Adsorption and desorption isotherms of quinoa seeds (Chenopodium quinoa Willd.) were measured using the static gravimetric method at three temperatures (20, 40 and 60 °C). Water activity ranged
from 0.118 to 0.937. The moisture sorption behaviour of quinoa was temperature dependent, as indicated by a decrease in equilibrium
moisture content, at all levels of a
w, with increasing temperature. Eight mathematical equations available in the literature were used to model the experimental
data, namely, GAB, BET, Caurie, Henderson, Oswin, Halsey, Smith and Iglesias–Chirife. All the equations showed generally a
good fit; however, the Iglesias–Chirife and Oswin equations were considered the best to predict the experimental data for
both isotherms. Effect of temperature on model parameters was analysed and studied through an Arrhenius-type equation. The
net isosteric heats of desorption and adsorption were determined by applying the Clausius–Clapeyron equation resulting in

The effect of reuterin and high hydrostatic pressure (HHP) processing at 450 MPa for 5 min on the inactivation of Listeria monocytogenes and the characteristics of cold-smoked salmon during 35 days at 4 and 10 °C were investigated. The growth rate of the pathogen was reduced by reuterin addition and a synergistic antimicrobial effect against L. monocytogenes was recorded when the biopreservative was applied in combination with HHP at 450 MPa for 5 min. This combined treatment prevented the pathogen recovery observed with individual treatments and delayed the spoilage of smoked salmon maintaining total viable counts under 3.5 log units during 35 days of storage at 4 °C. All treatments assayed induced changes in lightness (L*) and redness (a*), resulting in a brighter appearance of smoked salmon, whereas no modifications were recorded in shear strength values immediately after treatments. Moreover, reuterin and HHP treatments, individually or in combination, avoided the formation of biogenic amines during the 35 days of storage at 4 and 10 °C. The addition of reuterin in combination with HHP at 450 MPa for 5 min might be applied as a hurdle technology to improve the safety and extend the shelf life of lightly preserved seafood products, such as cold-smoked salmon. 相似文献

Although cysteine desulphydrase activity has been purified and characterized from Saccharomyces cerevisiae, the gene encoding this activity in vivo has never been defined. We show that the full‐length IRC7 gene, encoded by the YFR055W open reading frame, encodes a protein with cysteine desulphydrase activity. Irc7p purified to homogeneity is able to utilize l ‐cysteine as a substrate, producing pyruvate and hydrogen sulphide as products of the reaction. Purified Irc7p also utilized l ‐cystine and some other cysteine conjugates, but not l ‐cystathionine or l ‐methionine, as substrates. We further show that, in vivo, the IRC7 gene is both necessary and sufficient for yeast to grow on l ‐cysteine as a nitrogen source, and that overexpression of the gene results in increased H2S production. Strains overexpressing IRC7 are also hypersensitive to a toxic analogue, S‐ethyl‐l ‐cysteine. While IRC7 has been identified as playing a critical role in converting cysteine conjugates to volatile thiols that are important in wine aroma, its biological role in yeast cells is likely to involve regulation of cysteine and redox homeostasis. The antimicrobial effect against L. monocytogenes of biodegradable films (alginate, zein and polyvinyl alcohol) containing enterocins was investigated. Survival of the pathogen was studied by means of challenge tests performed at 6 degrees C during 8 and 29 days, for air-packed and vacuum-packed sliced cooked ham, respectively. Air packaging was tested with two concentrations of enterocins (200 and 2000 AU/cm2). Control air-packed cooked ham showed an increase of L. monocytogenes from 10(4) to 10(7) CFU/g after 8 days. By contrast, packaging with antimicrobial films effectively slowed down the pathogen's growth, leading to final counts lower than in control lots. Air-packaging with alginate films containing 2000 AU/cm2 of enterocins effectively controlled L. monocytogenes for 8 days. An increase of only 1 log unit was observed in zein and polyvinyl alcohol lots at the same enterocin concentration. Vacuum packaging with films containing enterocins (2000 AU/cm2) also delayed the growth of the pathogen. No increase from inoculated levels was observed during 15 days in antimicrobial alginate films. After 29 days of storage, the lowest counts were obtained in samples packed with zein and alginate films containing enterocins, as well as with zein control films. The most effective treatment for controlling L. monocytogenes during 6 degrees C storage was vacuum-packaging of sliced cooked ham with alginate films containing 2000 AU/cm2 of enterocins. From the results obtained it can concluded that antimicrobial packaging can improve the safety of sliced cooked ham by delaying and reducing the growth of L. monocytogenes. 相似文献

