Loss of β-cell mass and function can lead to insufficient insulin levels and ultimately to hyperglycemia and diabetes mellitus. The mainstream treatment approach involves regulation of insulin levels; however, approaches intended to increase β-cell mass are less developed. Promoting β-cell proliferation with low-molecular-weight inhibitors of dual-specificity tyrosine-regulated kinase 1A (DYRK1A) offers the potential to treat diabetes with oral therapies by restoring β-cell mass, insulin content and glycemic control. GNF4877, a potent dual inhibitor of DYRK1A and glycogen synthase kinase 3β (GSK3β) was previously reported to induce primary human β-cell proliferation in vitro and in vivo. Herein, we describe the lead optimization that lead to the identification of GNF4877 from an aminopyrazine hit identified in a phenotypic high-throughput screening campaign measuring β-cell proliferation.  相似文献

Several days after heart surgery, a patient discovered his upper right canine tooth had broken at the root. Such tooth damage, recognized post-operatively, is usually assumed to be caused by blunt mechanical force from an instrument used by the anesthesiologist during placement of a breathing tube at the start of surgery. In this case, the patient had saved the crown portion of the broken tooth, and it was possible to examine the root fracture characteristics. The curvature and direction of the crack path and natural tooth situation suggested that failure could be described through a cantilever beam model. This was confirmed when a whole extracted sample tooth was embedded and broken by a measured force in a manner consistent with the model. The resulting fracture surface matched that of the patient’s broken canine tooth. However, the high load and force direction necessary to fracture the root was inconsistent with forces applied during the anesthesia procedure. The failure analysis and further investigation indicated tooth clenching on the breathing tube during recovery was the likely cause of fracture. This paper presents an alternate explanation for intubation-related dental injury, demonstrates the practicality of fractographic analysis of biological materials, and introduces a methodology for simulating in vitro tooth settings for mechanical testing.  相似文献

In many legal proceedings, fact finders scrutinize the demeanor of a defendant or witness, particularly his or her nonverbal behavior, for indicators of deception. This meta-analysis investigated directly observable nonverbal correlates of deception as a function of different moderator variables. Although lay people and professionals alike assume that many nonverbal behaviors are displayed more frequently while lying, of 11 different behaviors observable in the head and body area, only 3 were reliably associated with deception. Nodding, foot and leg movements, and hand movements were negatively related to deception in the overall analyses weighted by sample size. Most people assume that nonverbal behaviors increase while lying; however, these behaviors decreased, whereas others showed no change. There was no evidence that people avoid eye contact while lying, although around the world, gaze aversion is deemed the most important signal of deception. Most effect sizes were found to be heterogeneous. Analyses of moderator variables revealed that many of the observed relationships varied as a function of content, motivation, preparation, sanctioning of the lie, experimental design, and operationalization. Existing theories cannot readily account for the heterogeneity in findings. Thus, practitioners are cautioned against using these indicators in assessing the truthfulness of oral reports. OBJECTIVE: The purpose of our study was to test the hypothesis that surfactant dosing through a proximal sideport adapter on an endotracheal tube leads to more dosing-associated hypoxemia compared with a method of dosing that uses a double-lumen endotracheal tube. STUDY DESIGN: Using adequate sample size to compare significant changes in O2 saturation (power > 0.8, alpha < 0.05) we enrolled 36 infants with respiratory distress syndrome in this randomized trial. A 10% change in O2 saturation was considered clinically significant. Nineteen infants received 38 doses of surfactant through the sideport adapter. Seventeen infants received 31 doses of surfactant through the nonventilation lumen of a double-lumen endotracheal tube. Two main outcome measures were assessed: time-averaged O2 saturation values 30 minutes after dosing and the largest absolute fall in O2 saturation for each patient. RESULTS AND CONCLUSIONS: Time-averaged O2 saturation measures were higher in the proximal sideport group (p = 0.02), but the magnitude of difference was probably not clinically significant. No significant difference was detected between groups when we compared largest absolute drop in O2 saturation. Secondary analyses found no effect of birth weight or dose number (second vs third dose) on either outcome measure.  相似文献

Chicken acidic leucine-rich EGF-like domain containing brain protein (CALEB) was identified by combining binding assays with immunological screens in the chicken nervous system as a novel member of the EGF family of differentiation factors. cDNA cloning indicates that CALEB is a multidomain protein that consists of an NH2-terminal glycosylation region, a leucine-proline-rich segment, an acidic box, a single EGF-like domain, a transmembrane, and a short cytoplasmic stretch. In the developing nervous system, CALEB is associated with glial and neuronal surfaces. CALEB is composed of a 140/130-kD doublet, an 80-kD band, and a chondroitinsulfate-containing 200-kD component. The latter two components are expressed in the embryonic nervous system and are downregulated in the adult nervous system. CALEB binds to the extracellular matrix glycoproteins tenascin-C and -R. In vitro antibody perturbation experiments reveal a participation of CALEB in neurite formation in a permissive environment.  相似文献

