The behaviour of sperm from egg penetration until creation of the zygote, the development of the maternal pronucleus, and the two first cleavage divisions were studied by use of fluorescence microscopy. It was found that 4-12 sperm penetrate the egg membranes prior to oviposition. Contrary to previous reports, we found that only 1-7 sperm move from their initial location just beneath the vitelline membrane and into the cytoplasm, where they develop into paternal pronuclei. At the time of oviposition, the oocyte nucleus was usually at the stage of metaphase I, rather than anaphase I as previously reported. At 26+/-2.5 minutes the meiotic process had entered the stage of metaphase II. The paternal and maternal pronuclei formed at 55+/-2.6 minutes, and they fused at 93+/-7.3 minutes. The mitotic division of the zygote was completed at 119+/-6.5 minutes. 相似文献

With the increasing flow of biological data there is a growing demand for mathematical tools whereby essential aspects of complex causal dynamic models can be captured and detected by simpler mathematical models without sacrificing too much of the realism provided by the original ones. Given the presence of a time scale hierarchy, singular perturbation techniques represent an elegant method for making such minimised mathematical representations. Any reduction of a complex model by singular perturbation methods is a targeted reduction by the fact that one has to pick certain mechanisms, processes or aspects thought to be essential in a given explanatory context. Here we illustrate how such a targeted reduction of a complex model of melanogenesis in mammals recently developed by the authors provides a way to improve the understanding of how the melanogenic system may behave in a switch-like manner between production of the two major types of melanins. The reduced model is shown by numerical means to be in good quantitative agreement with the original model. Furthermore, it is shown how the reduced model discloses hidden robustness features of the full model, and how the making of a reduced model represents an efficient analytical sensitivity analysis. In addition to yielding new insights concerning the melanogenic system, the paper provides an illustration of a protocol that could be followed to make validated simplifications of complex biological models possessing time scale hierarchies. 相似文献

When selecting in a finite population of honeybees there is a conflict between gain in a quantitative trait and increasing homozygosity, and therefore the frequency of inviable diploid drones. The consequences when using different mating, import, and selection strategies on diploid drone frequency and genetic gain, was explored with Monte Carlo computer simulations.Within a closed population breeding structure, mass selection gave the highest genetic gain in the quantitative trait, but also the largest increase in percentage diploid drones and queens with unacceptably-low brood viability. Mass selection combined with truncation selection against queens having more than 15% diploid drones gave a comparable genetic gain and was the best strategy of the ones studied to avoid diploid drones. Within-family selection (one replacement per sib group) gave the least genetic gain, and a frequency of diploid drones comparable to random (no) selection. It was intermediate between mass selection and mass selection combined with viability selection concerning the frequency of diploid drones.Insemination with pooled and homogenized semen originating from all breeder queens (30), as compared to natural mating with 12 randomly-selected drones, had little effect on the genetic gain and on the overall frequency of diploid drones (10 to 15% by generation 20).The effect of opening the closed breeding population for the import of external queens every generation, by exchanging breeder queens of lowest performance with a corresponding number of new queens (5, 10and 15 out of 30), was also investigated. Under mass selection (natural mating as well as artificial insemination) the frequency of diploid drones and the proportion of queens discarded were reduced because of low brood viability. However, artificial insemination was superior to natural mating considering the latter criterion. If the imported queens were at the same genetic level for the quantitative trait under selection as the whole breeding population at that generation, or 10% better, the genetic gain was respectively slightly reduced and approximately maintained. If the imported queens were of inferior quality (equal to the initial population) the import of queens slowed genetic progress considerably. 相似文献

