Gravel constitutes the filter medium in subsurface flow constructed wetlands (SSF CWs) and its porosity and hydraulic conductivity decrease over time (clogging), limiting the lifespan of the systems. Using gravel of poor quality accelerates clogging in wetlands. In this study, gravel samples from six different wetland systems were compared with regards to their mineral composition and mechanical resistance properties. Results showed that both mineralogy and texture are related to mechanical resistance. Accordingly, gravel with high content of quartz (> 80%) showed a lower percentage of broken particles (0.18-1.03%) than those with lower content of quartz (2.42-4.56% media broken). Although granite is formed by high durability minerals, its non-uniform texture results in a lower resistance to abrasion (ca. 10% less resistance than calcareous gravel). Therefore, it is recommended to use gravels composed mainly of quartz or, when it is not available, limestone gravels (rounded and uniform) are recommended instead. The resistance to abrasion (LAA test) seems to be a good indicator to determine the mechanical properties of gravels used in CWs. It is recommended to use gravels with LAA below 30% in order to avoid a rapid clogging due to gravel crumbling and subsequent mineral solids accumulation.  相似文献

Here we report results from a multi-laboratory (n = 11) evaluation of four different PCR methods targeting the 16S rRNA gene of Catellicoccus marimammalium originally developed to detect gull fecal contamination in coastal environments. The methods included a conventional end-point PCR method, a SYBR® Green qPCR method, and two TaqMan® qPCR methods. Different techniques for data normalization and analysis were tested. Data analysis methods had a pronounced impact on assay sensitivity and specificity calculations. Across-laboratory standardization of metrics including the lower limit of quantification (LLOQ), target detected but not quantifiable (DNQ), and target not detected (ND) significantly improved results compared to results submitted by individual laboratories prior to definition standardization. The unit of measure used for data normalization also had a pronounced effect on measured assay performance. Data normalization to DNA mass improved quantitative method performance as compared to enterococcus normalization. The MST methods tested here were originally designed for gulls but were found in this study to also detect feces from other birds, particularly feces composited from pigeons. Sequencing efforts showed that some pigeon feces from California contained sequences similar to C. marimammalium found in gull feces. These data suggest that the prevalence, geographic scope, and ecology of C. marimammalium in host birds other than gulls require further investigation. This study represents an important first step in the multi-laboratory assessment of these methods and highlights the need to broaden and standardize additional evaluations, including environmentally relevant target concentrations in ambient waters from diverse geographic regions.  相似文献

This paper deals with the weighted earliness-tardiness resource-constrained project scheduling problem with minimum and maximum time lags (WET-RCPSP/max). The problem consists of scheduling the activities of a project subject to prescribed resource and temporal constraints such that the total weighted deviation of the activities' completion times from prescribed due dates is minimized. Key applications are planning of just-in-time production and reactive scheduling. For the (approximative) solution of the WET-RCPSP/max, we present a population-based iterated-local-search heuristic. We also report the results of an experimental performance analysis where this heuristic outperformed state-of-the-art methods.  相似文献

OBJECTIVES: In neurogenic dysphagia a good cough is important for airway protection. If triggering of cough, or its effectiveness, is impaired this might result in an increased aspiration risk. Capsaicin, an agent which induces cough through sensory nerve stimulation, was used to test cough sensitivity in groups of patients with and without neurogenic dysphagia. METHODS: On the basis of swallowing speed (ml/s) in a validated water test 28 alert neurological inpatients (16 women, aged 22-71 years) were classified into 13 with abnormal and 15 with normal swallowing (median swallowing speed 23% and 99%, median volume/swallow 43% and 106% of that predicted for age and sex respectively: p<0.001). Capsaicin nebulised on air in saline was inhaled via a low resistance valve using a mouthpiece and noseclip. Up to seven incremental concentrations of capsaicin ranging from 0.07-20.0 x 10(-4) mol/l were each inhaled for up to a minute. A pneumotachograph connected to the expiratory limb gave a paper recording of expiratory air flow. Coughs were recorded as high flow expirations of short duration. Capsaicin concentrations at first cough (threshold) were recorded; concentrations at frequencies of 10 and 20 coughs/minute were interpolated from the dose-response curve. RESULTS: Cough threshold tended to be lower in those with abnormal swallowing (non-significant): the (log) concentration of capsaicin producing 10 or 20 coughs/ minute also tended to be lower (p=0.12 and 0.07 respectively) in those with abnormal swallowing. CONCLUSION: Contrary to expectation, these results suggest that cough responsiveness is enhanced in alert patients with neurogenic dysphagia even after allowing for diagnostic category, the possible presence of a bulbar upper motor neuron lesion, or voluntary respiratory capacity. It is concluded that these patients with neurogenic dysphagia do not have a reduced sensitivity of cough triggering.  相似文献

