The decellularized extracellular matrix (ECM) obtained from human and porcine adipose tissue (AT) is currently used to prepare regenerative medicine bio-scaffolds. However, the influence of these natural biomaterials on host immune response is not yet deeply understood. Since macrophages play a key role in the inflammation/healing processes due to their high functional plasticity between M1 and M2 phenotypes, the evaluation of their response to decellularized ECM is mandatory. It is also necessary to analyze the immunocompetence of macrophages after contact with decellularized ECM materials to assess their functional role in a possible infection scenario. In this work, we studied the effect of four decellularized adipose matrices (DAMs) obtained from human and porcine AT by enzymatic or chemical methods on macrophage phenotypes and fungal phagocytosis. First, a thorough biochemical characterization of these biomaterials by quantification of remnant DNA, lipids, and proteins was performed, thus indicating the efficiency and reliability of both methods. The proteomic analysis evidenced that some proteins are differentially preserved depending on both the AT origin and the decellularization method employed. After exposure to the four DAMs, specific markers of M1 proinflammatory and M2 anti-inflammatory macrophages were analyzed. Porcine DAMs favor the M2 phenotype, independently of the decellularization method employed. Finally, a sensitive fungal phagocytosis assay allowed us to relate the macrophage phagocytosis capability with specific proteins differentially preserved in certain DAMs. The results obtained in this study highlight the close relationship between the ECM biochemical composition and the macrophage’s functional role.  相似文献

An analysis of pulsewidth-modulation inverter nonlinearities influencing high-frequency carrier-signal voltage injection for saliency-tracking-based rotor/flux position estimation is presented in this paper. Distortion of the injected carrier voltage caused by the nonlinear behavior of the inverter has been reported to cause errors in the estimated rotor/flux position. Though a number of techniques have been developed to compensate for inverter nonlinearities, they have not been proven to be effective when a high-frequency low-magnitude voltage needs to be generated. Both the origins of the distortion as well as the requirements for compensation methods to be effective when producing such high-frequency voltages will be established in this paper.  相似文献

The analysis and design of current regulators for multiphase AC loads in AC machines is presented using complex vector notation. The use of complex vector notation provides a way of comparing the performance of controller topologies through their complex vector root locus and complex vector frequency-response functions. Transfusion-associated graft-versus-host disease (TA-GVHD), has rarely been reported associated with B-chronic lymphocytic leukaemia (B-CLL). We report a patient diagnosed with B-CLL, previously treated with fludarabine, who developed TA-GVHD after being transfused during surgery for splenectomy. Diagnosis was confirmed by polymerase chain reaction (PCR) detection of donor DNA in the patient, by amplification of Y-chromosome sequence and analysis of minisatellite polymorphisms. B-CLL patients treated with fludarabine appear to be at risk for TA-GVHD and should be regarded as candidates for transfusions with irradiated blood products. This case illustrates that PCR is a rapid technique for the early diagnosis of TA-GVHD.  相似文献

We investigated whether 2-chlorodexoyadenosine could induce apoptosis in B cell chronic lymphocytic leukemia (B-CLL) cells in vitro using clinically achievable drug doses, measuring apoptosis ratio by flow cytometry. B cells were isolated from previously untreated patients and apoptosis was measured in these cells immediately after isolation and following incubation in vitro, without and with 2-chlorodeoxyadenosine at different concentrations, for 24 and 48 h. Distribution of cellular DNA content and quantitative analysis of apoptosis were determined by standard propidium iodide staining and flow cytometry. Spontaneous apoptosis occurred in B-CLL cells incubated in vitro in the absence of drug, but the level of apoptosis was greater in cells treated with 2-chlorodeoxyadenosine after the second day of culture. The present in vitro study of B-CLL cells from previously untreated patients suggests this chemotherapeutic agent activates a program of cell death by apoptosis using a drug dose equivalent to the physiological concentration used in patients in vivo. These data reveal an interesting possibility in the 2-chlorodeoxyadenosine treatment of untreated patients by neoplastic B cell apoptosis induction.  相似文献

Temperature monitoring using the injection of a high-frequency carrier signal voltage is proposed in this paper. The carrier signal voltage is used to estimate the transient impedance of the system, which is a function of the stator transient resistance and, therefore, the windingspsila temperature. The variation of the stator transient impedancepsilas phase angle is used to track the temperature of the stator windings. The method can be implemented in standard inverter-fed drives with no additional hardware, with minimal computational requirements, and with practically no interference with the regular operation of the drive.  相似文献

This paper studies the application of high-frequency voltage excitation-based stator winding diagnostic methods to three-phase ac machines operated from power converters that create the necessary high-frequency excitation as part of their normal operation. This paper focuses on two specific operating modes: 1) machines operated from inverters in the overmodulation region and 2) machines operated from soft-starters during startup. In both cases, high-frequency (in the range of the hundred hertz) voltage components at well-defined frequencies are created. The negative-sequence currents induced from these high-frequency voltages are shown to contain accurate information on the level of asymmetry (fault) in the machine. This information is significantly richer than exists in other modes of operation, i.e., inverters working in the linear modulation region or soft-starters in the steady state, and provides interesting opportunities to complement other diagnostic methods.  相似文献

This paper compares pulsating- and rotating-vector carrier-signal injection for self-sensing (or sensorless) control of permanent-magnet synchronous machines (PMSMs). The main focus of this paper is the evaluation of estimation errors due to the physical nonideal attributes of both the machine and the inverter. Initial-position and magnet-polarity estimations are analyzed as well. Transient response and signal processing for both techniques are discussed for completeness. The theoretical analysis is supported by experimental and simulation evidence obtained using interior PMSMs.   相似文献

In the present work, an improved version of the conventional-droop control for microgrid converter is presented. The modifications added to the control are based on a feed-forward current control that allows the converter to work in several modes, both when it is grid connected or in island. The use of this control represents the main contribution of this paper, permitting the inverter to work as a grid supporting source or ancillary services provider when it works grid connected. In this mode the converter varies the injected active and reactive power with the variation of voltage module and frequency using the same main control loop as when it is working in island mode.  相似文献
