Gynecologic problems in older women

Regular gynecologic evaluation in older women is an integral part of medical care, just as it is for women of reproductive age. This should be emphasized since older women often neglect early symptoms of gynecologic diseases, some of which are potentially lethal. With this in mind, the health care provider must be cognizant of not only gynecologic problems that affect all women, but also those disease processes which are either specific to or more prevalent in an older population. This article emphasizes these aspects in caring for the older gynecologic patient.     

Construction, properties and specific fluorescent labeling of a bovine prothrombin mutant engineered with a free C-terminal cysteine

To define the role of phosphatidylserine-induced conformationalchanges in prothrombin activation during blood coagulation,a recombinant bovine prothrombin was constructed, characterizedand shown to have a globally native-like conformation. We introduceda cysteine to replace the penultimate residue (Gly581) of apreviously constructed active site mutant, and expressed thedouble mutant in Chinese hamster ovary cells at the level of0.6 µg/ml of cell culture medium. Specific labeling withfluorescein maleimide was accomplished by limited reductionwith dithiothreitol to free the engineered cysteine while maintainingthe native-like functional properties of the molecule. The averagestoichiometry of labeling was 0.84 probe/protein. The locationof the probe at the C-terminus was confirmed by proteolysisby native thrombin, by Taipan venom, and by carboxypeptidaseY. Both the double mutant and labeled prothrombin could be activatedby snake venoms and the prothrombinase but, as expected, thedouble mutant meizothrombin did not autolyze as does nativemeizothrombin. Thus, for the first time, a native-like but specificallylabeled prothrombin has been constructed. This molecule willbe an essential tool for elucidating the structural role ofmembranes during prothrombin activation. In addition, the methodsdescribed might be usefully applied to labeling of an odd, engineeredcysteine in other disulfide bond-containing proteins.     

Mie scattering from a sonoluminescing air bubble in water

A single bubble of air in water can emit pulses of blue-white light that have durations of less than 50 ps while it is oscillating in an acoustic standing wave. The emission is called sonoluminescence. A knowledge of the bubble diameter throughout the cycle, and in particular near the time of sonoluminescence emission, can provide important information about the phenomenon. A new Mie scattering technique is developed to determine the size of the bubble through its expansion and collapse during the acoustic cycle. The technique does not rely on an independent means of calibration or on accurate measurements of the scattered intensity.     

Cytochemical localization of acetylcholine receptors at the neuromuscular junction by means of horseradish peroxidase-labeled alpha-bungarotoxin

The cytochemical localization of acetylcholine (ACh) receptors at the neuromuscular junction was investigated with a procedure utilizing alpha-bungarotoxin (alpha-BtX) labeled directly with horseradish peroxidase (HRP). Following incubation of tissues in the conjugate and reaction for peroxidase, activity was observed on the junctional folds of the motor endplate. A uniform layer of reaction product approximately 15 nm thick occurred over the apical portions of the junctional folds. Membranes at the bases of the synaptic cleft showed only small amounts of reaction product. Non-junctional regions of the muscle fiber were unreactive. Activity was also observed in the membrane of the axon facing the muscle surface, often including the axolemma overlying the "active zones" of the nerve terminal. Such presynaptic activity was still evident on nerve terminals disjuncted from the synapse by enzymatic treatment prior to incubation in the conjugate. This localization indicates the possible presence of presynaptic ACh receptors within the axolemma. In muscle denervated for 7-12 days, motor endplates were reactive and parajunctional sarcolemma showed slight activity, but most extrajunctional regions contained no obvious accumulations of reaction product. Activity at all sites was prevented by preincubation of tissues in native alpha-BTX prior to incubation in the conjugate and reaction for HRP. This procedure represents a simple and convenient method for the high resolution localization of ACh receptors.     

Hot working behavior of a WE54 magnesium alloy

The plastic deformation and recrystallization behavior of the commercial magnesium alloys WE54 was analyzed using the strain rates 0.01, 0.1, 1, and 5 s^?1 in the temperature range from 400 to 550 °C. The dependence of the flow stress on the temperature and the strain rate was modeled using the Garofalo hyperbolic sine equation. Thereby, the activation energy for plastic deformation of 224 kJ mol^?1 was determined considering the flow stress at a strain of 0.5. The analysis revealed a stress exponent of 3.2. Furthermore, processing maps were generated by plotting the efficiency of power dissipation and the instability parameter considering different instability criteria as a function of the temperature and the strain rate. Depending on these parameters the extent of the recrystallization and the localization of the nucleation varied, significantly. At 400 °C, the recrystallization is very limited and was observed at grain boundaries (GB), shear bands (SB), and twin boundaries (TW). Increasing temperatures result in an increased recrystallized fraction, while lower strain rates promote grain boundary nucleation and reduce the amount of SBN and TW. The prediction of the processing map was verified by large scale extrusion trials, which proof that the evaluation of hot compression data can provide an effective tool to establish viable processing parameters.     

Ground-based measurements of inflight antenna patterns for imagingradar systems

An approach is presented for determining the inflight antenna pattern in the cross-track direction for air- and spaceborne synthetic aperture radar (SAR) systems. In the 1991 Oberpfaffenhofen DC-8/E-SAR calibration campaign, ground-based measurement, equipment comprising 18 precision calibration receivers and nine polarimetric active radar calibrators, all operating in C-band, were tested. These instruments are capable of handling various pulse lengths and repetition frequencies, and they have a very high dynamic range. Together with precise internal clocks, these instruments are suitable for recording the actual radar transmit pulse shape for the later evaluation of the desired inflight antenna pattern. Lining up these devices in the cross-track direction, each receiver yields an azimuth cut of the three-dimensional antenna pattern. The elevation pattern was then obtained by time correlation of these azimuth cuts. Further results concerning pulse shapes, squint angles, and H-V pattern misalignment are presented     

A student design project to improve power quality for a commercialfacility

A recent paper (see ibid., vol.7, no.4, p.1550-8, 1992) reported on the success of industry-sponsored student projects to promote power quality education. One such project, sponsored by BCM Engineers in Plymouth Meeting, PA, was entitled “Power Quality Survey of a Commercial System.” The results of the survey confirmed the suspicion that power line disturbances (i.e., normal-mode and common-mode impulses and “noise”) were responsible for the outages, premature failures and data communication errors of critical microprocessor-based equipment (e.g., terminals, workstations, network file servers, teleconferencing video equipment, etc.). As a follow-up to this project, this paper reports on the results of a student design project to develop a cost-effective implementation plan to mitigate the causes of the BCM power quality problems (i.e., grounding, wiring methods, load configurations, surge protection, and employee practices). Since BCM only leases several floors of the commercial building, it was important that the design be cost-effective and unobtrusive to the other tenants in the building. The design presented is simple and effective, and it conforms to the NEC and accepted industry practice. The students have benefited from this experience by being exposed to real-world constraints on power quality design which cannot be taught in the classroom     

Career Benefits Associated With Mentoring for Proteges: A Meta-Analysis.

Meta-analysis was used to review and synthesize existing empirical research concerning the career benefits associated with mentoring for the protege. Both objective (e.g., compensation) and subjective (e.g., career satisfaction) career outcomes were examined. Comparisons of mentored versus nonmemored groups were included, along with relationships between mentoring provided and outcomes. The findings were generally supportive of the benefits associated with mentoring, but effect sizes associated with objective outcomes were small. There was also some indication that the outcomes studied differed in the magnitude of their relationship with the type of mentoring provided (i.e.. career or psychosocial).
