The murine interleukin 5 receptor (IL-5R) was identified by utilizing an immobilized IL-5 and an immobilized monoclonal antibody against the murein IL-5R (designated H7 mAb). The H7 mAb immunoaffinity-purified materials from the extract of cell-surface radioiodinated T88-M cells (an IL-5-dependent early B cell line) using 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate (CHAPS) were reacted with an immobilized IL-5 matrix. SDS-PAGE of the adsorbed fraction revealed a single band at approximately 60 kDa. The binding of the 60 kDa protein to the immobilized IL-5 matrix was inhibited by the excess IL-5. The CHAPS-extract depleted of the 60 kDa protein by the absorption with H7 mAb did not contain any IL-5 binding proteins. Immunoaffinity procedure provided a final 7400-fold purification, based on an estimation of the content of the 60 kDa protein (approximate purity: 20%) from the silver-stained pattern of SDS-PAGE. Actin was copurified with the 60 kDa protein at an approximate ratio of 1:1, suggesting that the intracytoplasmic domain of the IL-5R may interact with actin. Furthermore, soluble IL-5R (molecular mass: 50 kDa) was purified by the H7 mAb-immunoaffinity chromatography. The purified soluble IL-5R was capable of inhibiting the binding of IL-5 to T88-M cells. Preparative SDS-PAGE followed by electroblotting onto a membrane permitted the determination of the N-terminal sequence of the IL-5R. The determined N-terminal sequence of the IL-5R and the deduced primary sequence from recently isolated cDNA were compared.  相似文献

Summary The vestibulo-ocular reflex (VOR) can be suppressed in darkness if a subject tries to imagine that he looks at a head fixed target. This mental suppression of VOR was used to induce adaptive changes in VOR gam during 3 h of active head oscillations in complete darkness. VOR gain changes were tested by asking the subject to look at a visual target; then passively or actively the head was turned in darkness while the subject fixated the same target. Corrective saccades occurring at the end of the movement when lights were turned on give an elegant measure of VOR gain. Three hours of training induced in 3 subjects a mean of 10.9% and 11.4% decrease of VOR gain for passive and active conditions, respectively. This demonstrates that reflex adaptation can be obtained without external cues, and probably with only an internal reconstruction of target and eye movement.  相似文献

The burgeoning of consumer health informatics and virtual health care can help people improve their health. However, little is known about individuals' reactions to such systems. We conducted an evaluation of the telephone-linked care (TLC) system, a computer-based telecommunications system, that functions as an at home monitor, educator, and counselor for patients with chronic health conditions. Our multimethod assessment of individuals' reactions to using TLC included both quantitative and qualitative methods. Ethnographic in-depth open-ended interviews indicated more subtle and surprising reactions to TLC than the overall positive responses from surveys: individuals formed personal relationships with this technology. This relationship formation suggests that TLC designers may have been successful in their attempts to emulate a conversation with a human being. Our study adds to evidence that technology can serve as a projective device for peoples' values and psychological issues. Both designers and users project values and goals onto computer-based technologies and take on different identities through it. Different groups of users, therefore, may see the same technology differently. People also form relationships with technologies, as they did with TLC. These findings, as well as implications for system design and health outcomes, need to be explored in additional studies.  相似文献

Oseltamivir (Tamiflu) is now being stockpiled by several governments as a first line treatment for an anticipated outbreak of avian influenza caused by H5N1. However, abnormal behaviors and death associated with the use of Tamiflu have developed into a major issue in Japan where Tamiflu is often prescribed for seasonal influenza. Thus, it is critical to determine neuropsychiatric effects of oseltamivir and to establish methods for safe administration. Using juvenile rats and rat hippocampal slices, we investigated whether oseltamivir has adverse effects on the central nervous system. Systemic injection of oseltamivir (50 mg/kg i.p.) produced no change in behavior within 2 h. However, prior injection of oseltamivir significantly altered the duration of loss of lightning reflex following ethanol injection (3.3 g/kg, i.p.). Ethanol injection in the presence of oseltamivir also resulted in enhanced hypothermia. In the CA1 region of hippocampal slices, oseltamivir (100 μM) induced paired-pulse facilitation in population spikes without changes in excitatory postsynaptic potentials. Similarly, 3 μM oseltamivir carboxylate, the active metabolite of oseltamivir, facilitated neuronal firing, though the facilitation did not involve GABAergic disinhibition. Moreover, oseltamivir carboxylate produced further facilitation following administration of 60 mM ethanol. These findings indicate that oseltamivir has effects on the central nervous system, especially when combined with other agents.  相似文献

