Northern pintail duck semen and sperm traits were characterized, and the fertility of cold-stored spermatozoa was investigated using artificial insemination. Excellent quality ejaculates containing high proportions of motile spermatozoa were collected from drakes within 20 s by a massage technique. Semen was collected in Beltsville poultry semen extender, pooled and cold-stored (4 degrees C) for 0, 24, 48 or 72 h. Hens were inseminated with 100 microl twice a week, and eggs were assessed for fertilization and hatch success. Fertilization success was similar (P > 0.05) for semen cold-stored for 0 (51.6%), 24 (51.5%), 48 (41.1%) and 72 h (22.3%; P > 0.05). Similar (P > 0.05) percentages of fertilized eggs hatched to live offspring (73.1, 71.4, 87.0 and 80.0%, respectively). Fresh semen was also equilibrated with 1 or 4% dimethylsulphoxide or glycerol, and cryopreserved at the following rates: (1) approximately 60 degrees C min(-1) (in liquid nitrogen [LN(2)] vapour) for 10 min; (2) 1 degrees C min(-1) to -20 degrees C, LN(2) vapour for 10 min; and (3) 1 degrees C min(-1) to -35 degrees C, all followed by immersion in LN(2). After thawing for 30 s at 37 degrees C or 20 min at 4 degrees C, sperm motility and viability were assessed. The highest numbers of motile spermatozoa were recovered after slow-fast freezing (2) and thawing at 0 degrees C (P < 0.05), but survival was inadequate to allow artificial insemination. Nonetheless, cold storage provides an effective means of short-term storage with no loss of fertility in this waterfowl species.  相似文献

This study examines ultrastructural changes in explants of fetal mouse cerebellum following exposure to castanospermine, an inhibitor of N-linked glycosylation and therefore likely to interfere with cell-cell interactions in the developing nervous system. Explants were grown in castanospermine enriched medium for a period of 10 days and compared with identical explants maintained in control culture medium. Both control and treated explants contained many neurons and numerous synaptic profiles. A detailed analysis of 75 thin sections from each group of explants did however reveal highly significant differences between the two. The castanospermine-treated cells contained smaller numbers of cisternae in each Golgi stack; much larger numbers of secretory vesicles which were also substantially larger in size and four times as many coated vesicles per section as control cells. In addition, at 10 days in culture the first changes in the rough endoplasmic reticulum were observed, with individual profiles of reticulum occasionally associated with large vacuoles. Thus, although cerebellar neurons in culture are able to survive prolonged exposure to castanospermine, there are marked ultrastructural changes in subcellular organelles associated with glycoprotein synthesis, packaging and secretion.  相似文献

Dietary and lifestyle change is a recognised strategy for the management and prevention of socio‐economically important non‐infectious diseases. Dietary fibre is composed largely of non‐starch polysaccharides (NSP), and greater consumption of NSP‐rich foods relieves simple constipation very effectively. Starch (as resistant starch, RS) is also receiving attention for its potential role in promoting colonic function. Resistant starch is that fraction of starch which escapes human small intestinal digestion and enters the large bowel where (together with a variable fraction of NSP) it is fermented by the resident microflora. The resulting short chain fatty acids (SCFA) are taken up by the large bowel and metabolised. SCFA mediate many of the health benefits ascribed to NSP and RS. Starch consumption is low in affluent westernised countries, despite recommendations by health agencies. In Australia, foods enriched in RS as a high amylose maize starch have gained consumer acceptance. However, scope remains for additional products and ingredients and a company, Ascentia Pty Ltd, has been established to develop a novel barley cultivar (Hordeum vulgare var. himalaya 292) for this purpose. This cultivar has a specific gene alteration leading to a loss of starch synthetase lla activity resulting in a grain which is low in starch but disproportionately higher in amylose and also NSP. Animal and human studies have shown that Himalaya 292 is high in RS relative to existing products and has a low glycaemic index and the cultivar offers promise as a vehicle to deliver health benefits to consumers.  相似文献
