Objective To investigate the possible role of brain-derived neurotrophic factor(BDNF)in the pathophysiology of attempted suicide in depressive episode patients.Methods The subjects were 23 depressive episode patients who had recently attempted suicide.24 depressive episode patients without suicide attempt.and 30 sex-age-and education matched healthy controls.The Hamihon Depression Rating Scale-24 items(HAMD24),Beck Helpless Rating Scale(BHS)and Self-rating Idea of Suicide Scale (SIOSS)was used to evaluate the severitv of symptoms.The seruln level of BDNF was detected with sandwich ELISA.Pearson correlation coefficients wag used to measure the correlations between serum BDNF and the related illness variables.Results There were significant difierences in the serum level of BDNF between patients with attempted suicide,pailents without suicide attempt and normal controls(F=21.927,P<0.05).Post hoc analysis showed the serum BDNF levels in patients with attempted suicide [(57 ±16)ng/L]were lower than that in patients without[(75±28)ns/L;P<0.05],and were lower in patients without suicide attempt[(75 ±28)ng/L]than that in normal controls[(111 ±39)ng/L;P<0.01].In patients with attempted suicide,the serum BDNF was negatively correlated to the duration of depressive episode(r=-0.541,P=0.008),the mean BHS score(r=-0.494,P=0.017)and the mean SIOSSscore(r=-0.754.P=0.000).Conclusion The results suggest that the decreased serum brain-derived neurotrophic factor is related to the pathophysiology of attempted suicide in depressive episode patients.  相似文献

背景:由于分子遗传学技术的发展,关于精神疾病与分子遗传的研究日趋增多,但结论不尽一致,对5-羟色胺2A受体基因T102C基因型频率颁布是否有地区、种族差异尚需要更多证实。 目的:分析精神分裂症患者5-羟色胺2A受体基因T102C基因型频率在地理上的分布。 设计:以精神分裂症患者为观察对象抽样调查。 单位:常州和平医院精神科。 对象:实验于1999—01/2003-08在解放军第102医院完成。选取符合中国精神疾病分类和诊断标准第三版关于精神分裂症诊断标准的患者177例,来自不同省区汉族军人,年龄18-45岁,病程1个月~20年。方法:采集1l7例精神分裂症患者的血样进行聚合酶链反应检测,并比较不同省区正常对照人群5-羟色胺2A受体基因型频率的分布有何差异,应用常规苯酚氯仿法抽提DNA,目的DNA的蒙合酶链反应扩增及电泳检测,5-羟色胺2A受体基因T102C基因片断扩增的引物序列:5’-TCT GCT ACA AGT TCT GGC TT-3’,5’-CTG CAG CTT TTT CTC TAG GG-3’;采用50μL反应体系,DNA 0.5μg,引物50pmol,TagDNA酶2U加dNTP至终浓度200μmol/L。 主要观察指标:精神分裂症患者DNA电泳检测及5-羟色胺2A受体基因型频率分布。 结果:5-羟色胺2A受体各基因型频率分布为:A1A1,0.07~0.33;A1A2,0.50-0.72;A2A2,0.17-0、29。基因型频率在不同地区的相关分析:A1A1,X^2=4、44,P=0.6171,P〉0.05;A1A2,X^2=1.14,P=0.9422,P〉0.05;A2A2,在0.93,P=0.9857,P〉0.05。 结论:117例精神分裂症患者5-羟色胺2A受体基因T102C基因型频率的地理分布比较均一。  相似文献

