An experimental technique is described to determine contact angles on bacterial layers deposited on cellulose triacetate filters. Measurements with water, water-n-propanol mixtures, and alpha-bromonaphthalene were employed to calculate surface free energies of various oral bacteria. Differences of 30 to 40 erg cm-2 were obtained for four different bacterial species isolated from the human oral cavity, if the concept of dispersion and polar surface free energies is applied. The free energies obtained were used to calculate interfacial free energies of adhesion of these bacteria from saliva onto tooth surfaces. Bacterial adhesion is energetically unfavorable, if the enamel surface free energy is less than 50 erg cm-2.  相似文献

A cDNA complementary to the mRNA of the ADP/ATP carrier from Neurospora crassa was identified among ordered cDNA clones by hybridizing total polyadenylated RNA to pools of 96 cDNA recombinant plasmids and subsequent cell-free translation of hybridization-selected mRNA. Further carrier cDNAs were found by colony filter hybridization at a frequency of 0.2-0.3%. The gene of the carrier was cloned and isolated on a 4.6-kbp EcoRI fragment of total Neurospora DNA, and the start of the mRNA was determined by S1 nuclease mapping. From the nucleotide sequence of the cDNA and the genomic DNA, the primary structure of the gene, of the mRNA and of the ADP/ATP carrier protein could be deduced. The gene occurs in a single copy in the genome and related genes are absent. It contains two short introns, and a pyrimidine-rich promoter region. The mRNA has a 46-bp 5' end and a 219-bp 3' end. There is an open reading frame coding for the 313 amino acid residues of the Neurospora carrier protein. The amino acid sequence is homologous in 148 positions with the established primary structure of the beef heart carrier.  相似文献

At present there is karyological information on ca 10% of the species and ca 30% of the genera of the Apocynaceae. Basic numbers of x = 6, 8, 9, 10, 11, 12, 16, 18, 20, 21 and 23 have been assessed. Of these x = 11 is primitive, occurring in ca 60% of the genera. Those of x = 6, 8, 9 and 10 have evolved by reduction, and x = 12 by increase from x = 11. In the subtribe Secondatiinae however, x = 12 is most likely the result of doubling x = 6. The numbers x = 16, 18 and 20 are likewise doubles of x = 8, 9 and 10 respectively. Those of x = 21, 23, and in one case, x = 20 are probably aneuploid products of doubles of x = 11. The two larger subfamilies, Plumerioideae and Apocynoideae have the basic numbers x = 8, 9, 10 and 11 in common and are not separable on the basis of chromosomal evidence. The third small subfamily Cerberoideae is more homogeneous according to basic number, i.e. x = 10 and 20. Most genera are characterized by a constant basic number, but some have two basic numbers; these clearly are cases of infrageneric aneuploidy. Based on records in the literature two closely related generaApocynum andTrachomitum appear to be characterized by a basic number of x = 8 as well as x = 11. This conflicting situation should be clarified by further karyological research. From the level of subtribe onwards some taxa have one basic number, but others are characterized by two or more numbers. The occurrence of similar basic numbers in different phylads of the family is considered to be the result of similar chromosomal evolution mechanisms. Approximately 22% of the investigated species are polyploid. Intrageneric polyploidy occurs with a frequency of about 12.5% and infraspecific polyploidy with less than 4%. The karyotypes observed are symmetrical: the chromosomes within a karyotype are similar in length with primary constrictions usually in a median position. In the Tabernaemontaneae however, it was observed that the karyotypes comprise one pair of distinctly heterobrachial chromosomes in addition to the metacentric ones. This tribe is also characterized by chromosomes which are relatively long. Most genera of the African continent, which are well known regarding their chromosome number, are characterized by x = 11. Exceptions areStrophantus (x = 9) with a mainly tropical African distribution. Two other genera with derived numbers, i.e.Gonioma with x = 10 andPachypodium with x = 9, occur in southern Africa and Madagascar. The genera with a non-African distribution are less known for their chromosome number. However, the available evidence suggests that evolution of derived numbers has occurred more frequently outside Africa than on this continent.  相似文献

