Campylobacter jejuni is a frequently detected food-borne pathogenic bacterium. Clinical cases are mostly sporadic but campylobacteriosis can have serious consequences, such as the development of Guillain-Barré syndrome as well as diarrheal diseases. We examined 265 retail raw chickens from Korean markets for the presence of C. jejuni using the US Food and Drug Administration standard cultural method and multiplex polymerase chain reaction (mPCR). The mPCR-confirmed C. jejuni isolates were subtyped by pulsed field gel electrophoresis (PFGE) and flaA-typing for investigating the genetic diversity of the microorganism in retail raw chickens. Restriction enzymes SmaI and DdeI were used for PFGE and flaA-typing, respectively. Campylobacter spp. were found in 181 samples (68.3%) and C. jejuni in 100 samples (37.74%). For C. jejuni, 73 pulsotypes and 30 flaA types were detected. Antibiotic resistance tests performed by disk diffusion assay indicated that most C. jejuni isolates were resistant to tetracycline, nalidixic acid, and ciprofloxacin, and 87 composite types were revealed by PFGE, flaA-typing, and the antibiotic resistance tests. Our results show that the genetic diversity of C. jejuni isolates is very high and the correlation between genotype and antibiotic resistance was low even though many bacteria showed multi-drug resistance.  相似文献

Tunneling spectra of intermediate-valence semiconductor SmB6 are reported for in-situ break junctions, being able to make nano-scale planar tunnel junctions. The electron tunneling using break junction method is a powerful probe of the intrinsic energy gap. The investigated tunneling conductance dI/dV curves are mostly reproducible and symmetric with respect to the applied voltage. Two kinds of characteristic energy gaps are observed at 2E(d) = 20 mV and 2E(a) = 9 mV, which coincides well with those previously studied by point-contact spectroscopy and the activation energy fitted by our electrical resistivity data. The positions of the gap structures are independent of the zero-bias conductance, implying no additional voltage drop induced by the break junctions. The small anomaly at the activation energy 2E(a) indicates a relatively low density of in-gap states. Furthermore, the results of magnetic properties reveal the ratio of Sm2+:Sm3+ = 3.7:6.3 and the antiferromagnetic nature at high temperature.  相似文献

We present a method to produce stable proton and helium ion beams with energies of 10-100 keV from a 30-year-old Van De Graaff accelerator using an external stabilized DC voltage supply instead of the belt charging system. Requiring no other modifications, this makes an ideal system for ion irradiation with fluences up to 1015 ions/cm2. Such ion energies and fluences are required in the emerging fields such as silicon micromachining using ion irradiation and we give examples of structures created with sizes as small as 200 nm.  相似文献

The application of a single cube beam splitter (SCBS) microscope to micro-optics characterization is presented. The SCBS in the optical path, with a small angle between the optical axis and its central semireflecting layer, not only gives off-axis digital holograms but also provides dual-channel imaging. It is a unique and easy way to perform uniformity inspection across the entire microlens array. Experimental results on physical spherical phase compensation, single lens characterization, dual-channel imaging, and uniformity inspection are provided to demonstrate the unique properties of SCBS microscopy.  相似文献

This work focused on the degradation of toxic organic compounds such as trichloroethylene (TCE) in water, using a combined photocatalysis/microfiltration (MF) system. The performances of the hybrid system were investigated in terms of the removal efficiency of TCE and membrane permeability, in the presence or absence of background species, such as alkalinity and humic acids. The mass balancing of the fate of TCE during photocatalytic reactions was performed in order to evaluate the feasibility of the photocatalytic membrane reactor (PMR). Greater TCE degradation (>60%) was achieved with an increase in the TiO2 dosage (up to 1.5g/L) in PMR, but a substantially large TiO2 dosage brought about a decrease in TCE degradation efficiency. The photocatalytic decomposition of TCE appeared to be more effective in acidic pH conditions than with a neutral or alkaline pH. The addition of alkalinity and humic acid into the feedwater did not have a significant effect on TCE degradation, while humic acids (whose dose was 1mg/L as TOC) in the feedwater played a part in a decline of permeability by 60%. Membrane permeability in the PMR was also affected by tangential velocities. An improvement of 60% in flux was achieved when the tangential velocity increased from 0.19 to 1.45m/s. This is because flow regimes can govern the deposition of TiO2 particles on the membrane surface.  相似文献

Centromere protein B is a highly conserved constitutive protein found at centromeres. Gene knockout studies in mice have unexpectedly identified Cenpb as a candidate gene involved in uterine function. The present study further explores the role of Cenpb in mice by intermating Cenpb-null mice over several generations. Breeding studies and analysis of uterine tissue indicate that Cenpb-null mice lose reproductive fitness over a number of generations due to a significant reduction in endometrial glands. These results suggest that Cenpb may play an important function in the short- and long-term maintenance of uterine integrity.  相似文献

