The increasing emergence of fungicide-resistant pathogens requires urgent solutions for crop disease management. Here, we describe a structural investigation of new fungicides obtained by combining strobilurin and succinate dehydrogenase inhibitor pharmacophores. We identified compounds endowed with very good activity against wild-type Pyricularia oryzae, combined in some cases with promising activity against strobilurin-resistant strains. The first three-dimensional model of P. oryzae cytochrome bc1 complex containing azoxystrobin as a ligand was developed. The model was validated with a set of commercially available strobilurins, and it well explains both the resistance mechanism to strobilurins mediated by the mutation G143A and the activity of metyltetraprole against strobilurin-resistant strains. The obtained results shed light on the key recognition determinants of strobilurin-like derivatives in the cytochrome bc1 active site and will guide the further rational design of new fungicides able to overcome resistance caused by G143A mutation in the rice blast pathogen.  相似文献

A noninvasive eye tracking system based on infrared 3-D video-oculographic techniques is proposed for the automatic monitoring of eye position and orientation in external beam radiotherapy of ocular tumors. The presented method can be applied for the real-time estimation of lesion position and tumor-beam misalignments, allowing automatic patient setup and eye movement gated treatments. A prototypal eye tracker was developed and tested on five subjects, achieving gaze estimation errors of 0.5° and eye monitoring frequencies of 125 Hz. The proposed application can potentially improve quality and efficacy of ocular radiotherapy treatments, currently based on invasive, qualitative, and manual control procedures.  相似文献

The case of magnetization reversal in a boundless plate of magnetically homogeneous material is treated in the framework of micromagnetics. It is assumed that the magnetocrystalline symmetry axis and the applied field lie along the normal of the plate surface. The mathematical analysis performed facilitates the determination of the nucleation field in an exact analytical way, leading to the conclusion that nucleation takes place by spin rotation in unison. Some indirect experimental evidences are discussed.  相似文献

Blockade of adenosine receptors can reduce cerebral infarct size in the model of global ischaemia. Using the potent and selective A2A adenosine receptor antagonist, SCH 58261, we assessed whether A2A receptors are involved in the neuronal damage following focal cerebral ischaemia as induced by occluding the left middle cerebral artery. SCH 58261 (0.01 mg/kg either i.p. or i.v.) administered to normotensive rats 10 min after ischaemia markedly reduced cortical infarct volume as measured 24 h later (30% vs controls, p < 0.05). Similar effects were observed when SCH 58261 (0.01 mg/kg, i.p.) was administered to hypertensive rats (28% infarct volume reduction vs controls, p < 0.05). Neuroprotective properties of SCH 58261 administered after ischaemia indicate that blockade of A2A adenosine receptors is a potentially useful biological target for the reduction of brain injury.  相似文献

Since the discovery of the human T-cell leukemia virus-1 (HTLV-1), cellular and animal models have provided invaluable contributions in the knowledge of viral infection, transmission and progression of HTLV-associated diseases. HTLV-1 is the causative agent of the aggressive adult T-cell leukemia/lymphoma and inflammatory diseases such as the HTLV-1 associated myelopathy/tropical spastic paraparesis (HAM/TSP). Cell models contribute to defining the role of HTLV proteins, as well as the mechanisms of cell-to-cell transmission of the virus. Otherwise, selected and engineered animal models are currently applied to recapitulate in vivo the HTLV-1 associated pathogenesis and to verify the effectiveness of viral therapy and host immune response. Here we review the current cell models for studying virus–host interaction, cellular restriction factors and cell pathway deregulation mediated by HTLV products. We recapitulate the most effective animal models applied to investigate the pathogenesis of HTLV-1-associated diseases such as transgenic and humanized mice, rabbit and monkey models. Finally, we summarize the studies on STLV and BLV, two closely related HTLV-1 viruses in animals. The most recent anticancer and HAM/TSP therapies are also discussed in view of the most reliable experimental models that may accelerate the translation from the experimental findings to effective therapies in infected patients.  相似文献

LIDAR (LIght Detection And Ranging) data are a primary data source for digital terrain model (DTM) generation and 3D city models. This paper presents a three-stage framework for a robust automatic classification of raw LIDAR data as buildings, ground and vegetation, followed by a reconstruction of 3D models of the buildings. In the first stage the raw data are filtered and interpolated over a grid. In the second stage, first a double raw data segmentation is performed and then geometric and topological relationships among regions resulting from segmentation are computed and stored in a knowledge base. In the third stage, a rule-based scheme is applied for the classification of the regions. Finally, polyhedral building models are reconstructed by analysing the topology of building outlines, building roof slopes and eaves lines. Results obtained on data sets with different ground point density, gathered over the town of Pavia (Italy) with Toposys and Optech airborne laser scanning systems, are shown to illustrate the effectiveness of the proposed approach.  相似文献