目的 分化抑制因子3(inhibitor of differentiation 3,Id3)是重要的细胞生长调控分子,在细胞增殖、分化等过程中发挥重要作用.文中探讨在大肠杆菌中表达及纯化重组人Id3蛋白,制备抗人Id3单克隆抗体(McAb),以进一步研究Id3的生物学功能及其临床应用.方法 将重组表达载体Id3/pET32a转化大肠杆菌BL21(DE3),用异丙基β-D硫代半乳糖苷(IPTG)诱导转化菌表达Id3重组融合蛋白(Trx-His-hId3),通过镍亲和层析柱纯化Trx-His-hId3.以纯化的Id3重组蛋白为免疫原,免疫BALB/c小鼠,采用杂交瘤技术制备Id3 McAb,应用酶联免疫吸附试验(ELISA)及Western blot进行抗体鉴定.结果 经免疫小鼠、脾细胞分离、细胞融合及克隆筛选等步骤,筛选出2株分泌抗人Id3 McAb的杂交瘤细胞株,分别命名为McAb 6G7和6G9,免疫球蛋白亚类鉴定均为IgG1亚类.Western blot分析表明,McAb 6G7和6G9能与重组人Id3蛋白特异性结合.间接ELISA法检测2株细胞冻融前后所产生上清中的抗体效价均分别为1∶ 1000、1∶ 5000.结论 成功制备了抗人Id3 McAb,为研究该蛋白的功能提供了有利工具.  相似文献

目的:探讨氯氮平对精神分裂症患者血浆神经肽Y(NPY)及体质量的影响。方法:30例女性精神分裂症患者(氯氮平组),于氯氮平治疗前、治疗4周,分别测定体质量、血浆NPY水平,并与30例健康成年女性(对照组)比较。结果:氯氮平组治疗前血浆NPY(pg/ml)与对照组比较,差异无统计学意义(58.9±12.3,60.9±14.0,P>0.05);治疗后,氯氮平组体质量及血浆NPY均高于治疗前,差异有统计学意义(63.3±13.3,60.9±14.0;159.2±66.1,129.0±66.5,P<0.05)。治疗后、治疗前血浆NPY的差值,与治疗前后体质量的差值呈正相关(r=0.446,P=0.013)。结论:氯氮平引起体质量增加的同时,伴有血浆NPY增高,后者可能参与了氯氮平所致的体质量增加形成机制。  相似文献

目的 探讨精神分裂症患者冲动、自杀行为与血浆瘦素及胆固醇之间的相关性.方法 106例精神分裂症首次发病患者用简明精神病评定量表(BPRS)、阳性和阴性症状量表(PANSS)、汉密尔顿抑郁量表(HAMD24)及Beck绝望量表(BHS)进行评定;按有无冲动、自杀行为分为自杀组(24例)、冲动组(31例)、非自杀非冲动组(以下简称患者对照组,51例),32名体检职工为正常对照组,所有入组对象(患者在服药前)测定体质量指数(BMI)、血浆瘦素和总胆固醇.结果 (1)正常对照组[(4.8±0.9)mmol/L]和患者对照组[(4.3±1.1)mmol/L]总胆固醇均高于自杀组[(3.7±1.0)mmol/L]及冲动组[(3.6±1.0)mmoL/L;P<0.05~0.01],而冲动组与自杀组之间、正常对照组与患者对照组之间的差异无统计学意义(P>0.05);正常对照组血浆瘦素[(13.4±6.7)μg/L]高于患者对照组[(8.9±3.8)μg/L]、自杀组[(6.7±2.6)μg/L]及冲动组[(5.6±4.2)μg/L;P<0.05~0.01],患者对照组高于自杀组及冲动组(P<0.05),而自杀组与冲动组的差异无统计学意义(P>0.05).(2)4组的血浆瘦素及总胆固醇均与BMI呈正相关(r=0.49~0.64;P<0.01);自杀组、冲动组的血浆瘦素和总胆固醇与PANSS阳性分、PANSS攻击分、BPRS分、HAMD分、BHS分均呈显著负相关(r=-0.35~-0.72;P<0.05~0.01);患者对照组的血浆瘦素、总胆固醇与PANSS攻击分、BPRS分、HAMD分、BHS分均呈显著负相关r=-0.29~-0.48;P<0.05~0.01).结论 精神分裂症患者的冲动、自杀行为及疾病严重程度与血浆瘦素及总胆固醇有一定的相关性.  相似文献