A new iron-based T 1 contrast agent consisting of a complex of iron ions coordinated to phosphate and amine ligands (Fe(phos) in short) has been characterized by spectroscopic and magnetic measurements. NMR relaxation studies showed r 1 values to be dependent on the phosphate salt concentration, K2HPO4, present in the medium. r 1 reaches a maximum value of 2.5 mM?1 s?1 for measurements carried out at 7 T and 298 K. 31P MRS, Mössbauer spectroscopy and magnetic measurements of Fe(phos) solutions suggest paramagnetic Fe3+ ions present in the studied iron–phosphate complex. In vitro and in vivo toxicity experiments with C6 cells and CD1 mice, respectively, demonstrated lack of toxicity for Fe(phos) at the highest dose tested in the MRI experiments (12 mM iron for C6 cells and 0.32 mmol iron/kg for mice). Finally, T 1 weighted images of brain tumours in mice have shown positive contrast enhancement of Fe(phos) for tumour afflicted regions in the brain.  相似文献

This paper describes a real problem in a market-driven medium sized foundry delivering a wide range of castings to different markets. The problem consists of finding an efficient production plan to schedule the different processes (moulding, furnacing, cutting, tooling, etc.) needed to the manufacture of the pieces. Different objectives and resources and technical constraints must be taken into account. To solve this problem we have first developed a more classical integer linear programming approach based on a rolling horizon strategy. The most innovative contribution of the paper is that it models the problem as a project scheduling problem. Based on this model we present a metaheuristic algorithm that adapts techniques from the area. Computational experiments comparing both approaches are provided on instances created by a generator simulating real instances.  相似文献

The multi-mode resource-constrained project scheduling problem with minimum and maximum time lags MRCPSP/max is a very general project scheduling problem with multiple execution modes per activity, renewable and non-renewable resources and minimum and maximum time lags between activities. In this paper, we describe SA-EVA, an algorithm for the problem. SA-EVA first searches for the best mode for each activity, without considering renewable resources. In this phase a simulated annealing is applied. Once a mode vector has been chosen, the problem reduces to the RCPSP/max, which SA-EVA solves with EVA, an algorithm designed in Ballestín et al. The extensive application of emerging technologies is revolutionizing warehouse management. These technologies facilitate working with complex and powerful warehouse management models in which products do not have assigned fixed locations (random storage). Random storage allows the utilization of the available space to be optimized. In this context, and motivated by a real problem, this article presents a model that looks for the optimal allocation of goods in order to maximize the storage space availability within the restrictions of the warehouse. For the proposed model a construction method, a local search algorithm and different metaheuristics have been developed. The introduced algorithms can also be used for other purposes such as to assess when and how it is convenient to perform relocation of stored items to improve the current level of storage space availability. Computational tests performed on a set of randomly generated and real warehouse instances show the effectiveness of the proposed methods.  相似文献

Constrained binding peptides (peptide aptamers) may serve as tools to explore protein conformations and disrupt protein-protein interactions. The quality of the protein scaffold, by which the binding peptide is constrained and presented, is of crucial importance. SQT (Stefin A Quadruple Mutant-Tracy) is our most recent development in the Stefin A-derived scaffold series. Stefin A naturally uses three surfaces to interact with its targets. SQT tolerates peptide insertions at all three positions. Peptide aptamers in the SQT scaffold can be expressed in bacterial, yeast and human cells, and displayed as a fusion to truncated pIII on phage. Peptides that bind to CDK2 can show improved binding in protein microarrays when presented by the SQT scaffold. Yeast two-hybrid libraries have been screened for binders to the POZ domain of BCL-6 and to a peptide derived from PBP2', specific to methicillin-resistant Staphylococcus aureus. Presentation of the Noxa BH3 helix by SQT allows specific interaction with Mcl-1 in human cells. Together, our results show that Stefin A-derived scaffolds, including SQT, can be used for a variety of applications in cellular and molecular biology. We will henceforth refer to Stefin A-derived engineered proteins as Scannins.  相似文献
