病原微生物如细菌、病毒、真菌等广泛存在于土壤环境中,其严重威胁着全球公共卫生安全。发展病原微生物快速检测技术,对于减少病原菌扩散传播、防控传染性疾病、维护生物安全具有重要意义。本文对病原微生物检测技术的最新研究进展进行了综述,包括显色培养技术、基质辅助激光解吸电离飞行时间质谱、分子生物学技术、拉曼光谱技术、流式细胞技术和生物传感器等,并对其原理、应用及优缺点进行了全面比较。最后,对病原微生物检测技术的未来发展提出展望,旨在促进土壤环境病原微生物的快速检测和生物风险防控。 相似文献

Background
Feline diabetes mellitus (DM) shares many pathophysiologic features with human type 2 DM. Human genome‐wide association studies have identified genes associated with obesity and DM, including melanocortin 4 receptor (MC4R), which plays an important role in energy balance and appetite regulation.Hypothesis/Objectives
To identify single nucleotide polymorphisms (SNPs) in the feline MC4R gene and to determine whether any SNPs are associated with DM or overweight body condition in cats.Animals
Two‐hundred forty domestic shorthaired (DSH) cats were recruited for the study. Of these, 120 diabetics were selected (60 overweight, 60 lean), along with 120 nondiabetic controls (60 overweight and 60 lean). Males and females were equally represented.Methods
A prospective case‐control study was performed. Genomic DNA was extracted from blood samples and used as template for PCR amplification of the feline MC4R gene. The coding region of the gene was sequenced in 10 cats to identify polymorphisms. Subsequently, genotyping by restriction fragment length polymorphism (RFLP) analysis assessed MC4R:c.92C > T allele and genotype frequencies in each group of cats.Results
No significant differences in MC4R:c.92C>T allele or genotype frequencies were identified between nondiabetic overweight and lean cats. In the overweight diabetic group, 55% were homozygous for the MC4R:c.92C allele, compared to 33% of the lean diabetics and 30% of the nondiabetics. The differences between the overweight diabetic and the nondiabetics were significant (P < .01).Conclusions and Clinical Importance
We identified a polymorphism in the coding sequence of feline MC4R that is associated with DM in overweight DSH cats, similar to the situation in humans. 相似文献75.

Retentions of total n-3 and n-6 essential fatty acids (EFAs) were assessed in Atlantic salmon (Salmo salar L.) parr held at 8 °C and 2 °C until they increased in weight from ca. 19 g to 38 g. Feeds contained sandeel oil or a rapeseed:linseed
oil blend at 21 and 34% dietary fat. EFA retention efficiencies [(g EFA gained g EFA ingested-1) × 100] were estimated by
the 'mass balance method' from measurements of feed intake, changes in biomass for each tank of fish, and fatty acid compositions
of the feeds and fish. The n-3 EFA retentions were higher (overall mean 71%) across feed treatments and temperatures than
the n-6 EFA retentions (overall mean 63%). Retentions of the n-3 fatty acids were higher in the fish given the feeds with
the lower fat content (77% vs. 65%), implying improved retention with reduced n-3 EFA availability. n-3 EFA retention tended
to be higher at 2 °C than at 8 °C, although this was not consistent across feeds. At low temperature there was very high retention
of the n-3 EFAs in feeds containing sandeel oil (80%). Such high retention may represent an adaptation response to low temperature.
Lower n-6 EFA retentions imply that more n-6 fatty acids were metabolized than n-3 EFAs. Feed oil influenced retention of
the n-6 fatty acids, retention being lower for the salmon parr given the feeds containing sandeel oil (56% vs. 71%). This

Our understanding of habitat selection by reef fish larvae has changed dramatically in the last 10 years. Fish larvae have long been considered passive particles at the mercy of ocean currents, tides and weather events. For reef fishes, ecological evidence has shown that passive dispersal alone often cannot explain larval distributions, suggesting active behaviour by reef fish larvae. While behaviourally modified passive transport may be important, recent work demonstrates that some reef fish larvae have the capability to actively swim to settlement habitat. They can orientate to reefs from distances of at least 1 km and can swim at speeds of 13.5 cm s?1 for several days. The question then becomes, what are the cues that fish larvae use to orientate to reefs? Ambient reef sound has been shown to be attractive to reef fish larvae, and current research aims to understand better the nature and use of these acoustic cues. Other potential cues include chemoreception/olfaction, wave cues and visual location of reefs. A better understanding of active habitat selection and the underlying sensory and behavioural mechanisms will improve our knowledge of recruitment processes in reef fish ecology, and may have implications for active management of reef fish populations. 相似文献