We investigated changes in the anterior tibial muscle during lengthening of the lower leg in rabbits. In 37 rabbits, an osteotomy of the right middle tibia was performed and was fixed by a unilateral external fixator. The rabbits were randomized into 6 groups. In groups 1, 2, and 3 the tibiae were distracted 0.5 mm/day. In groups 1 and 2, the rabbits were killed after 14 and 28 days of distraction, respectively, and in group 3 after 28 days of distraction, followed by 14 days of rest. Groups 1a, 2a, and 3a served as controls. They were treated similarly as groups 1, 2, and 3, but no distraction was performed. Proliferating cell nuclei were labeled with 5-bromo-2-deoxyuridine and were identified by immunohistochemical staining. The weight of the muscle was measured. During bone lengthening the muscle showed signs of growth, as indicated by increasing weight and number of proliferating cell nuclei. This was observed only during lengthening and it ceased when the lengthening was stopped.  相似文献

Zusammenfassung In grünem (Taiwan Gunpowder) und schwarzem (Ceylon high OP) Tee sind beträchtliche Mengen an Aromastoffen in glykosidischer Bindung vorhanden; ihr Anteil ist sogar höher als der an freien Aromastoffen. Hauptkomponenten sind in beiden Teesorten cis-3-Hexen-1-ol, Benzylalkohol und 2-Phenylethanol in gebundener Form, dazu im Schwarztee Linalool und im grünen Tee Geraniol. Die unterschiedlichen Anteile dieser beiden Terpenalkohole spiegeln die unterschiedlichen Tee-Varietäten wieder, denen die untersuchten Teesorten angehören: der Ceylon-Tee ist der Varietätassamica zuzuordnen, der Tee aus Taiwan der Varietätsinensis. Die Zusammensetzung der aus gebundenen Formen freisetzbaren Aromastoffe war ähnlich komplex wie die der freien Aromastoffe, wobei die Unterschiede zwischen den beiden Teesorten auf den verschiedenen Herstellungsverfahren beruhen. Das Auftreten gebundener Formen solcher Aromastoffe, die nicht originär vorhanden sind, sondern erst während der Welk- oder der Fermentationsphase gebildet werden, impliziert das Vorhandensein glykosidierender Aktivitäten im Teeblatt noch in relativ späten Stadien der Schwarzteeherstellung. Die Untersuchung der gebundenen Anteile an Aromastoffen trägt neben der Analyse der freien Aromastoffe wesentlich zum Verständnis der Reaktionen im Teeblatt wahrend der verschiedenen Produktionsstadien bei.
Bound flavour compounds in plants 2. Part. Free and bound flavour compounds in green and black tea
Summary Green and black tea contains considerable amounts of flavour compounds which are present in glycosidic bonds, their portion being even higher than that of free flavour compounds. The main components in both of these tea varieties are 3-hexene-1-ol, benzylalcohol and 2-phenylethanol, which are present in the bound forms, and additionally linalool in black tea and geraniol in green tea. The different amounts of these terpene alcohols reflect the different varieties the teas under investigation belong to: the Ceylon tea belongs to the variety assamica, and the Taiwan tea to the variety sinensis. The composition of the flavour compounds liberated from bound forms had a similar complexity to that of the free flavour compounds, the difference being based upon the different production procedures. The occurrence of bound forms of such components that are not originally present but are produced during the withering or fermentation stage implies that glycosidating activities are still present in the tea leaf, even in a relatively late stage of the black tea production process. The investigation of the bound parts of flavour compounds delivers a valuable contribution to a better understanding of the reactions occurring in the tea leaf during the different stages of tea production.

In neonatally allylestrenol treated animals in adult age a single benzpyrene treatment significantly decreases the female and significantly increases the male rat's sexual activity. Three month old females display the negative sexual behavioral effect of neonatal allylestrenol treatment less than the six month old ones. The benzpyrene treatment in adult age decreases the sexual activity of male rats. The experiments call the attention to the modifying effect of perinatal steroid treatments to similar exposure in adult age.  相似文献

We determined the effects of aspirin and a novel thromboxane A2/prostaglandin endoperoxide (TP)-receptor antagonist, BMS-180291, on thrombosis and bleeding times in skin and mesenteric arteries. In anesthetized rats, occlusive thrombosis was induced in the carotid artery by topical application of ferrous chloride and in the vena cava by blood flow stasis combined with either infusion of thromboplastin or hypotonic saline. Aspirin (1, 10, and 50 mg/kg) did not reduce arterial or venous thrombus weight significantly. BMS 180,291 (150 micrograms/kg/min) decreased arterial thrombus weight and hypotonic saline-induced caval thrombus weight by 58 and 57%, respectively. BMS-180291 lacked antithrombotic activity at a lower dose (50 micrograms/kg/min) and failed to inhibit thromboplastin-induced caval thrombosis. BMS-180291 (150 micrograms/kg/min) significantly reduced arterial thrombus weight by 40% when plasma epinephrine concentration was increased to 5 ng/ml. BMS-180291 and aspirin produced increases of only < or = 30% in bleeding times. These results demonstrate that BMS-180291 has antithrombotic activity in experimental aspirin-resistant arterial and venous thrombosis. Both aspirin and BMS-180291 have only modest effects on small artery hemostasis in rats.  相似文献
