Summary To isolate a novel gene that contains an SH2 domain, we devised a rapid and nonradioactive cDNA library screening method using polymerase chain reaction (PCR). For PCR amplification, we designed degenerate oligonucleotide primers from the multialigned DNA sequences of SH2 domains. Parthenogenetic sporophytes were obtained from three strains of Laminaria japonica Areschoug. These sporophytes grew to maturity in the sea, producine spores that all grew into female gametophytes. These female gametophytes gave rise to another generation of parthenogenetic sporophytes during the next year, so that by the year 1990 parthenogenetic sporophytes had been cultivated for 12, 9, and 7 generations, respectively, for the three strains. When female gametophytes from parthenogenetic sporophytes were combined with normal male gametophytes, normal sporophytes that reproduced and gave rise to both female and male gametophytes were obtained. The parthenogenetic sporophytes were shorter and narrower than the normal sporophytes of the same strain. Chromosome counts on mature sporophytes showed that normal sporophytes (from fertilized eggs) were diploid (2n = approximately 40) and that the spores they produced were haploid (n = approximately 20), while nuclei from both somatic and sporangial cells in parthenogenetic sporophytes were haploid. All gametophytes were haploid. Young sporophytes derived from cultures with both female and male gametophytes were diploid, while young, sporophytes obtained from female gametophytes from parthenogenetic sporophytes had haploid, diploid, or polyploidy chromosome numbers. Polyploidy was associated with abnormal cell shapes. The presence of haploid parthenogenetic sporophytes should be use in breeding kelp strains with useful characteristics, since the sporophyte phenotype is expressed from a haploid genotype which can be more readily selected. 相似文献

黄河干流流量季节性变化和小浪底“调水调沙”形成了黄河河南段特殊的游荡型滩涂生态系统,为揭示游荡型黄河滩涂(嫩滩、二滩、高滩和大堤)群落组成和种间关系及群落稳定性规律,于2015年9月至2018年11月,采用典型样方法在郑州黄河湿地自然保护区游荡型黄河滩涂开展群落学调查,利用149个草本层样方分析群落物种组成、多样性、种间联结性和群落稳定性。结果表明:(1)研究区植物共有90科289属445种,其中被子植物87科286属440种,裸子植物2科2属3种;蕨类植物仅见1属2种。(2)不同滩涂生境优势种具有显著差异,且总体狗牙根(Cynodon dactylon)、芦苇(Phragmites australis)和小蓬草(Erigeron canadensis)为优势种,国家二级保护植物野大豆(Glycine soja)呈斑块聚集分布,且为嫩滩和二滩亚优势种。(3)大堤物种多样性各指标取值最大,嫩滩次之,二滩和高滩较小。(4)χ2检验、Jaccard指数、Pearson相关检验和Spearman秩相关检验表明,优势种种对种间联结性不强,各物种间相对独立。(5)二滩总体联结... 相似文献

与缓慢发展的干旱过程不同,骤旱具有发生速度快,短期内可致害的特点。目前,关于作物骤旱致害的临界阈值及其调控机制尚不清楚。以春小麦为供试作物,通过桶栽试验,模拟研究骤旱过程中小麦受旱致害的过程特征及其控制因素。结果发现,发生骤旱时土壤含水量下降呈先快后慢的变化趋势,叶片水分和叶水势则呈先慢后快的指数变化趋势。叶片光合生理指标对土壤水分的下降存在明显的阈值响应,且不同生理指标的阈值并不完全相同,其中净光合速率与表征叶片光合能力的指标(最大羧化速率)对土壤有效含水量的响应阈值为0.4,气孔导度和蒸腾速率对土壤有效含水量的响应阈值分别为0.5和0.4。而小麦光合生理指标对叶片水分和叶水势的阈值响应并不明显。同时依据各生理指标相关和通径分析结果得出,骤旱发生时引起小麦叶片净光合速率快速降低的主导因子为非气孔因素,而并不是以往作物受旱研究中的气孔因素。本研究结果有望丰富干旱影响认知,并可为科学应对干旱提供依据。 相似文献

