The species of Beauveria bassiana is widely used for the management of agricultural insect pests. In this study, we integrated egfp-double-stranded RNA (dsRNA) to a previously generated egfp-expressing B. bassiana transformant (Bb-egfp#3) using a protoplast integration method. The Bb-egfp#3 protoplast was mixed with the dsRNA under PEG/CaCl2 conditions and liquid-cultured in Sabouraud dextrose broth for 5 days. A control culture followed the same procedure without dsRNA. Bb-egfp#3/egfp-dsRNA cultures showed very low fungal growth (OD630 = 0.2) compared to the control culture, Bb-egfp#3 only (OD630 = 1.1). Screening of possible transformants on Sabouraud dextrose agar revealed a transformant T3, without egfp signal. T3 was confirmed as B. bassiana through sequencing of conserved genes and insect bioassays. Interestingly, the genomic egfp fragment of T3 was disrupted, and the egfp signal was not detected over four subcultures, which was also confirmed by RNA-seq of Bb-egfp#3 and T3. This study provides an interesting observation that protoplast integration with dsRNA could possibly generate significantly reduced gene expression in B. bassiana and it is stable across several generations. 相似文献

Elevated blood pressure (BP) is the most common cause of cardiovascular disease. Salt intake has a strong influence on BP, and plasma sodium (pNa) is increased with progressive increases in salt intake. However, the associations with pNa and BP had been reported inconsistently. We evaluated the association between pNa and BP, and estimated the risks of all-cause-mortality according to pNa levels. On the basis of data collected from health checkups during 1995-2009, 97,009 adult subjects were included. Positive correlations between pNa and systolic BP, diastolic BP, and pulse pressure (PP) were noted in participants with pNa ≥138 mM/L (P<0.001). In participants aged ≥50 yr, SBP, DBP, and PP were positively associated with pNa. In participants with metabolic syndrome components, the differences in SBP and DBP according to pNa were greater (P<0.001). A cumulative incidence of mortality was increased with increasing pNa in women aged ≥50 yr during the median 4.2-yr-follow-up (P<0.001). In women, unadjusted risks for mortality were increased according to sodium levels. After adjustment, pNa ≥145 mM/L was related to mortality. The positive correlation between pNa and BP is stronger in older subjects, women, and subjects with metabolic syndrome components. The incidence and adjusted risks of mortality increase with increasing pNa in women aged ≥50 yr. 相似文献

[Purpose] The aim of this study was to determine the effect of application of a
metatarsal bar on the pressure in the metatarsal bones of the foot using a foot analysis
system (pressure on the forefoot, midfoot, and rearfoot). [Subjects and Methods] Forty
female university students in their twenties were selected for this study, and an
experiment was conducted with them as the subjects, before and after application of a
metatarsal bar. The static foot regions were divided into the forefoot, midfoot, and
rearfoot, and then the maximum, average, and low pressures exerted at each region were
measured, along with the static foot pressure distribution ratio. 1) Static foot pressure:
The tips of both feet were aligned to match the vertical and horizontal lines of the foot
pressure measuring plate. The subjects were told to look toward the front and not to wear
shoes. 2) Distribution ratio: The distribution ratio was measured in four regions (front,
back, left, and right) using the same method as used for static foot pressure measurement.
[Results] The results of this study showed that the maximum, average, and minimum static
pressures in the forefoot were significantly decreased. The minimum static pressure in the
midfoot was significantly increased, and the pressure in the other parts was significantly
decreased. The maximum and average static pressures in the rearfoot were also
significantly decreased. [Conclusion] As reduction of foot pressure with a metatarsal bar
results in lowering of the arch and an increased contact surface, the foot pressure was
dispersed. These results suggest that wearing shoes with a bar that can decrease the foot
pressure is therapeutically helpful for patients with a diabetic foot lesion or rheumatoid
