Five TLRs are thought to play an important role in antiviral immunity, sensing viral products and inducing IFN-alpha/beta and -lambda. Surprisingly, patients with a defect of IRAK-4, a critical kinase downstream from TLRs, are resistant to common viruses. We show here that IFN-alpha/beta and -lambda induction via TLR-7, TLR-8, and TLR-9 was abolished in IRAK-4-deficient blood cells. In contrast, IFN-alpha/beta and -lambda were induced normally by TLR-3 and TLR-4 agonists. Moreover, IFN-beta and -lambda were normally induced by TLR-3 agonists and viruses in IRAK-4-deficient fibroblasts. We further show that IFN-alpha/beta and -lambda production in response to 9 of 11 viruses tested was normal or weakly affected in IRAK-4-deficient blood cells. Thus, IRAK-4-deficient patients may control viral infections by TLR-3- and TLR-4-dependent and/or TLR-independent production of IFNs. The TLR-7-, TLR-8-, and TLR-9-dependent induction of IFN-alpha/beta and -lambda is strictly IRAK-4 dependent and paradoxically redundant for protective immunity to most viruses in humans. 相似文献

Fifty children diagnosed with medulloblastoma completed 188 psychological evaluations using the Wechsler Intelligence Scales for Children (D. Wechsler, 1974, 1991) over a 7-year study period following 35-40 Gy postoperative craniospinal irradiation. Random coefficient models were used to predict the trend in the children's intellectual performance as a function of time since diagnosis, with both patient and treatment variables as parameters of this function. A quadratic model demonstrated a delay prior to decline in performance for older patients, whereas the younger patients showed an immediate loss of performance with a plateau at approximately 6 years postdiagnosis. A steeper decline was found for those with higher baseline performance. Clinicians may use the proposed predictive model to identify those patients who are at risk of significant intellectual decline. 相似文献

采用密度梯度离心和贴壁筛选方法分离纯化了恒河猴骨髓间充质干细胞(mesenchymal stem cells from bone marrow of Rhesus monkey,RhBMSCs);通过表面抗原检测和核型分析对细胞进行了初步鉴定;通过生长曲线的绘制和细胞凋亡的检测探讨了细胞的生长特点。本实验获得的RhBMSCs形态以梭型为主,核型正常,CD29表达率为99.2%,CD34、CD45、HLA-DR表达率均低于3.2%,RhBMSCs表型符合间充质干细胞的特点,且纯度较高,细胞生长旺盛,但倍增时间有随传代数的增加而逐渐延长的趋势。本实验探讨了RhBMSCs的体外分离、扩增培养及鉴定的方法,并对其生长特点进行了初步观察,这不仅有助于进一步了解与之近似的人的骨髓间充质干细胞的相关特性,而且为以猕猴为对象的BMSC定向分化和组织修复等动物体内实验打下了基础。 相似文献

目的:为选择性脊髓血管造影和椎管内显微外科提供解剖学资料。方法:采用尸体标本30例,于节段性动脉内逐一注入红色乳胶,在完整脊髓上解剖观察。结果:每例脊髓胸腰段的后髓动脉数目5~13支,平均8.4支。后髓动脉起始情况分3型10亚型,其中后髓动脉与前髓动脉由同一肋间后动脉发出仅34支(13.5%)。胸下部及腰部后髓动脉呈典型“发卡”样折曲。后髓动脉平均外径0.37±0.12mm。约1/3标本在T_(11)~L_3间有1支后髓动脉较粗(>0.5mm),它是脊髓后下部血供的主要来源。结论:多数后髓动脉单独起始,较大的后髓动脉应引起注意。 相似文献

目的:通过检测病毒血清抗体,探讨相关病毒感染与特发性右室心律失常(IRVA)发生的相关性.方法:病例对照研究分为3组:IRVA组、其他心脏病平行对照组(Heart-Disease-Control)和健康对照组(Healthy-Control),每组30例,性别年龄匹配.接受常规检查后进行血清学检查,随访6~12个月.结果:IRVA组与其他2组的X线心胸比值、超声心脏测值比较,无显著性差异(P>0.05).3组的柯萨奇B组病毒(CVB)血清IgM阳性率组间差异无显著性;而IRVA组的巨细胞病毒(CMV)血清IgM阳性率(73.3%)显著高于其他2组(P<0.01),随访6~12个月后,该组CMV IgM阳性率仍然持续增高(66.7%).相关性分析发现,CVB感染与IRVA发生的联系强度低(P>0.05),而CMV感染与IRVA发生的联系强度高(P<0.001).4种常见的病毒血清抗心肌自身抗体检测中发现IRVA组抗β1受体抗体阳性率(60.0%)显著高于其他2组(P<0.01).结论:IRVA患者血清CMV IgM阳性率高,该抗体的出现与IRVA的发生高度相关;CMV感染引起IRVA的发生可能与免疫机制(抗β1受体抗体介导)有关. 相似文献

The present study aimed to investigate the interaction between the coexistent SP receptor and 5-HT3 receptor in trigeminal ganglion (TG) neurons using whole-cell patch clamp technique. The majority of the neurons examined responded to 5-HT with an inward current (I5-HT) (78.2%, 79/101) that could be blocked by 5-HT3 receptor antagonist, ICS-205,930. The I5-HT was potentiated by preapplication of SP (10(-10) to 10(-8) M) in most 5-HT-sensitive cells(78.5%, 62/79). Coapplication of SP and GR-82334, antagonist of NK1 receptor, had no enhancing effect on I5-HT. The concentration-response curves for 5-HT with and without SP preapplication show that: (1) the threshold 5-HT concentrations with and without SP preapplication are basically the same, while SP preapplication increased the maximal value of I5-HT by 38.0% of its control; (2) the EC50 values of the curves with and without SP pretreatment are very close, i.e. 1.89 x 10(-5) M and 2.08 x 10(-5) M (P > 0.1; n = 9), respectively. Intracellular dialysis of GDP-beta-S, a non-hydrolyzable GDP analog, and GF-109203X, a selective protein kinase C inhibitor, removed the SP potentiation of I5-HT. These results may offer a clue to understanding the mechanism underlying the generation and/or regulation of peripheral pain caused by tissue damage inflammation, etc. 相似文献

二尖瓣环的非平面特性对二尖瓣返流的超声诊断和二尖瓣环成形术的合理设计具有重要意义.基于心脏的实时三维超声图像,我们研究了一种对二尖瓣环三维重建及运动分析的方法.首先通过人机交互方式提取出二尖瓣环的特征点,并根据位置关系对特征点排序,然后利用非均匀有理B样条曲线建立二尖瓣环三维形态模型,并编程实现二尖瓣环的动态显示和运动分析.通过对20组病例分析,初步证明此方法所建模型较准确反映患者的二尖瓣环的运动,能满足二尖瓣环三维可视化和分析研究的需要. 相似文献
