Blended SRC-II process streams, representing a full boiling range distillate material, were fractionally distilled into non-overlapping 50 °F cuts with boiling points between 300 and 850 °F. Another set of 18 distillate cuts were obtained with boiling points ranging between 138 and 1055 °F. Distillate cuts were assayed for mutagenic activity using the histidine reversion assay with Salmonella typhimurium strains TA98, TA100, TA1535 and TA1537, as well as for mammalian-cell transformation activity in the Syrian hamster embryo test, and DNA damage in the prophage induction assay. Samples were also separated into chemical class fractions by alumina column chromatography and analysed by high resolution gas chromatography so that the chemical composition of the cuts could be related to their relative activity in the different assays. In the mammalian cell transformation and microbial mutagenicity assays, significant activity was found almost exclusively in distillate cuts with components boiling > 700 °F, with the highest activity in the transformation assay observed for cuts > 800 °F. All of the distillate cuts showed increased levels of DNA damage as expressed by lambda prophage induction in Escherichia coli 8177. However, the greatest activity was associated with distillate cuts with boiling points in the 800 °F + range. Chemical analysis of the 50 °F distillate cuts showed a variety of polycyclic aromatic hydrocarbons (PAH) and amino-PAH compounds to be present in the distillate cuts boiling > 700 °F and essentially absent from cuts boiling < 700 °F. The sample set of non-overlapping (50 °F) cuts were reblended according to the proportions of each cut found in the original blend material. These reblended composites were then assayed to compare their activity with that predicted from the activities of the component distillate cuts. The reblending experiments indicated the microbial mutagenicity response was essentially additive. Mammalian cell transformation activities were non-additive, indicating a compositional effect on the expression of transforming agents in the complex mixture.  相似文献

We show here for the first time that a stable parallel double helix with Hoogsteen pairing can exist independently of the triple helix of which it is a component part. The experiments employ DNA oligonucleotides with mixed sequences of normal bases. These duplexes are distinct from previously reported ribopolynucleotide helices containing bulky substituents which prevent Watson-Crick pairing as well as from parallel duplexes with Donohue, or reversed Watson-Crick, pairing. Stoichiometry is established by mixing curves and gel electrophoresis. Tm depends linearly upon pH, increasing with acidity because of the need to protonate N3 of C. The Tm of the 20-mer studied here is 52 degrees C at pH 5.2 and 0.1 M NaCl. At pH above 6, the molecule rearranges to form an antiparallel duplex with imperfect Watson-Crick pairing and loops, and the Tm is then independent of pH. The CD spectrum of the parallel duplex is very similar to that of the corresponding triple helix but quite different from that of the Watson-Crick helix. The infrared spectrum in the double bond region closely resembles that of the triple helix but, as with the CD, is quite different from that of the Watson-Crick duplex. The infrared spectra of the duplex and triple helix are also nearly identical in the region form 800 to 1000 cm-1, which is sensitive to backbone conformation. Readers' eye movements were monitored as they read sentences containing lexically ambiguous words. The ambiguous words were either biased (one strongly dominant interpretation) or nonbiased. Readers' gaze durations were longer on nonbiased than biased words when the disambiguating information followed the target word. In Experiment 1, reading times on the disambiguating word did not differ whether the disambiguation followed the target word immediately or occurred several words later. In Experiment 2, prior disambiguation eliminated the long gaze durations on nonbiased target words but resulted in long gaze durations on biased target words if the context demanded the subordinate meaning. The results indicate that successful integration of one meaning with prior context terminates the search for alternative meanings of that word. This results in selective (single meaning) access when integration of a dominant meaning is fast (due to a biasing context) and identification of a subordinate meaning is slow (a strongly biased ambiguity with low-frequency meaning). The purpose of the paper is to provide quantitative characterization of metallic microneedles. Mechanical and fluid flow experiments were performed to evaluate the buckling force, the penetration force, and the pressure versus flow rate characteristics of the microneedles. The microneedle design variations characterized included varying the shaft lengths, varying the tip taper angles/geometries, and the inclusion of micromechanical barbs. The penetration force was found to range from 7.8 gF for a microneedle of shaft length 500 /spl mu/m, to 9.4 gF for a length of 1500 /spl mu/m, both with a tip taper angle of 30/spl deg/. Microneedles with a linear tip taper angle of 30/spl deg/ penetrated 95 +% of the time without failure. The microneedles with a 15/spl deg/ and 20/spl deg/ linear tip taper penetrated 10% and 25% of the time, respectively. The buckling force was found to be 98.4 gF for a 500 /spl mu/m long microneedle shaft, 72.3 gF for a needle of shaft length 1000 /spl mu/m, and 51.6 gF for a 1500 /spl mu/m long shaft. The results demonstrate that the penetration force was 7.9% of the buckling force for 500 /spl mu/m long shafts, 11.6% for a 1000 /spl mu/m long shaft, and 18.2% for a 1500 /spl mu/m long microneedle shafts. The microneedle fluid flow characteristics were studied. An inlet pressure of 49.0 Pa was required for a flow rate of 1000 /spl mu/L/h and 243.0 Pa for a flow rate of 4000 /spl mu/L/h using air as the fluid medium. For water, an average pressure of 30.0 kPa was required for a flow rate of 1000 /spl mu/L/h and 106.0 kPa for a flow rate of 4000 /spl mu/L/h.  相似文献