The adhesion of the oral bacterium Streptococcus sanguis CH3 to various polymeric surfaces with surface free energies (gamma s) ranging from 22 to 141 erg cm-2 was investigated. Suspensions containing nine different bacterial concentrations (2.5 X 10(7) to 2.5 X 10(9) cells per ml) were used. After adhesion for 1 h at 21 degrees C and a standardized rinsing procedure, the number of attached bacteria per square centimeter (nb) was determined by scanning electron microscopy. The highest number of bacteria was consistently found on polytetrafluorethylene (gamma s = 22 erg cm-2), and the lowest number was found on glass (gamma s = 141 erg cm-2) at all bacterial concentrations tested. The overall negative correlation between nb and gamma s was weak. However, the slope of the line showing this decrease, calculated from an assumed linear relationship between nb and gamma s, appeared to depend strongly on the bacterial concentration and increased with increasing numbers of bacteria in the suspension. Analysis of the data for each separate polymer showed that the numbers of attached cells on polyvinyl chloride and polypropylene were higher but that those on polycarbonate were lower than would be expected on basis of a linear relationship between nb and gamma s. Desorption experiments were performed by first allowing the bacteria to attach to substrata for 1 h, after which the substrata and attached bacteria were removed to bacterial suspensions containing 10-fold lower bacterial concentrations.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The kinetics of adhesion of Streptococcus sanguis CH3 from suspension to polymers with different surface free energies were studied by using three bacterial concentrations (2.5 X 10(7), 2.5 X 10(8), and 2.5 X 10(9) cells per ml-1). Substratum surface free energies (gamma s) ranged from 18 to 120 erg cm-2. The kinetics of bacterial adhesion to these surfaces showed a typical two-step adhesion process, indicating an equilibrium in both steps. In the initial adhesion step (step 1), low equilibrium numbers of adhering bacteria were counted on substrata with surface free energies lower than 55 erg cm-2. A maximal number adhered on substrata with higher surface free energies. At the lowest bacterial concentration tested, the highest number of bacteria were found on substrata with a surface free energy around 55 erg cm-2. For each substratum, step 2 started after a characteristic time interval tau, being short (30 min) for gamma s less than 50 and long (120 min) for gamma s greater than 50 erg cm-2. The relationship between the substratum surface free energy and the number of bacteria adhering at equilibrium after step 2 was similar to, although less distinct than, that during step 1 with a slight indication of a bioadhesive minimum around gamma s = 35 erg cm-2. The results are indicative of a two-step adhesion model, in which step 1 is controlled by macroscopic substratum properties.  相似文献

Trigeminal (V) nucleus principalis (PrV) is the requisite brainstem nucleus in the whisker-to-barrel cortex model system that is widely used to reveal mechanisms of map formation and information processing. Yet, little is known of the actual PrV circuitry. In the ventral “barrelette” portion of the adult mouse PrV, relationships between V primary afferent terminals, thalamic-projecting PrV neurons, and gamma-aminobutyric acid (GABA)-ergic terminals were analyzed in the electron microscope. Primary afferents, thalamic-projecting cells, and GABAergic terminals were labeled, respectively, by Neurobiotin injections in the V ganglion, horseradish peroxidase injections in the thalamus, and postembedding immunogold histochemistry. Primary afferent terminals (Neurobiotin- and glutamate-immunoreactive) display asymmetric and multiple synapses predominantly upon the distal dendrites and spines of PrV cells that project to the thalamus. Primary afferents also synapse upon GABAergic terminals. GABAergic terminals display symmetric synapses onto primary afferent terminals, the somata and dendrites (distal, mostly) of thalamic-projecting neurons, and GABAergic dendrites. Thus, primary afferent inputs through the PrV are subject to pre- and postsynaptic GABAergic influences. As such, circuitry exists in PrV “barrelettes” for primary afferents to directly activate thalamic-projecting and inhibitory local circuit cells. The latter are synaptically associated with themselves, the primary afferents, and with the thalamic-projecting neurons. Thus, whisker-related primary afferent inputs through PrV projection neurons are pre- and postsynaptically modulated by local circuits.  相似文献

Five Enterococcus italicus strains harbouring tet genes responsible for the tetracycline resistance were subjected to plasmid profile determination studies. For four strains tested the profiles showed between three and six plasmid bands, the size of which ranged between 1.6 and 18.5 kb. Southern hybridization experiments associated tetS and tetK genes with chromosomal DNA in all strains and tetM gene with plasmids of around the same size (18.5 kb) in two of the tested strains. The ability of the new species to transfer tetM gene was studied by transfer experiments with the tetracycline-susceptible recipient strains E. faecalis JH2-2 and OG1RF; mobilization experiments were performed with E. faecalis JH 2-2 harbouring the conjugative plasmid pIP501as helper plasmid. The results obtained show that the new enterococcal species was able to acquire antibiotic resistance by conjugation, but not to transfer its plasmids to other bacteria. Further PCR and hybridization experiments carried out to assess the presence of mobilization sequences also suggest that the tetM plasmid from E. italicus is a non-mobilizable plasmid.  相似文献

MOTIVATION: R/qtl is free and powerful software for mapping and exploring quantitative trait loci (QTL). R/qtl provides a fully comprehensive range of methods for a wide range of experimental cross types. We recently added multiple QTL mapping (MQM) to R/qtl. MQM adds higher statistical power to detect and disentangle the effects of multiple linked and unlinked QTL compared with many other methods. MQM for R/qtl adds many new features including improved handling of missing data, analysis of 10,000 s of molecular traits, permutation for determining significance thresholds for QTL and QTL hot spots, and visualizations for cis-trans and QTL interaction effects. MQM for R/qtl is the first free and open source implementation of MQM that is multi-platform, scalable and suitable for automated procedures and large genetical genomics datasets.
