目的利用生物大分子表面加强的激光解析/电离时间飞行质谱技术(Seldi-Tof-MS)检测大鼠抗Thy-1模型在系膜增殖期血清中的差异多肽,寻找与系膜增殖相关的生物标志物。方法12只200g雄性Wistar大鼠,分为对照组和模型组,对照组尾静脉注射0.9%氯化钠注射液,模型组尾静脉注射抗Thy-1抗体,7d后处死。过碘酸-希夫(PAS)染色检测病理变化,Seldi-Tof-MS检测血清中差异多肽。结果模型组在第7天出现明显的系膜细胞增殖和基质积聚,与对照组相比,模型组有两个多态峰发生变化(P<0.01),其中质荷比为2746ku的多肽在模型组高表达,7399ku在模型组低表达。结论利用Seldi-Tof-MS在抗Thy-1模型血清中成功发现了两个差异多肽,他们可能是系膜增殖相关生物标志物。 相似文献

Advances in spectroscopy now enable researchers to obtain information about chemical and physical components in food or biological
materials at the molecular level. Various spectroscopic techniques (e.g., atomic absorption spectroscopy, Raman and Fourier-transform
infrared spectroscopy, near infrared spectroscopy, nuclear magnetic resonance spectroscopy, mass spectroscopy, X-ray fluorescence
spectroscopy, ultra-violet spectroscopy) have been used to study structure-function relationships in foods (both liquid and
solid) to improve overall food quality, safety and sensory characteristics; to investigate fungal infections in plant materials
(e.g., fruits, seeds); or to study mobility of different chemical components in food materials. Processing, analyzing, and
displaying these data can often be difficult, time-consuming, and problem-specific. Chemometrics is well established for calibrating
the spectral data to predict concentrations of constituents of interest. Similarly, proteomics deals with the structure-function
relationship of proteins. Since most of the food processing industries are becoming increasingly automated, there is a need
to understand how the spectroscopic data can be used for automation. In this paper, we have provided basic working principles
of the above mentioned spectroscopic techniques, examples of the use of spectral data in food processing, methods of analysis
of spectral data and their integration in the automation process.

The coagulation of soymilk proteins induced by magnesium chloride at 30 °C was investigated using the SDS-PAGE and two-dimensional electrophoresis. Approximately 88.4% of the soymilk proteins were coagulated into the soymilk pellet fraction (SPF) by 5 mM magnesium chloride, and the total protein in the soymilk supernatant fraction (SSF) decreased from 7.80 ± 0.12 mg mL−1 to 0.90 ± 0.18 mg mL−1. SDS-PAGE showed that the total intensities of the protein bands corresponding to the 7S α′ subunit, the 7S α subunit, the 7S β subunit and the 11S A3 subunit were decreased to 10.5 ± 0.3%, 11.8 ± 1.1%, 6.6 ± 0.2% and 15.1 ± 2.2%, respectively. Two-dimensional electrophoresis indicated that most of the soymilk proteins in the SSF, including the 7S subunits, 11S subunits, β-amylase, sucrose binding protein 2, Bd 30K, lectin and trypsin inhibitor A, were coagulated into the SPF by 5 mM magnesium chloride. 相似文献

采用辐射生存率测定、双向电泳和MALDI-TOF/TOF质谱分析方法,对芽胞杆菌T61的γ-射线辐射抗性和辐射后的蛋白质组学进行分析。结果表明,T61的D10值为4 kGy,远高于辐射敏感的大肠杆菌。在10 kGy γ-射线辐照后,34个蛋白质出现显著变化,包括多种抗氧化酶类、核苷酸合成代谢酶类、能量代谢酶类、转录相关因子、蛋白质合成与更新相关因子。其中,一些抗氧化防御系统重要成员上调表达,暗示出抗氧化保护在芽胞杆菌T61的辐射抗性中发挥重要作用。 相似文献

For the analysis of longitudinal data with nonignorable and nonmonotone missing responses, a full likelihood method often requires intensive computation, especially when there are many follow-up times. The authors propose and explore a Monte Carlo method, based on importance sampling, for approximating the maximum likelihood estimators. The finite-sample properties of the proposed estimators are studied using simulations. An application of the proposed method is also provided using longitudinal data on peptide intensities obtained from a proteomics experiment of trauma patients. 相似文献
