The publication of the UKCP09 climate change projections for the United Kingdom provides the opportunity for more rigorous inclusion of climate change uncertainty in water resources planning. We set out how the current approach to incorporating climate change and other uncertainties in water resources planning may be updated to incorporate the UKCP09 projections. In an uncertain future, the frequency with which customers will experience water shortages cannot be predicted for sure, so a water company cannot predict definitely whether it will or will not fulfil its Level of Service commitments. We therefore go on to propose that the probability of failing to meet Level of Service (for given populations of customers) provides an appropriate metric of risk, which conveniently summarises the uncertainties associated with supply and demand, including climate change uncertainties. We sketch out how this risk metric can be calculated based upon simulation modelling of the water resource system.  相似文献

By exploiting a genome-wide collection of bacterial single-gene deletion mutants, we have studied the toxicological pathways of a 60-nm cationic (amino-functionalized) polystyrene nanomaterial (PS-NH(2)) in bacterial cells. The IC(50) of commercially available 60 nm PS-NH(2) was determined to be 158 μg/mL, the IC(5) is 108 μg/mL, and the IC(90) is 190 μg/mL for the parent E. coli strain of the gene deletion library. Over 4000 single nonessential gene deletion mutants of Escherichia coli were screened for the growth phenotype of each strain in the presence and absence of PS-NH(2). This revealed that genes clusters in the lipopolysaccharide biosynthetic pathway, outer membrane transport channels, ubiquinone biosynthetic pathways, flagellar movement, and DNA repair systems are all important to how this organism responds to cationic nanomaterials. These results, coupled with those from confirmatory assays described herein, suggest that the primary mechanisms of toxicity of the 60-nm PS-NH(2) nanomaterial in E. coli are destabilization of the outer membrane and production of reactive oxygen species. The methodology reported herein should prove generally useful for identifying pathways that are involved in how cells respond to a broad range of nanomaterials and for determining the mechanisms of cellular toxicity of different types of nanomaterials.  相似文献

The HUmic-LIke Substances (HULIS) fraction isolated from aerosol samples collected at a rural location of the Pearl River Delta Region (PRD), China, during the harvest season was analyzed by both positive and negative mode electrospray ionization (ESI) coupled with an ultrahigh resolution mass spectrometer (UHRMS). With the remarkable resolving power and mass accuracy of ESI-UHRMS, thousands of elemental formulas were identified. Formulas detected in the positive (ESI+) and the negative (ESI-) mode complement each other due to differences in the ionization mechanism, and the use of both provides a more complete characterization of HULIS. Compounds composed of C, H, and O atoms were preferentially detected in ESI- by deprotonation, implying their acidic properties. Tandem MS and Kendrick Mass Defect analysis implies that carboxyl groups are abundant in the CHO compounds. This feature is similar to those of natural fulvic acids, but relatively smaller molecular weights are observed in the HULIS samples. A greater number of reduced nitrogen organic compounds were observed in the ESI+ compared to ESI-. Compounds with biomass burning origin including alkaloids, amino acids, and their derivatives are their probable constituents. Sulfur-containing species were dominantly detected in ESI-. The presence of sulfate fragments in the MS/MS spectra of these species and their high O/S ratios implies that they are mainly organosulfates. Organosulfates and nitrooxy-organosulfates were often the most intensive peaks in the ESI- spectra. They are believed to be products of reactive uptake of photooxidation products of reactive volatile organic compounds by acidic sulfate particles. The elemental compositions deduced from the UHRMS analysis confirm the conclusion from our previous study that biomass burning and SOA formation are both important sources of HULIS in the PRD region.  相似文献

An increase in on-farm mortality (euthanasia and death) in dairy herds has been reported in several countries in the last decade. This does not only imply possible problems with animal welfare, but it also causes economic losses to the farmer. The objective of this study was to evaluate time trends in on-farm dairy cow mortality in Sweden and identify potential herd-level risk factors. Data were retrieved on all Swedish dairy herds enrolled in the milk recording scheme between 2002 and 2010. Herds with a herd size of <20 cows or a mortality rate (MR) of >40 dead or euthanized cows per 100 cow-years were excluded. Two different models were used: 1 multiple-year analysis, which included 6,898 herds during the period 2002 to 2010 and 1 single-year analysis including 4,252 herds for the year 2010, where other variables that were not present during the entire multiple year study were analyzed. The outcome variable was the number of euthanized and dead cows per year and season. A negative binomial regression model, adjusted for clustering within herd, was applied to both models. Fixed effects in the multiple-year analysis were breed, calving interval, herd size, milk yield, region, season, pasture period, and year. The fixed effects in the single-year analysis were breed, calving interval, conventional versus organic farming, herd size, housing system, milk yield, region, and season. The results demonstrated that MR gradually increased from 5.1 to 6.6 events per 100 cow-years during the study period. Swedish MR are consequently on par with, or even greater than, MR among dairy herds in other comparable countries. Higher mortality was associated with larger herd size, longer calving intervals, and herds that had Swedish Holstein as the predominant breed. Lower mortality was observed in herds with a higher herd average milk yield, during the fall and winter, and in organically managed herds. There were regional differences in mortality. An interaction between herd size and season was found in both models. Also, an interaction between housing system and milk yield was found in the single-year analysis. This first assessment of on-farm mortality in Swedish dairy herds confirmed that the MR has increased over the last few years. The study also identified some herd-level risk factors.  相似文献

