We have examined the 3'-terminal sequence of the "small" structural ribosomal RNA ("13S") of hamster cell mitochondria, using a procedure involving [3H]isoniazide labeling of samples subjected to sequential periodate oxidation and beta-elimination. The terminus was found to be PyUAUUAOH, which is similar, but not identical, to the corresponding terminus of eukaryotic cytoplasmic 18S rRNA.  相似文献

Microbial-, and chemical-based burden of disease associated with lack of access to safe water continues to primarily impact developing countries. Cost-effective health risk-mitigating measures, such as of solar disinfection applied to microbial-contaminated water stored in plastic bottles have been increasingly tested in developing countries adversely impacted by epidemic water-borne diseases. Public health concerns associated with chemical leaching from water packaging materials led us to investigate the magnitude and variability of antimony (Sb) and bromine (Br) leaching from reused plastic containers (polyethylene terephthalate, PET; and polycarbonate, PC) subject to UV and/or temperature-driven disinfection. The overall objective of this study was to determine the main and interactive effects of temperature, UV exposure duration, and frequency of bottle reuse on the extent of leaching of Sb and Br from plastic bottles into water. Regardless of UV exposure duration, frequency of reuse (up to 27 times) was the major factor that linearly increased Sb leaching from PET bottles at all temperatures tested (13-47 °C). Leached Sb concentrations (∼360 ng L−1) from the highly reused (27 times) PET bottles (minimal Sb leaching from PC bottles, <15 ng L−1) did not pose a serious risk to human health according to current daily Sb acceptable intake estimates. Leached Br concentrations from both PET and PC containers (up to ∼15 μg L−1) did not pose a consumer health risk either, however, no acceptable daily dose estimates exist for oral ingestion of organo-brominated, or other plasticizers/additives compounds if they were to be found in bottled water at much lower concentrations. Additional research on potential leaching of organic chemicals from water packaging materials is deemed necessary under relevant environmental conditions.  相似文献

We have recently developed approaches for the generation of encephalitogenic T cell clones from mouse strains considered resistant to experimental allergic encephalomyelitis (EAE). By allowing for the direct use of knockout and mutant strains of mice, such clones allow for the efficient characterization of the relevance of specific gene products in the effector phase of EAE. Recent studies have suggested that Fas/FasL-mediated cell death may play a role in the pathogenesis of MS. To assess the role of Fas/FasL in EAE, we have tested the ability of wild-type C57BL/6-derived, encephalitogenic T cell clones to mediate adoptively transferred EAE in Fas-deficient C57BL/6-lpr mice. We now report that mice with the lpr mutation are fully susceptible to the adoptive transfer of EAE. Our results suggest that Fas/FasL-mediated cell death in the central nervous system does not play an integral role in the effector phase of acute EAE.  相似文献

This paper reports the results of a study on the effect of the cooling rate during solidification on the shear creep and low cycle shear fatigue behavior of 60 Sn/40 Pb solder joints, and on bulk solder tensile properties. Solder joints were made with three different initial microstructures by quenching, air-cooling and furnace-cooling. They have similar steady-state strain rates under creep at relatively high shear stresses (i.e. in the matrix creep region) but creep at quite different strain rates at lower shear stresses (i.e. in the grain boundary creep region). These results are ascribed to the refined grain size and less lamellar phase morphology that results on increasing the cooling rate. Tensile tests on bulk solders that were cold-worked, quenched and furnace-cooled show that a faster cooling rate decreases the ultimate strength and increases the ductility at low strain rates. The fatigue life of quenched solder joints is shown to be longer than that of the furnace-cooled joints.  相似文献

A multicentered study was performed to determine the mortality rate of patients with multiple injuries with major pelvic and long bone fractures who have early total care of their injuries. A 2-year review of patients with ISSs > or = 18 with major fractures treated at the trauma centers in Buffalo, New York, Camden, New Jersey, Nashville, Tennessee, Baltimore, Maryland, Tampa, Florida, and Seattle, Washington was performed. This group of 676 patients was compared with a similar group of 906 patients from the American College of Surgeons' Multiple Trauma Outcome Study. Mortality was significantly reduced in the patients who had early total care of all their injuries including fracture stabilization for patients less than 50 years of age and those 50 years and older. In a subgroup of patients less than 50 years of age and an ISS of 18-34 and 35-45 there was a mortality reduction from 11.8% to 5.1% and from 25.8% to 11.5%, respectively, when the fractures were managed acutely. Similar reductions in mortality were found in the patients 50 years of age and older with early fracture stabilization with a reduction from 26.4% to 8% in patients with ISSs of 18-24 and a reduction from 42.3% to 18.4% in the patients with ISSs of 35-45. This study clearly shows the additional benefit of early fracture stabilization in reducing mortality rates in the patient with multiple injuries.  相似文献

Human galanin is a 30 amino acid neuropeptide that elicits arange of biological activities by interaction with G protein-coupledreceptors. We have generated a model of the human GALR1 galaninreceptor subtype (hGALR1) based on the alpha carbon maps offrog rhodopsin and investigated the significance of potentialcontact residues suggested by the model using site-directedmutagenesis. Mutation of Phe186 within the second extracellularloop to Ala resulted in a 6-fold decrease in affinity for galanin,representing a change in free energy consistent with hydrophobicinteraction. Our model suggests interaction between Phe186 ofhGALR1 and Ala7 or Leu11 of galanin. Receptor subtype specificitywas investigated by replacement of residues in hGALR1 with thecorresponding residues in hGALR2 and use of the hGALR2-specificligands hGalanin(2–30) and [D-Trp2]hGalanin(1–30).The His267Ile mutant receptor exhibited a pharmacological profilecorresponding to that of hGALR1, suggesting that His267 is notinvolved in a receptor–ligand interaction. The mutationPhe115Ala resulted in a decreased binding affinity for hGalaninand for hGALR2-specific analogues, indicating Phe115 to be ofstructural importance to the ligand binding pocket of hGALR1but not involved in direct ligand interaction. Analysis of Glu271Trpsuggested that Glu271 of hGALR1 interacts with the N-terminusof galanin and that the Trp residue in the corresponding positionin hGALR2 is involved in receptor subtype specificity of binding.Our model supports previous reports of Phe282 of hGALR1 interactingwith Trp2 of galanin and His264 of hGALR1 interacting with Tyr9of galanin.  相似文献