Summary In the lateral geniculate body (LGB), intra- and quasi-intracellular records were done. With small light stimuli shone into different parts of the receptive field, EPSPs and IPSPs could be elicited. Stimulation of the exact center of an on-center cell produced a pure excitatory response, that of an off-center neurone pure inhibition. This response lasted throughout the stimulus. At light off, inhibition was elicited in on-center cells and excitation in off-center cells. A stimulus in the field periphery produced a mixed response with a small and short excitation followed by large inhibition in on-center cells, and a short inhibition followed by postsynaptic depolarization in off-center cells. At light off, on-center cells showed depolarization after a short polarizing phase, and off-center cells a broad polarization which interrupted the initial small excitation. The latencies of both the excitatory and inhibitory center responses at light on and off characteristic for the two types of neurones, were 20–30 msec shorter than the reversed responses elicited by stimulation of the receptive field surround.The findings are compatible with a model in which each geniculate on-center cell gets its major excitatory input from one optic tract on-center fibre and inhibitory input from several off-center fibres with nearby receptive fields. An off-center LGB-cell receives its main excitation essentially from one offcenter fibre and inhibition from several on-center cells. The responses to moving stimuli also agreed with this model. The presence of recurrent inhibition within the LGB could be confirmed by electrical stimulation. But it could not be decided whether the reciprocal inhibition of on- and off-center cells was due to forward or backward inhibition. The spontaneous activity of on- and off-center cells which were simultaneously recorded with one electrode, showed a mutual inhibition 6–8 msec after one cell had fired. Anatomical data relevant to the model are discussed and some functional implications are suggested.  相似文献

Syncytia or multinucleated giant-cell formation is one of the major cytopathic effects induced by human immunodeficiency virus (HIV) infection. Cell fusion results from the strong interaction of CD4 molecules on the surface of the uninfected T cells and gp120, an external envelope glycoprotein of HIV on the infected T cells. We studied the production of HIV in fusion cells between MOLT-4 and virus-infected MOLT-4/HIV cells and found that HIV production was enhanced up to three- to fivefold, which showed a good correlation with the appearance and extent of syncytia formation. Blocking the fusion by monoclonal antibody against a binding epitope of CD4 molecule to gp120 decreased the HIV production significantly. Enhancement of HIV production was observed by more than five-fold in comparison with chronically infected cells, which were fusion free 20 hr postcocultivation. Electron microscopic observation also showed the presence of abundant HIV particles inside the fused cells and on the outer surface. AZT blocked the HIV augmentation of fused cells in coculture completely. Southern blot analysis revealed that both integrated and unintegrated HIV DNA were highly accumulated in fusion cells, as compared with fusion-free MOLT-4/HIV cells. Among unintegrated DNA, circular and linear DNA were accumulated to a similar degree. Northern blot hybridization showed that rapid enhancement of all three species of HIV-specific RNA containing genomic (9.2 kb) and subgenomic (4.3 and 1.9 kb) RNAs were found 20 hr postinfection in fusion cells. These data suggest that syncytia formation is an extremely active infection process of HIV, by which multiple rounds of reinfection might take place.  相似文献

3D MRI-Based Multicomponent FSI Models for Atherosclerotic Plaques   总被引:1,自引:0,他引:1  
A three-dimensional (3D) MRI-based computational model with multicomponent plaque structure and fluid-structure interactions (FSI) is introduced to perform mechanical analysis for human atherosclerotic plaques and identify critical flow and stress/strain conditions which may be related to plaque rupture. Three-dimensional geometry of a human carotid plaque was reconstructed from 3D MR images and computational mesh was generated using Visualization Toolkit. Both the artery wall and the plaque components were assumed to be hyperelastic, isotropic, incompressible, and homogeneous. The flow was assumed to be laminar, Newtonian, viscous, and incompressible. The fully coupled fluid and structure models were solved by ADINA, a well-tested finite element package. Results from two-dimensional (2D) and 3D models, based on ex vivo MRI and histological images (HI), with different component sizes and plaque cap thickness, under different pressure and axial stretch conditions, were obtained and compared. Our results indicate that large lipid pools and thin plaque caps are associated with both extreme maximum (stretch) and minimum (compression when negative) stress/strain levels. Large cyclic stress/strain variations in the plaque under pulsating pressure were observed which may lead to artery fatigue and possible plaque rupture. Large-scale patient studies are needed to validate the computational findings for possible plaque vulnerability assessment and rupture predictions.  相似文献

O'Goiman's (1979) criticisms of the orienting response (OR) significance hypothesis are not well-founded. The hypothesis is not based solely on electrodermal data—studies reporting significance effects for heart rate and pupillary OR are cited. The hypothesis does not discard novelty but suggests that an interaction between uncertainty and significance triggers OR. Differential reactivity across OR components reflects stimulus- and individual-response factors, and does not require separate “significance registers.” Since the hypothesis specifically maintains that appraisals of stimulus significance affect the OR, not the mere fact of cognitive appraisal per se, no general “cognitive appraisal effect” was ever expected. O'Gorman confuses the difficulty of estimating the subject's spontaneous judgments of significance with the relative ease of manipulating significance in the laboratory. He considers classic OR theory more “objective” only because he fails to look closely enough at the complex judgmental processes involved in stimulus intake, model building, and matching functions. O'Gorman's contentions of different initial- and test-ORs, and of a simple additive relationship between significance and OR, lack supportive data and are challenged by available evidence. Literature is cited suggesting that: a) scanning is continuously biased toward the detection of significant stimuli; b) detection of a sgnificant stimulus feature triggers increased information scanning, increasing the possibility that previously unrecognized uncertainty will be detected, and c) lowers the criterion level of the OR “threshold,” increasing the likelihood of OR if uncertainty is detected. Evidence is cited suggesting that the significance-OR, and perhaps attention in general, is mediated by neocortical-limbic interaction, the limbic' “motivational” evaluator of stimulus input steering and switching neocortical “intellectual” analyzers of stimulus pattern.  相似文献