BACKGROUND: Fish wastes has been used for many years as an alternative in feeds for aquaculture. In the present study weight gain of juvenile white shrimp Litopenaeus vannamei fed diets including fish waste silage (WS), fish waste silage with soybean meal SBM (WS + S) or fish waste meal (WM) was compared. A conventional acidic silage process was applied to obtain from wastes (skin, heads, bones and viscera) of snapper (Lutjanus spp.), grunt (Haemulon plumieri), and grouper (Epinephelus spp.) an ingredient rich in protein. RESULTS: After 3 days ensilage more than 90% protein was hydrolysed. Waste material processed at pH 3.8 lost about 24% tryptophan. Butylated hydroxytoluene (BHT) prevented lipid oxidation, as shown after 45 days with malonaldehyde production. Shrimp fed WS + S diet gained 0.7 g per week higher than those fed WS and WM diets with 0.3 g per week (P < 0.05). CONCLUSION: WS processed with formic acid under conditions of low pH is beneficial for the white shrimp L. vannamei. It sustained reasonable weight gain combined with soybean meal in practical diets. This article proposes methods for testing the null hypothesis that a number of so‐called long run canonical correlations (LRCCs) are zero. Two test statistics are proposed and their limiting distributions are derived under the null hypothesis. The finite sample properties of the tests are illustrated via a number of simulation studies that reveal the asymptotic theory provides a good guidance to behaviour in moderate or large sized samples. It is shown that the statistics provide a natural way for testing the asymptotic independence of two standardized sums. The usefulness of the tests is illustrated via the following examples: inference about cointegrating vector in a particular cointegration model; inference about break points in a cointegration model; moment estimation; parameter estimation in Generalized Method of Moments estimation.  相似文献

We reconstruct the in vivo spatial distribution of linear and nonlinear elastic parameters in ten patients with benign (five) and malignant (five) tumors. The mechanical behavior of breast tissue is represented by a modified Veronda-Westmann model with one linear and one nonlinear elastic parameter. The spatial distribution of these elastic parameters is determined by solving an inverse problem within the region of interest (ROI). This inverse problem solution requires the knowledge of the displacement fields at small and large strains. The displacement fields are measured using a free-hand ultrasound strain imaging technique wherein, a linear array ultrasound transducer is positioned on the breast and radio frequency echo signals are recorded within the ROI while the tissue is slowly deformed with the transducer. Incremental displacement fields are determined from successive radio-frequency frames by employing cross-correlation techniques. The rectangular regions of interest were subjectively selected to obtain low noise displacement estimates and therefore were variables that ranged from 346 to 849.6 mm2 . It is observed that malignant tumors stiffen at a faster rate than benign tumors and based on this criterion nine out of ten tumors were correctly classified as being either benign or malignant.  相似文献

Polytetrafluoroethylene (PTFE) is widely regarded as an excellent candidate for solid lubrication in vacuum. However, it is often precluded from many practical applications due to its intrinsically high wear rate. Over the past decade, it has been discovered that small loading fractions of alumina nanofillers can increase the wear resistance of PTFE by three to four orders of magnitude. This dramatic increase in wear resistance has in turn prompted numerous tribological studies to examine the robustness of this performance. In this study, the wear and friction behavior of unfilled PTFE and PTFE and alumina nanocomposites were evaluated under a broad range of vacuum environments from 760 to 4 × 10?6 Torr. The nanocomposites of PTFE/alumina showed a dramatic increase in wear of over two orders of magnitude at the highest vacuum conditions. There appears to be an optimal vacuum environment around 1–10 Torr, in which these samples achieved the lowest wear rates of approximately 2.5 × 10?7 mm3/(Nm).  相似文献

Shiga toxin-producing Escherichia coli (STEC) produce toxins that have been associated with several human illnesses. E. coli O157:H7 is the most well-studied STEC and was first associated with consumption of improperly cooked ground beef in 1982. E. coli O157:H7 is not the only foodborne STEC because other STEC serotypes are also associated with human illnesses. The objective of this study was to assess prevalence of STEC in 23 yearling beef (Angus) heifers grazing an irrigated grass pasture in spring (April), summer (July), fall (October), and winter (December) of 1999. A total of 86 fecal samples were rectally collected and were subjected to microbiological testing for the presence of STEC. Nine E. coli isolates from five heifers (one in spring and fall and three in winter) were toxic to Vero cells. Of these isolates, four were E. coli O157:H7, two belonged to the serogroup O6, one O39:NM, one O113:H-, and the final isolate was untypable. The STEC prevalence rate in our herd ranged from 4% (spring) to 15% (winter). Based on detecting both O157:H7 and non-O157:H7 STEC in our heifers, it is clear that screening fecal samples should not be limited to E. coli O157:H7. Identification of STEC-positive cattle prior to slaughter should help in reducing the risk of beef contamination with such foodborne pathogens if pre- and/or postharvest control measures are applied to such animals.  相似文献
