Acute myocardial infarction (MI) is one of the most common causes of death worldwide. Pituitary adenylate cyclase activating polypeptide (PACAP) is a cardioprotective neuropeptide expressing its receptors in the cardiovascular system. The aim of our study was to examine tissue PACAP-38 in a translational porcine MI model and plasma PACAP-38 levels in patients with ST-segment elevation myocardial infarction (STEMI). Significantly lower PACAP-38 levels were detected in the non-ischemic region of the left ventricle (LV) in MI heart compared to the ischemic region of MI-LV and also to the Sham-operated LV in porcine MI model. In STEMI patients, plasma PACAP-38 level was significantly higher before percutaneous coronary intervention (PCI) compared to controls, and decreased after PCI. Significant negative correlation was found between plasma PACAP-38 and troponin levels. Furthermore, a significant effect was revealed between plasma PACAP-38, hypertension and HbA1c levels. This was the first study showing significant changes in cardiac tissue PACAP levels in a porcine MI model and plasma PACAP levels in STEMI patients. These results suggest that PACAP, due to its cardioprotective effects, may play a regulatory role in MI and could be a potential biomarker or drug target in MI.  相似文献

Functional properties of glyco-protein conjugates of the anionic polysaccharide pectin with whey protein isolate, obtained by dry heat treatment at 60 °C for 14 days, have been investigated in O/W emulsions containing 20% (w/w) soybean oil and 0.4% (w/w) protein both at pH 4.0 and 5.5. Emulsion stabilizing properties of mixtures and conjugates were compared at five protein to pectin weight ratios by determining changes in droplet size distribution and extent of serum separation with time. The results indicated that the dry heat-induced covalent binding of low methoxyl pectin to whey protein, as shown by SDS-PAGE, led to a substantial improvement in the emulsifying behaviour at pH 5.5, which is near the isoelectric pH of the main protein β-lactoglobulin. At pH 4.0, however, a deterioration of the emulsifying properties of whey protein was observed using either mixtures of protein and pectin or conjugates.The observed effects could be explained by protein solubility and electrophoretic mobility measurements. The protein solubility at pH 5.5 was hardly changed using mixtures of protein and low methoxyl pectin or conjugates, whereas at pH 4.0 it was decreased considerably. Electrophoretic mobility measurements at pH 5.5 revealed a much more pronounced negative charge on the emulsion droplets in the case of protein–pectin conjugates, which clearly indicated that conjugated pectin did adsorb at the interface even at pH conditions above the protein's iso-electric point. Hence, the improved emulsifying properties of whey protein isolate at pH 5.5 upon conjugation with low methoxyl pectin may be explained by enhanced electrosteric stabilization.Comparing two different commercial pectin samples, it was clearly shown that the dextrose content during dry heat treatment of protein–pectin mixtures should be as low as possible since protein–sugar conjugates not only resulted in increased brown colour development, but also gave raise to a largely decreased protein solubility which very badly affected the emulsifying properties.  相似文献

In most cell types and heterologous expression systems, the electrogenic sodium-bicarbonate cotransporter NBCe1 operates with a 1Na+–2HCO3 stoichiometry that, given typical transmembrane electrochemical gradients, promotes Na+ and HCO3 influx. However, NBCe1 in the kidney mediates HCO3 efflux (HCO3 reabsorption), a direction that has been predicted to be favored only if NBCe1 operates with a 1:3 stoichiometry. The phosphorylation state of Ser982 in the cytosolic carboxy-terminal domain of NBCe1 has been reported to be a key determinant of the transporter stoichiometry, with non-phosphorylated Ser982 favoring a 1:3 stoichiometry. Conversely, phosphoproteomic data from renal cortical preparations have revealed the presence of NBCe1 peptides including phosphoserine982 (pSer982) and/or pSer985 although it was not known what proportion of NBCe1 molecules were phosphorylated. In the present study, we report the generation, characterization, and application of a novel phosphospecific antibody raised against NBCe1/pSer982 and show that, contrary to expectations, Ser982 is more prevalently phosphorylated in murine kidneys (in which NBCe1 mediates HCO3 efflux) than in murine colons (in which NBCe1 mediates HCO3 influx). Using phosphomimetic mutants of murine NBCe1 expressed in Xenopus oocytes, we found no evidence that the phosphorylation state of Ser982 or Ser985 alone influences the transport stoichiometry or conductance. Furthermore, we found that the phosphorylation of NBCe1/Ser982 is enhanced in murine kidneys following a 24 h induction of metabolic acidosis. We conclude that the phosphorylation status of Ser982 is not a key determinant of NBCe1 stoichiometry but correlates with presumed NBCe1 activity.  相似文献

The effect of heat on the decomposition of two grades of soybean lecithin has been examined up to 600°C using differential scanning calorimetry and thermogravimetry. Studies were carried out in both air and argon and a kinetic analysis of the isothermal rates of reaction was attempted. Three principal decomposition stages were distinguished in argon and four in air for a water-dispersible lecithin typical of the quality of the material used in many industrial applications. Gas chromatography showed that the principal gases evolved from 50 to 600°C in argon from the water-dispersible lecithin in contact with aluminum surfaces were C1-C6 hydrocarbons, carbon dioxide, carbon monoxide, water and traces of hydrogen.  相似文献

The flexible ligand docking problem is divided into two subproblems: pose/conformation search and scoring function. For successful virtual screening the search algorithm must be fast and able to find the optimal binding pose and conformation of the ligand. Statistical analysis of experimental data of bound ligand conformations is presented with conclusions about the sampling requirements for docking algorithms. eHiTS is an exhaustive flexible-docking method that systematically covers the part of the conformational and positional search space that avoids severe steric clashes, producing highly accurate docking poses at a speed practical for virtual high-throughput screening. The customizable scoring function of eHiTS combines novel terms (based on local surface point contact evaluation) with traditional empirical and statistical approaches. Validation results of eHiTS are presented and compared to three other docking software on a set of 91 PDB structures that are common to the validation sets published for the other programs.  相似文献