Background
Blastocystis is a highly prevalent anaerobic eukaryotic parasite of humans and animals that is associated with various gastrointestinal and extraintestinal disorders. Epidemiological studies have identified different subtypes but no one subtype has been definitively correlated with disease.Results
Here we report the 18.8 Mb genome sequence of a Blastocystis subtype 7 isolate, which is the smallest stramenopile genome sequenced to date. The genome is highly compact and contains intriguing rearrangements. Comparisons with other available stramenopile genomes (plant pathogenic oomycete and diatom genomes) revealed effector proteins potentially involved in the adaptation to the intestinal environment, which were likely acquired via horizontal gene transfer. Moreover, Blastocystis living in anaerobic conditions harbors mitochondria-like organelles. An incomplete oxidative phosphorylation chain, a partial Krebs cycle, amino acid and fatty acid metabolisms and an iron-sulfur cluster assembly are all predicted to occur in these organelles. Predicted secretory proteins possess putative activities that may alter host physiology, such as proteases, protease-inhibitors, immunophilins and glycosyltransferases. This parasite also possesses the enzymatic machinery to tolerate oxidative bursts resulting from its own metabolism or induced by the host immune system.Conclusions
This study provides insights into the genome architecture of this unusual stramenopile. It also proposes candidate genes with which to study the physiopathology of this parasite and thus may lead to further investigations into Blastocystis-host interactions.27.

Most aspects of molecular biology can be understood in terms of biological design principles. These principles can be loosely defined as qualitative and quantitative features that emerge in evolution and recur more frequently than one would expect by chance alone in biological systems that perform a given type of process or function. Furthermore, such recurrence can be rationalized in terms of the functional advantage that the design provides to the system when compared with possible alternatives. This paper focuses on those design features that can be related to improved functional effectiveness of molecular and regulatory networks. We begin by reviewing assumptions and methods that underlie the study of such principles in molecular networks. We follow by discussing many of the design principles that have been found in genetic, metabolic, and signal transduction circuits. We concentrate mainly on results in the context of Biochemical Systems Theory, although we also briefly discuss other work. We conclude by discussing the importance of these principles for both, understanding the natural evolution of complex networks at the molecular level and for creating artificial biological systems with specific features. 相似文献

In mammals, two carotenoid cleaving oxygenases are known; beta-carotene 15,15′-monooxygenase (BCMO1) and beta-carotene 9′,10′-oxygenase (BCO2). BCMO1 is a key enzyme in vitamin A synthesis by symmetrically cleaving beta-carotene into 2 molecules of all-trans-retinal, while BCO2 is responsible for asymmetric cleavage of a broader range of carotenoids. Here, we show that the Atlantic salmon beta-carotene oxygenase (bco) gene family contains 5 members, three bco2 and two bcmo1 paralogs. Using public sequence databases, multiple bco genes were also found in several additional teleost species. Phylogenetic analysis indicates that bco2a and bco2b originate from the teleost fish specific genome duplication (FSGD or 3R), while the third and more distant paralog, bco2 like, might stem from a prior duplication event in the teleost lineage. The two bcmo1 paralogs (bcmo1 and bcmo1 like) appear to be the result of an ancient duplication event that took place before the divergence of ray-finned (Actinopterygii) and lobe-finned fish (Sarcopterygii), with subsequent nonfunctionalization and loss of one Sarcopterygii paralog. Gene expression analysis of the bcmo1 and bco2 paralogs in Atlantic salmon reveals regulatory divergence with tissue specific expression profiles, suggesting that the beta-carotene oxygenase subtypes have evolved functional divergences. We suggest that teleost fish have evolved and maintained an extended repertoire of beta-carotene oxygenases compared to the investigated Sarcopterygii species, and hypothesize that the main driver behind this functional divergence is the exposure to a diverse set of carotenoids in the aquatic environment. 相似文献

The honey bee originated in tropical Africa and later dispersed to northern Europe. It has been suggested that a higher hemolymph
storage capacity for the glycolipoprotein vitellogenin evolved in temperate regions, and that the trait constitutes an adaptation
to a strongly seasonal environment. We have investigated whether the relative vitellogenin levels of European and African
honey bees are in accordance with this hypothesis. Our data indicate that European workers have a higher set-point concentration
for vitellogenin compared to their African origin. Considered together with available life history information and physiological
data, the results lend support to the view that “winter bees”, a longlived honey bee worker caste that survives winter in
temperate regions, evolved through an increase in the worker bees’ capacity for vitellogenin accumulation.
