, but not posterior uveal melanomas, also contained detectable VEGF protein. To further study the expression of VEGF in these tumor cells we performed Northern blotting on a retinoblastoma celline, Y79, and on an uveal melanoma celline, OM431. Both of these cellines expressed low levels of VEGF mRNA under normal culture conditions. However, when the cells were cultured under hypoxic conditions, a strong increase in VEGF mRNA could be seen in Y79 cells but not in OM431 cells. By using a bioassay, we also found that hypoxia stimulated the secretion of VEGF protein into the culture medium of Y79 cells. In conclusion, we have shown that VEGF mRNA and protein are expressed in retinoblastomas but not in posterior uveal melanomas. Moreover we have shown that VEGF is hypoxia-inducible in retinoblastoma cells. These results suggest that focal hypoxia may act as a stimulus for VEGF production in retinoblastomas, that in turn may contribute to tumor growth by stimulating the formation of a vascular stroma.  相似文献

In this paper, we propose that the urinary toxins from the wastewater be adsorbed on an adsorbent such as spherical activated carbon and the latter be regenerated by subjecting it to high temperatures to recycle activated carbon and also to recycle the water used in dialysis. We studied the adsorption of artificial waste dialysate, which is a mixed solution of urea, creatinine, and uric acid, and the separate solutions for each of these and found that their extents of adsorption onto the spherical activated carbon material were nearly identical. The amount of adsorption was approximately 1.4 mg·g-1 for urea, 18 mg·g-1 for creatinine, and 20 mg·g-1 for uric acid. The urea, creatinine, and uric acid adsorbed onto the spherical activated carbon decomposed on heat treatment at 500℃, and the adsorption capacity of the spherical activated carbon was regenerated. Our study successfully demonstrated that the spherical activated carbon can be recycled in the waste dialysate treatment process.  相似文献

Corrosion behaviour of Si3N4 ceramics containing Y2O3, Al2O3 and AIN as sintering aids was investigated under hydrothermal conditions at 200–300 C and saturated vapour pressures of water for 1–10 days. Hydrothermal corrosion resulted in the dissolution of the Si3N4 matrix and the formation of a product layer consisting of the original grain-boundary phases and hydrated silica. The dissolution rate of Si3N4 ceramics decreased with decreasing crystallinity of the grain-boundary phase. The dissolution rate could be adequately described by a parabolic plot in the initial stage of the reaction. The apparent activation energies were 83.5–108 kJ mol–1, and the bending strength of the corroded samples decreased from 600 to 400 MPa in the initial stage of the reaction upto a weight loss of 0.004 g cm–2, and then was almost constant up to a weight loss of 0.012 g cm–2.  相似文献

DNA machines consisting of consecutive hairpins, which we have previously described, have various potential applications in DNA computation. In the present study, a 288-base DNA machine containing four consecutive hairpins was successfully constructed by ligation and PCR. PAGE and fluorescence spectroscopy experiments verified that all four hairpins were successfully opened by four opener oligomers, and that hairpin opening was dependent on the proper openers added in the correct order. Quantitative analysis of the final results by fluorescence spectroscopy indicated that all four hairpins were open in about 1/4 to 1/3 of the DNA machines.  相似文献

An adaptive workload balancing scheme is analyzed. This scheme is intended to automatically dissolve the imbalance in resource utilization of computing systems and to bring about the proper balance between throughput and turnaround time. This is done by means of the invisible hand due to the bidding of priorities of resource use, with no contradiction between the flexible bid for each resource and the stable price for the entire service. The result of this analysis shows that this scheme would regulate those imbalances, and that it includes the load balancing policy of IBM OS/VS2-2 as a special case. Sodium laureth sulfate and amino acid type surfactants have been categorized as low‐skin irritation chemicals based on results of previous skin irritation tests. However, detergent‐induced skin irritation still occurs. Detergents with a low‐skin irritation effect are required, since the number of cosmetics for sensitive skin has increased. Therefore, an in vitro method of testing the safety of cosmetics for sensitive skin is required. Skin irritation by anionic surfactants was investigated to determine the effect of laureth‐3 carboxylate (polyoxyethylene lauryl carboxymethyl ether) amino acid salt on skin irritation. In addition, sodium laureth sulfate and amino acid type surfactants, regarded as low‐irritation surfactants, were also tested for skin irritation. The skin irritation effect of laureth‐3 carboxylate lysine salt (Surfactant 1 ), sodium laureth sulfate (Surfactant 2 ), and sodium N‐lauroyl glutamate (Surfactant 3 ) were investigated using a reconstructed human cultured epidermal model, LabCyte EPI‐MODEL24 6D. Cell viabilities of cultured epidermal cells exposed to Surfactant 1 (5.0 % aq.), Surfactant 2 , and Surfactant 3 were 82.0, 45.0 and 19.1 %, respectively. There were significant differences in cell viability upon exposure to 5.0 % aqueous test solutions of the three test chemicals. The results of the current investigation indicate that Surfactant 1 has a low skin‐irritation effect.  相似文献

In this paper, we propose a new architecture for a multi-state DNA machine whose conformation of repeated hairpin structures changes sequentially in response to input oligomers. As an application of the machine, we also propose molecular memory in which the machine is used as a memory unit. Addressing in the memory is realized through state transitions of the machine. We then describe a method for designing DNA sequences of the machine, which exhaustively checks conformational changes of the machine by dividing its secondary structure into hairpin units. The method is based on the minimum free energy of the structure, the structure transition paths, and the total frequency of optimal and suboptimal structures. DNA sequences designed by the method were tested in a chemical experiment in which a machine consisting of two hairpins was actually constructed. As a result, we verified that the multi-state DNA machine realized the expected changes in its secondary structure.  相似文献

The interferometric monitor for greenhouse gases (IMG) was the precursor of the high-resolution Fourier-transform infrared radiometer (FTIR) onboard a satellite for observation of the Earth. The IMG endured the stress of a rocket launch, demonstrating that the high-resolution, high-throughput spectrometer is indeed feasible for use onboard a satellite. The IMG adopted a newly developed lubricant-free magnetic suspension mechanism and a dynamic alignment system for the moving mirror with a maximum traveling distance of 10 cm. We present the instrumentation of the IMG, characteristics of the movable mirror drive system, and the evaluation results of sensor specifications during space operation.  相似文献