Various fine pitch chip-on-film (COF) packages assembled by (1) anisotropic conductive film (ACF), (2) nonconductive film (NCF), and (3) AuSn metallurgical bonding methods using fine pitch flexible printed circuits (FPCs) with two-metal layers were investigated in terms of electrical characteristics, flip chip joint properties, peel adhesion strength, heat dissipation capability, and reliability. Two-metal layer FPCs and display driver IC (DDI) chips with 35 μm, 25 μm, and 20 μm pitch were prepared. All the COF packages using two-metal layer FPCs assembled by three bonding methods showed stable flip chip joint shapes, stable bump contact resistances below 5 mΩ, good adhesion strength of more than 600 gf/cm, and enhanced heat dissipation capability compared to a conventional COF package using one-metal layer FPCs. A high temperature/humidity test (85 °C/85% RH, 1000 h) and thermal cycling test (T/C test, ?40 °C to + 125 °C, 1000 cycles) were conducted to verify the reliability of the various COF packages using two-metal layer FPCs. All the COF packages showed excellent high temperature/humidity and T/C reliability, however, electrically shorted joints were observed during reliability tests only at the ACF joints with 20 μm pitch. Therefore, for less than 20 μm pitch COF packages, NCF adhesive bonding and AuSn metallurgical bonding methods are recommended, while all the ACF and NCF adhesives bonding and AuSn metallurgical bonding methods can be applied for over 25 μm pitch COF applications. Furthermore, we were also able to demonstrate double-side COF using two-metal layer FPCs.  相似文献

Poly(ethylene terephthalate) (PET)/Clay hybrids were prepared via in situ polymerization, in which the clay was modified with 12-aminododecanoic acid (ADA). Three types of bifunctional compounds were introduced into the system, respectively, as compatibilizers between PET and ADA-Clay. The three compounds contain 1 degree, 2 degree, and 3 degree amino groups, respectively, that could form ionic complex with carboxylic acid of ADA. They also contain an ester group that could react with PET chains through transesterification. The results show that adding the compound with 3 degree amino group can improve clay dispersion significantly, while the ones with 1 degree and 2 degree amino groups are not effective compatibilizers. The possible underlying mechanism is discussed.  相似文献

A fast and ultra-sensitive trace analysis of methyl parathion pesticides in a polydimethylsiloxane (PDMS) microfluidic channel was investigated using confocal surface-enhanced Raman spectroscopy (SERS). A three-dimensional PDMS-based passive micromixer was fabricated for this purpose. This PDMS micromixer showed a high mixing efficiency because a strong chaotic advection was developed by the simultaneous vertical and transverse dispersion of the confluent streams. The confocal SERS signal was measured after methyl parathion pesticides were effectively adsorbed onto silver nanoparticles while flowing along the upper and lower alligator-teeth-shaped PDMS channel. A quantitative analysis of the methyl parathion pesticides was performed based on the measured peak height at 1246 cm-1. Our method has a detection limit of 0.1 ppm. This value satisfies the requirement recommended by the Collaborative International Pesticides Analytical Council (CIPAC) for the determination of methyl parathion in pesticide formulations. This study demonstrates the feasibility of using confocal SERS for the highly sensitive detection of methyl parathion pesticides in a PDMS microfluidic channel.  相似文献

Pleural mesothelial cells (PMCs) play a central role in the progression of pleural fibrosis. As pleural injury progresses to fibrosis, PMCs transition to mesenchymal myofibroblast via mesothelial mesenchymal transition (MesoMT), and produce extracellular matrix (ECM) proteins including collagen and fibronectin (FN1). FN1 plays an important role in ECM maturation and facilitates ECM-myofibroblast interaction, thus facilitating fibrosis. However, the mechanism of FN1 secretion is poorly understood. We report here that myosin 5b (Myo5b) plays a critical role in the transportation and secretion of FN1 from human pleural mesothelial cells (HPMCs). TGF-β significantly increased the expression and secretion of FN1 from HPMCs and facilitates the close association of Myo5B with FN1 and Rab11b. Moreover, Myo5b directly binds to GTP bound Rab11b (Rab11b-GTP) but not GDP bound Rab11b. Myo5b or Rab11b knockdown via siRNA significantly attenuated the secretion of FN1 without changing FN1 expression. TGF-β also induced Rab11b-GTP formation, and Rab11b-GTP but not Rab11b-GDP significantly activated the actin-activated ATPase activity of Myo5B. Live cell imaging revealed that Myo5b- and FN1-containing vesicles continuously moved together in a single direction. These results support that Myo5b and Rab11b play an important role in FN1 transportation and secretion from HPMCs, and consequently may contribute to the development of pleural fibrosis.  相似文献