The aim of this research was to study and model the kinetics of the hot-air drying of Aloe vera (Aloe barbadensis Miller) and to evaluate the influence of temperature on the kinetic parameters for the proposed models. A convective dryer was used at 50, 60, 70, 80 and 90 °C with an air flow of 2.0±0.2 m/s. The sorption isotherm of the fresh product was mathematically described by the Guggenheim, Anderson and de Boer (GAB) model, giving as a result monolayer moisture of 0.20 g water/g d.b. The mathematical models evaluated in the kinetic research included five empirical equations (Newton, Henderson-Pabis, Page, modified Page and Fick's diffusional model). The fit quality of the proposed models was evaluated by using the linear regression coefficient (r2), sum square error (SSE), root mean square error (RMSE) and Chi-square statistic (χ2). The diffusivity coefficient increased with the temperature from 5.30 to 17.73×10−10 m2/s, for a range of temperatures between 50 and 90 °C, with an estimated activation energy of 30.37 kJ/mol. When comparing the experimental moisture values with those estimated by the proposed models, the modified Page model provided the best to fit of the data, showing that this equation correctly simulates the Aloe vera drying process and represents an excellent tool for estimating its drying time. 相似文献

In total 7351 meat preparations and fresh processed meat products were analysed from 555 different Serbian meat producers over a 10-year period, 4.5 years before and 5.5 years after mandatory Hazard Analysis and Critical Control Points (HACCP) implementation. From the obtained results, it could be concluded that HACCP has contributed to a better alignment of practices with the legal provisions. The share of non-compliant samples dropped from 18.6% before HACCP to 8.3% after its mandatory implementation. Average sulphite concentrations for all categories of meat preparations and fresh processed meat products decreased by 43%, declining from 33.6 to 19.3 mg kg?1. Typical misuse and frequent abuse of sulphites was independent of a season. Application of HACCP principles in the Serbian meat industry raised awareness about the misuse of sulphites and contributed to a better control, minimising exposure to sulphites. 相似文献

The effects of dietary inclusion of freeze-dried goat and cow milk on the utilization of zinc and selenium, and on the metabolic fate of zinc, were studied in transected (control) and resected rats (resection of 50% of the distal small intestine). Intestinal resection reduced the apparent digestibility coefficient and zinc retention in the cow milk diet, whereas these biological indices were similar in transected (control) and resected rats with standard (without milk) and goat milk diets. The apparent digestibility coefficient and retention of selenium were not affected by intestinal resection in the animals fed with the three diets studied. In transected (control) and resected rats, the apparent digestibility coefficient and retention of zinc and selenium were higher for the goat milk diet than for the other two diets. Zinc deposits in the organs, expressed as microg/g dry weight were in order of decreasing concentrations: femur, testes, sternum, liver, kidney, heart, spleen, longissimus dorsi muscle and brain. Deposits were greatest with the goat milk diet, followed by the standard diet and were lowest for the rats given the cow milk diet, both for transected (control) and resected animals. We conclude that consumption of the goat milk diet produces a greater bioavailability of zinc and selenium and a greater deposit of zinc in key organs, for both the transected (control) and the resected rats, with respect to the standard diet and the cow milk diet. 相似文献

Real-time PCR-based methods have been frequently used to detect and enumerate foodborne pathogens. However, these techniques have a major drawback since they cannot differentiate between DNA from live and dead cells. In this study, we developed a propidium monoazide (PMA)-based PCR method to detect and enumerate viable Salmonella cells in the presence of high number of dead cells (up to 108 CFU/g) in cooked ham. Three different specific PCR targets differing in length (95, 285, and 417 bp) were tested. We found that the inhibition effect was dependent on the PCR amplification product length, and only the longer product achieved suppression of 108 CFU/g of heat-killed cells. SYBR® Green and TaqMan® chemistries were compared to develop a highly efficient PMA-quantitative PCR system targeting the 417-bp fragment. Both chemistries showed similar detection (103 CFU/g) and quantification limits (104 CFU/g), but TaqMan® assay showed higher efficiency (98.6 %) than SYBR® Green assay (92.8 %). PMA-quantitative PCR assay developed is a rapid method for the selective detection and enumeration of viable Salmonella cells with further application in postprocessed meat products and safe shelf-life studies. 相似文献

This work aims to evaluate the influence of olive pomace drying (a solid by-product of the olive oil industry) on both antioxidant potential and drying kinetics. The two main fractions of olive pomace (pits, PI and pulps + peels, P + P) were characterized by image analysis and density measurement. The drying process was analyzed in experiments carried out at different temperatures (from 50 to 150 °C) and mathematically described from the diffusion and Weibull models. The antioxidant potential of the extracts (ethanol–water 80:20 v/v, 22 ± 1 °C, 170 rpm for 24 h) obtained from the dry product was analyzed by measuring the total phenolic content and antioxidant capacity and the main polyphenols were quantified by HPLC–DAD/MS–MS. 相似文献
