Studies of proteins' interaction in cells by FRET can take benefit from two important photo-physical properties describing fluorescent proteins: fluorescence emission spectrum and fluorescence lifetime. These properties provide specific and complementary information about the tagged proteins and their environment. However, none of them taken individually can completely quantify the involved fluorophore characteristics due to their multiparametric dependency with molecular environment, experimental conditions, and interpretation complexity. A solution to get a better understanding of the biological process implied at the cellular level is to combine the spectral and temporal fluorescence data acquired simultaneously at every cell region under investigation. We present the SLiM-SPRC160, an original temporal/spectral acquisition system for simultaneous lifetime measurements in 16 spectral channels directly attached to the descanned port of a confocal microscope with two-photon excitation. It features improved light throughput, enabling low-level excitation and minimum invasivity in living cells studies. To guarantee a fairly good level of accuracy and reproducibility in the measurements of fluorescence lifetime and spectra on living cells, we propose a rigorous protocol for running experiments with this new equipment that preserves cell viability. The usefulness of SLiM approach for the precise determination of overlapping fluorophores is illustrated with the study of known solutions of rhodamine. Then, we describe reliable FRET experiments in imaging mode realized in living cells using this protocol. We also demonstrate the benefit of localized fluorescence spectrum-lifetime acquisitions for the dynamic study of fluorescent proteins. proteins.  相似文献

The synthesis of new organic hole transporting materials (HTMs) based on 3,6‐disubstituted 9H‐carbazole‐3,6‐diamine,N,N,N′,N′‐tetraphenyl‐9‐(4‐methoxyphenyl) derivatives and their applications in solid state dye sensitizer solar cells (DSSCs) are described. The effect of the methoxy group localized on the para position of the diphenylamine moieties on the thermal, electronic and electrochemical properties and photovoltaic performance is discussed. In solid state DSSCs, utilization of the aforementioned HTMs in combination with the dye D102 (TiO2/D102/HTM/Au) shows a positive influence of the methoxy group on the photovoltaic conversion efficiency compared with unsubstituted diphenylamine grafted groups. A study on the concentration of the HTM is also carried out and shows an optimal concentration around 200 mg mL?1. Without further optimization, the best device gives a power conversion efficiency of 1.75% under AM 1.5 solar irradiation (100 mW cm?2).  相似文献

This paper investigates the manufacturing distortion of curved composite parts manufactured by a new Liquid Composite Molding (LCM) process called Flexible Injection (FI). This technique uses a deformable tool to speed up the fabrication but may generate manufacturing defects when strongly curved shapes are processed. The goal of the study is to evaluate the impact of such heterogeneities on the dimensional stability of the product. Curved components were first manufactured with varying processing conditions to achieve a wide range of layup quality. The shape stability of the samples was then recorded as a function of temperature to measure the thermoelastic component of distortion and experimental results were compared with predictions made by two modeling techniques. Under certain conditions, manufacturing defects can significantly affect the distortion behavior. This suggests that a robust preforming procedure is of primary importance to produce curved parts by Flexible Injection with a high level of repeatability.  相似文献

The optimal design of an energy-intensive process involves a compromise between costs and greenhouse gas emissions, complicated by the interaction between optimal process emissions and supply chain emissions. We propose a method that combines generic abatement cost estimates and the results of existing (LCA) life cycle assessment studies, so that supply chain emissions are properly handled during optimization. This method is illustrated for a (NGCC) natural gas combined cycle power plant model with the following design and procurement options: procurement of natural gas from low-emissions producers, fuel substitution with (SNG) synthetic natural gas from wood, and variable-rate CO2 capture and sequestration from both the NGCC and SNG plants. Using multi-objective optimization, we show two Pareto-optimal sets with and without the proposed LCA method. The latter can then be shown to misestimate CO2 abatement costs by a few percent, penalizing alternate fuels and energy-efficient process configurations and leading to sub-optimal design decisions with potential net losses of the order of $1/MWh. Thus, the proposed LCA method can enhance the economic analysis of emissions abatement technologies and emissions legislation in general.  相似文献

To design a cellular radio network (GSM, UMTS, WiFi …) it is indispensable today to use a radio planning software. In current commercialized tools, radio network planning is based on a numerical evaluation of a set of radio metrics, directly linked to quality of service notion, such as interference, throughput, coverage, capacity … The network configurations produced by such tools are generally faced with the human expert judgment who attaches great importance to the organizational aspects such as cells size and the topology. In other words, all those concepts related to the visual cells shape and geometry. In this article we emphasize the simplicity and the strength by which the geometrical concepts helps to bring out good network properties which are not taken into account by conventional radio criteria. On one hand, well-formed cells lead to reduce the number of handover calls, and to form transition zones between cells allowing proper completion of the handover procedure. On the other hand, cell geometry optimization produce performing network schemes in which the frequencies reuse (frequencies planning in GSM, WiFi) is simplified. We describe in this paper an original and practical modeling of the cellular geometry criteria granting the integration of this concept into the automatic planning process of mobile radio networks. The relevance of this criterion is assessed on three different levels. On a local level, we first establish the correlation between the geometric criteria and the improvement of point-by-point radio quality indicators. We secondly show geometry impact on radio frequency planning and the improving of the handover zones. Finally we analyze the impact of geometric criterion on improving indoor positioning systems.  相似文献

This article introduces a high-level system using belief functions for exchanging and managing imperfect information about events on the road in vehicular ad hoc networks. The main purpose of this application is to provide the most reliable information for the driver from multiple messages received informing the driver about events on the roads. This system and some variants are tested using a MATLAB? simulator. An implementation with Android smartphones using a Bluetooth technology to exchange the messages is also introduced.  相似文献