The microbiology and clinical features of empyema were studied retrospectively in 197 patients whose specimens yielded bacterial growth after inoculation for aerobic and anaerobic bacteria. Three hundred forty-three organisms (216 aerobic or facultative and 127 anaerobic organisms) were isolated. Aerobic bacteria were isolated in 127 (64 percent) patients, anaerobic bacteria in 25 (13 percent), and mixed aerobic and anaerobic bacteria in 45 (23 percent). The predominant aerobic or facultative organisms were Streptococcus pneumoniae (70 isolates), Staphylococcus aureus (58), Escherichia coli (17), Klebsiella pneumoniae (16), and Haemophilus influenzae (12). The predominant anaerobes were pigmented Prevotella and Porphyromonas species (24), Bacteroides fragilis group (22), anaerobic cocci (36), and Fusobacterium species (20). beta-Lactamase-producing organisms were recovered in 49 (38 percent) of 128 tested specimens. These included all 42 tested S aureus and 15 B fragilis group, 4 of 9 K pneumoniae, 3 of 9 H influenzae, 3 of 8 pigmented Prevotella and Porphyromonas species, and 2 of 6 E coli. Most patients from whom S pneumoniae and H influenzae were recovered had pneumonia, and most patients with S aureus had pneumonia, aspiration pneumonia, and lung abscesses. The recovery of anaerobic bacteria was mostly associated with the concomitant diagnosis of aspiration pneumonia, and lung, subdiaphragmatic, dental, and oropharyngeal abscesses. These data highlight the importance of anaerobic bacteria in selected cases of empyema.  相似文献

The composition and solubility properties of eight ammonium and potassium orthophosphites and hypophosphites were determined to evaluate the potential of these classes of materials for increasing the plant nutrient content of liquid fertilizers. Phase relationships for the systems (NH4)2O-P2O3-H2O, K2O-P2O3-H2O, (NH4)2O-P2O-H2O, and K2O-P2O-H2O were determined along with the crystallographic properties (X-ray and optical) of the solid phases. Invasive species disrupt landscape patterns and compromise the functionality of ecosystem processes. Non-native saltcedar poses significant threats to native vegetation and groundwater resources in the southwestern U.S. and Mexico, and quantifying spatial and temporal distribution patterns is essential for monitoring its spread. Considerable research focuses on determining the accuracy of various remote sensing techniques for distinguishing saltcedar from native woody riparian vegetation through sub-pixel, or soft classifications. However, there is a lack of research quantifying spatial distribution patterns from these classifications, mainly because landscape metrics, which are commonly used to statistically assess these patterns, require bounded classes and cannot be applied directly to soft classifications. This study tests a new method for discretizing sub-pixel data to generate landscape metrics using a continuum of fractional cover thresholds. The developed approach transforms sub-pixel classifications into discrete maps compliant with metric terms and computes and interprets metric results in the context of the region to explain patterns in the extent, distribution, and connectivity of saltcedar in the Rio Grande basin. Results indicate that landscape metrics are sensitive to sub-pixel values and can vary greatly with fractional cover. Therefore spectral unmixing should be performed prior to metric calculations. Analysis of metric trends provides evidence that saltcedar has expanded away from the immediate riparian zones and is displacing native vegetation. This information, coupled with control management strategies, can be used to target remediation activities along the Rio Grande.  相似文献

This paper presents a methodology to solve the Just-in-Time (JIT) sequencing problem for multiple product scenarios when set-ups between products are required. Problems of this type are combinatorial, and complete enumeration of all possible solutions is computationally prohibitive. Therefore, Genetic Algorithms are often employed to find desirable, although not necessarily optimal, solutions. This research, through experimentation, shows that Genetic Algorithms provide formidable solutions to the multi-product JIT sequencing problem with set-ups. The results also compare favourably to those found using the search techniques of Tabu Search and Simulated Annealing.  相似文献

A critical component of landscape dynamics is the recovery of vegetation following disturbance. The objective of this research was to characterize the forest recovery trends associated with a range of spectral indicators and report their observed performance and identified limitations. Forest disturbances were mapped for a random sample of three major bioclimate zones of North American boreal forests. The mean number of years for forest to recover, defined as time required to for a pixel to attain 80% of the mean spectral value of the 2 years prior to disturbance, was estimated for each disturbed pixel. The majority of disturbed pixels recovered within the first 5 years regardless of the index ranging from approximately 78% with normalized burn ratio (NBR) to 95% with tasselled cap greenness (TCG) and after 10 years more than 93% of disturbed pixels had recovered. Recovery rates suggest that normalized differenced vegetation index (NDVI) and TCG saturate earlier than indices that emphasize longer wavelengths. Thus, indices such as NBR and the mid-infrared spectral band offer increased capacity to characterize different levels of forest recovery. The mean length of time for spectral indices to recover to 80% of the pre-disturbance value for pixels disturbed 10 or more years ago was highest for NBR, 5.6 years, and lowest for TCG, 1.7 years. The mid-infrared spectral band had the greatest difference in recovered pixels among bioclimate zones 1 year after disturbance, ranging from approximately 42% of disturbed pixels for the cold and mesic bioclimate zone to 60% for the extremely cold and mesic bioclimate zone. The cold and mesic bioclimate zone had the longest mean years to recover ranging from 1.9 years for TCG to 4.2 years for NBR, while the cool temperate and dry bioclimate zone had the shortest mean years to recover ranging from 1.6 years for TCG to 2.9 years for NBR suggesting differences in pre-disturbance conditions or successional processes. The results highlight the need for caution when selecting and interpreting a spectral index for recovery characterization, as spectral indices, based upon the constituent wavelengths, are sensitive to different vegetation conditions and will provide a variable representation of structural conditions of forests.  相似文献
