构建AtNHX1基因植物表达载体,通过农杆菌介导法将其转入番茄。探讨了外植体类型、农杆菌菌株和不同筛选标记对芽诱导分化的影响。对抗性植株进行PCR检测,获得15株转基因植株。对转基因番茄T1代进行80mmol/LNaHCO胁迫处理,转基因植株的相对生长量高于对照植株,显示AtNHX1基因的导入提高了番茄对碱性盐的耐受性。  相似文献

fw2.2 is one of the few QTLs thus far isolated from plants and the first one known to control fruit size. While it has been established that FW2.2 is a regulator (either directly or indirectly) of cell division, FW2.2 does not share sequence homology to any protein of known function (Frary et al. Science 289:85–88, 2000; Cong et al. Proc Natl Acad Sci USA 99:13606–13611, 2002; Liu et al. Plant Physiol 132:292–299, 2003). Thus, the mechanism by which FW2.2 mediates cell division in developing fruit is currently unknown. In an effort to remedy this situation, a combination of yeast two-hybrid screens, in vitro binding assays and cell bombardment studies were performed. The results provide strong evidence that FW2.2 physically interacts at or near the plasma membrane with the regulatory (beta) subunit of a CKII kinase. CKII kinases are well-studied in both yeast and animals where they form part of cell cycle related signaling pathway. Thus while FW2.2 is a plant-specific protein and regulates cell division in a specialized plant organ (fruit), it appears to participate in a cell-cycle control signal transduction pathway that predates the divergence of single- and multi-cellular organisms. These results thus provide a glimpse into how ancient and conserved regulatory processes can be co-opted in the evolution of novel organs such as fruit.  相似文献

The butenolide, 3-methyl-2H-furo[2, 3-c]pyran-2-one, is an highly active compound isolated from plant-derived smoke. This compound is known to stimulate seed germination in a wide range of plants akin to smoke or aqueous extracts of smoke. The present study attempted to elucidate the role of the butenolide in overcoming detrimental effects of low and high temperatures on tomato seed germination and seedling growth. The germination percentage followed a parabolic curve for temperatures ranging from 10 to 40°C, with 25°C being the optimum for all treatments. Control seeds showed radicle emergence at two extreme temperatures (10 and 40°C) and seedlings failed to develop further, even upon prolonged incubation. By comparison the butenolide-treated seeds grew into phenotypically normal seedlings at these non-optimum temperatures. The smoke–water-treated seeds had an intermediate response as only a fraction of germinated seed developed into normal seedlings. Seedling vigour indices as well as seedling weight were significantly higher (p ≤ 0.05) for butenolide-treated seeds at all temperatures. Furthermore, seedlings developed in the presence of the butenolide had about a 1:1 correspondence between root and shoot length. Butenolide-treated seeds grew better than the control seeds in the temperature shift experiments. A gradual decline in the vigour index values was recorded with an increased duration of incubation at the extreme temperatures. Results of the present study are very important from an horticultural point of view as they indicate the potential use of the butenolide compound in restoring normal seed germination and seedling establishment in tomato below and above optimum temperatures.  相似文献

Four hexokinase (LeHXK1–4) and four fructokinase (LeFRK1–4) genes were identified in tomato plants. Previous GFP fusion studies indicate that the gene product of LeHXK3 is associated with the mitochondria while that of LeHXK4 is located within plastids. In this study we found that the enzyme encoded by the fructokinase gene LeFRK3 is also located within plastids. The presence of LeFrk3 enzyme in plastids raises the question of the origin of fructose in these organelles. The other three FRKs enzymes, LeFrk1&2&4, are located in the cytosol. Unlike LeFrk1&2&4, the two additional HXKs, LeHxk1&2, share a common membrane anchor domain and are associated with the mitochondria similar to LeHxk3. The difference in the locations of the cytoplasmic FRK and HXK isozymes suggests that glucose phosphorylation is confined to defined special intracellular localizations while fructose phosphorylation is less confined.Contribution from the Agriculture Research Organization, The Volcani Center, Bet Dagan, Israel, No. 126/2006 series.  相似文献

Matrix attachment region-binding filament-like protein 1 (MFP1) is a plant-specific long coiled-coil protein that binds double-stranded DNA. While originally identified as a component of the tobacco nuclear matrix, it was subsequently shown that the majority of MFP1 resides in mature chloroplast where it is located at the stroma side of the thylakoids and is able to bind to nucleoids. On the other hand, a 90 kDa MFP1-like protein from onion has been convincingly shown to be an intrinsic component of the onion meristematic nuclear matrix. Here, we have expanded the analysis of the subcellular location of MFP1 by using high-resolution confocal immunofluorescence microscopy and immunogold electron microscopy. Two different antisera raised against MFP1 from two species were used on isolated nuclei and chloroplasts from tomato, tobacco, and Arabidopsis. Our data show that both antibodies detect a signal in both compartments in all three species. An Arabidopsis MFP1 T-DNA insertional mutation abolishes both nuclear and chloroplast signals, indicating that the nuclear and plastidic antigens are derived from the same gene. We therefore suggest that MFP1 is a protein with a dual location, in both nuclei and chloroplasts, consistent with prior findings in onion and the dicot species investigated here.  相似文献