In the era of big data, univariate models have widely been used as a workhorse tool for quickly producing marginal estimators; and this is true even when in a high-dimensional dense setting, in which many features are “true,” but weak signals. Genome-wide association studies (GWAS) epitomize this type of setting. Although the GWAS marginal estimator is popular, it has long been criticized for ignoring the correlation structure of genetic variants (i.e., the linkage disequilibrium [LD] pattern). In this paper, we study the effects of LD pattern on the GWAS marginal estimator and investigate whether or not additionally accounting for the LD can improve the prediction accuracy of complex traits. We consider a general high-dimensional dense setting for GWAS and study a class of ridge-type estimators, including the popular marginal estimator and the best linear unbiased prediction (BLUP) estimator as two special cases. We show that the performance of GWAS marginal estimator depends on the LD pattern through the first three moments of its eigenvalue distribution. Furthermore, we uncover that the relative performance of GWAS marginal and BLUP estimators highly depends on the ratio of GWAS sample size over the number of genetic variants. Particularly, our finding reveals that the marginal estimator can easily become near-optimal within this class when the sample size is relatively small, even though it ignores the LD pattern. On the other hand, BLUP estimator has substantially better performance than the marginal estimator as the sample size increases toward the number of genetic variants, which is typically in millions. Therefore, adjusting for the LD (such as in the BLUP) is most needed when GWAS sample size is large. We illustrate the importance of our results by using the simulated data and real GWAS. 相似文献

【背景】水中的重金属污染是一个严峻的环境问题,严重危害人体健康,利用微生物吸附剂修复重金属污染的水体是一种高效环保的方法。Sphingopyxis能够去除重金属,但是其去除水体中镉的研究很少,且其吸附镉的机理尚不清楚。【目的】以从水体中分离的Sphingopyxis sp. YF1为对象,探究该菌对镉的吸附特性和机制。【方法】分析在不同pH、接触时间及重金属初始浓度条件下YF1活菌和死菌对Cd2+的吸附效果,对其进行动力学模型和等温模型拟合,通过扫描电镜和能谱(scanning electron microscopy and energy dispersive X-ray spectroscopy, SEM-EDS)观察镉在活菌和死菌细胞表面的富集,并通过傅里叶变换红外光谱(Fourier transform infrared spectroscopy, FTIR)和X射线光电子能谱(X-ray photoelectron spectroscopy, XPS)分析确定YF1菌中参与吸附Cd2+的官能团,阐明YF1对镉的吸附机理。【结果】当pH值为3.0-5.0时,随着pH值的升高,活菌与死菌的镉吸附量都随着增加,pH值为5.0-7.0时,活菌与死菌的镉吸附量均无较大变化,吸附主要发生在前10 min,之后吸附速率逐渐降低,活菌和死菌吸附Cd2+的过程更符合准二级动力学模型,表明菌体对镉主要是以化学吸附的方式进行;活菌和死菌等温模型拟合都更符合Langmuir模型,说明YF1对Cd2+的吸附为均相吸附;活菌和死菌对Cd2+的吸附量分别达到36.20 mg/g和62.98 mg/g;吸附后的活菌和死菌的细胞表面均有Cd(II)沉积在菌体表面,活菌和死菌的-OH、C-(O,N)和-NO2等基团参与了镉的吸附。【结论】Sphingopyxis sp. YF1菌具有较强的Cd2+去除能力,该菌株在去除水体Cd2+方面具有良好的应用前景。 相似文献

High-mobility group box1 (HMGB1) induces inflammatory injury, and emerging reports suggest that it is critical for brain ischemia reperfusion. Engeletin, a natural Smilax glabra rhizomilax derivative, is reported to possess anti-inflammatory activity. Herein, we examined the mechanism of engeletin-mediated neuroprotection in rats having transient middle cerebral artery occlusion (tMCAO) against cerebral ischemia reperfusion injury. Male SD rats were induced using a 1.5 h tMCAO, following by reperfusion for 22.5 h. Engeletin (15, 30 or 60 mg/kg) was intravenously administered immediately following 0.5 h of ischemia. Based on our results, engeletin, in a dose-dependent fashion, reduced neurological deficits, infarct size, histopathological alterations, brain edema and inflammatory factors, namely, circulating IL-1β, TNF-α, IL-6 and IFN-γ. Furthermore, engeletin treatment markedly reduced neuronal apoptosis, which, in turn, elevated Bcl-2 protein levels, while suppressing Bax and Cleaved Caspase-3 protein levels. Meanwhile, engeletin significantly reduces overall expressions of HMGB1, TLR4, and NF-κB and attenuated nuclear transfer of nuclear factor kappa B (NF-κB) p65 in ischemic cortical tissue. In conclusion, engeletin strongly prevents focal cerebral ischemia via suppression of the HMGB1/TLR4/NF-κB inflammatory network. 相似文献
