目的 选择固相金属亲和层析柱的金属离子,以优化分离纯化重组戊型肝炎病毒包涵体的条件。方法 在同一试验条件下用4种金属离子柱分离纯化目的蛋白。结果 包涵体经不同离子柱层析时目的蛋白的收获率、纯度、洗脱条件各异。结论 镍离子柱纯化效果最佳。  相似文献

目的 合成胸腺素α1(Tα1)基因并在原核细胞中表达。方法 用半酶半化学合成并克隆Tα1基因 ,将该基因插入表达质粒pGEX 4T 1并在大肠杆菌进行表达 ,对表达的融合蛋白GST Tα1利用亲和层析法纯化 ,再经Thrombin酶切后 ,获得重组Tα1进行生物学活性检测。结果 经测序证实合成的Tα1序列与设计的一致。构建的重组表达载体pGEX 4T 1 Tα1在大肠杆菌中得到高效表达。E玫瑰花结形成试验证明重组Tα1具有生物学活性。结论 合成的Tα1基因在大肠杆菌中表达出具有生物活性的蛋白。  相似文献

目的 克隆幽门螺杆菌(Helicobacter pylori,HP)过氧化氢酶(katA)基因并在大肠杆菌中进行表达。方法 采用PCR扩增幽门螺杆菌katA全长基因,将其克隆入pET-11c载体中,经测序证实后,在大肠杆菌中进行表达,产物用Western blot检测其抗原性并进行N末端氨基酸的测序。结果 幽门螺杆菌katA基因全长1 518bp,编码氨基酸505个。在BL21(DE3)中的表达量约占细菌总蛋白的24.9%,表达产物经SDS-PAGE显示其相对分子质量与软件预测结果 58 000相符,N末端5个氨基酸测序结果与Hp中天然的katA完全一致,经Western blot检测可被 Hp全菌抗血清识别。结论 katA能在大肠杆菌中进行高效表达,具有良好的免疫反应性,可望为研究 Hp的致病机理、实验诊断及亚单位疫苗等提供充足的katA原材料。  相似文献

The performance of adhesively-bonded joints under monotonic and cyclic-fatigue loading has been investigated using a fracture-mechanics approach. The joints consisted of an epoxy film adhesive which was employed to bond aluminium-alloy substrates. The effects of undertaking cyclic-fatigue tests in (a) a ‘dry’ environment of 55% relative humidity at 23°C, and (b) a ‘wet’ environment of immersion in distilled water at 28°C were investigated. In particular, the influence of employing different surface pretreatments for the aluminium-alloy substrates was examined. In addition, single-lap joints were tested under cyclic fatigue loading in the two test environments, and a back-face strain technique has been used which revealed that crack propagation, rather than crack initiation, occupied the dominant proportion of the fatigue lifetime of the single-lap joints. In Part II, the data obtained in the present Part I paper will be employed to predict theoretically the lifetime of the adhesively-bonded single-lap joint specimens.  相似文献

Effects of microwave heating on the oxidative stability ofd-tocopherols were studied in relation to the production of fatty acids in oils. During microwave heating, the stability of tocopherols decreased in the orderδ>β>γ>α. This order did not depend on the types of ethyl esters of fatty acids or oils present. But, the shorter the chainlength and the lower the degree of unsaturation of the fatty acid ethyl esters, the greater was the reduction in amount of individual tocopherols. A similar tendency was observed when tocopherol-stripped vegetable oils, with equimolar mixtures of tocopherols added, were treated under the same conditions. The reduction in tocopherols became greater with increasing levels of free fatty acids.  相似文献

This paper describes the design and application of the Atmospheric Evaluation and Research Integrated model for Spain (AERIS). Currently, AERIS can provide concentration profiles of NO2, O3, SO2, NH3, PM, as a response to emission variations of relevant sectors in Spain. Results are calculated using transfer matrices based on an air quality modelling system (AQMS) composed by the WRF (meteorology), SMOKE (emissions) and CMAQ (atmospheric-chemical processes) models. The AERIS outputs were statistically tested against the conventional AQMS and observations, revealing a good agreement in both cases. At the moment, integrated assessment in AERIS focuses only on the link between emissions and concentrations. The quantification of deposition, impacts (health, ecosystems) and costs will be introduced in the future. In conclusion, the main asset of AERIS is its accuracy in predicting air quality outcomes for different scenarios through a simple yet robust modelling framework, avoiding complex programming and long computing times.  相似文献

MaleArchips argyrospilus (Walker) were maximally attracted to a 60404200 admixture of (Z)-11-tetradecenyl, (E)-11-tetradecenyl, (Z)-9-tetradecenyl and dodecyl acetates. The identification of (Z)-9-tetradecenyl acetate, in addition to the other 3 components reported previously, is the first report of a 4-component pheromone system in moths.Archips mortuanus Kearfoot, a sibling species, was maximally attracted to a blend of the same components, but in a 90101 mix of the tetradecenyl acetates. Among a number of tortricine moths that co-occur on apples and other hosts in New York, British Columbia, and elsewhere, differences in diel rhythms of attraction and seasonal distribution are insufficient to maintain reproductive isolation. Although the attractant systems of these species often possess components in common, males are maximally attracted to a speciesspecific blend. In tortricine moths attraction specificity appears to be a paramount mechanism for species partitioning.  相似文献

Short syntheses of (Z)-7-dodecen-1-yl acetate, (Z)-7-tetradecen-1-yl acetate, (Z)-9-dodecen-1-yl acetate, and (Z)-9-tetradecen-1-yl acetate from 7-hydroxyheptanal and 9-oxononanoic acid precursors obtained by oxidative cleavage of easily available aleuritic acid are reported. The key step in these syntheses is a stereoselective Wittig reaction between aldehyde and alkyl-phosphonium salt. Wittig-Horner type reaction of 7-hydroxyheptanal and diethyl cyanomethylphosphonate gave the ,-unsaturated nitrile derivative which after protection of the hydroxyl group was reduced to the corresponding aldehyde. Wittig reaction of the latter, followed by acetylation, completed the synthesis of (E,Z)-7,9-dodecadien-1-yl acetate, the sex pheromone of the European grapevine mothLobesia botrana Schiff.  相似文献