The sensitivity of hydroponically cultivated tomato (Lycopersicon esculentum Mill. cv. Ibiza F1) submitted to nitrite treatments (0.25-10mM KNO(2)) for 7d was studied. Increasing nitrite levels in the culture medium led to several disruptions of tomato plants, reflected by reductions of both dry matter per plant, chlorophyll concentrations and the appearance of chlorosis symptoms at the leaf surface. This behaviour was accompanied by stimulation of nitrite, nitrate and ammonia ion accumulation, mainly in roots and old leaves. Higher proteolytic and gaiacol peroxidase (GPX, EC. 1.11.1.7) activities and malonyldialdehyde content were also noted. Protein content of the different plant organs was decreased by nitrite treatment. These physiological and biochemical parameters were chosen as they are stress indicators. Taken together, our data partly explain the harmful effects of nitrite ions, when excessive in the culture medium.  相似文献

Bacillus licheniformis N1, which has previously exhibited potential as a biological control agent, was investigated to develop a biofungicide to control the gray mold of tomato caused by Botrytis cinerea. Various formulations of B. licheniformis N1 were developed using fermentation cultures of the bacteria in Biji medium, and their ability to control gray mold on tomato plants was evaluated. The results of pot experiments led to the selection of the wettable powder formulation N1E, based on corn starch and olive oil, for evaluation of the disease control activity of this bacterium after both artificial infection of the pathogen and natural disease occurrence under production conditions. In plastic-house artificial infection experiments, a 100-fold diluted N1E treatment was found to be the optimum biofungicide spray formulation. This treatment resulted in the significant reduction of symptom development when N1E was applied before Bo. cinerea infection, but not after the infection. Both artificial infection experiments in a plastic house and natural infection experiments under production conditions revealed that the N1E significantly reduced disease severity on tomato plants and flowers. The disease control value of N1E on tomato plants was 90.5% under production conditions, as compared to the 77% conferred by a chemical fungicide, the mixture of carbendazim and diethofencarb (1:1). The prevention of flower infection by N1E resulted in increased numbers of tomato fruits on each plant. N1E treatment also had growth promotion activity, which showed the increased number of tomato fruits compared to fungicide treatment and non-treated control and the increased fruit size compared the non-treated control under production conditions. This study suggests that the corn starch-based formulation of B. licheniformis developed using liquid fermentation will be an effective tool in the biological control of tomato gray mold.  相似文献

Black shading nets are commonly used to protect agricultural crops from excessive solar radiation and wind, and for water saving. Recent studies have demonstrated that when black nets were replaced by either red, yellow, or pearl nets (ChromatiNets?) of equivalent shading capacity, it increased the fruit yield and improved the quality of bell peppers (Capsicum annuum L.) and tomatoes (Solanum lycopersicum L.) (both Solanaceae). We studied the effects of these colored shading nets on the infestation by aphids [Myzus persicae (Sulzer) and Aphis gossypii Glover (both Hemiptera: Aphididae)] and whiteflies [Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae)], and the incidence of the viral diseases transmitted by these insects, for five consecutive years (2006–2010). These studies were conducted in the semi‐arid Besor region in southern Israel. The plants were grown in ‘walk‐in’ tunnels that were covered by several nets of 35% shading capacity in the range of photosynthetically active radiation. Although the shading nets permit free passage of these pests, the infestation levels of aphids and whiteflies in tunnels covered by either the yellow or pearl nets were consistently 2–3× lower than in tunnels covered by the black or red nets. In accordance with the pest results, when the incidences of Cucumber mosaic virus in pepper grown under the black or red nets ranged between 35 and 89%, they were 2–10× lower under the yellow or pearl nets. Similarly, when the incidences of necrotic Potato virus Y in tomato grown under black or red nets ranged between 42 and 50%, they were 2–3× lower under the yellow or pearl nets. Also, when the incidences of Tomato yellow leaf curl virus in tomato grown under the black or red nets ranged between 15 and 50%, but they were 2–4× lower under the yellow or pearl nets. Putative mechanisms of crop protection achieved by the yellow and pearl nets are discussed.  相似文献
