The T cell antigen receptor chains are assembled through a rearrangement process that combines variable (V), diversity (D) and joining (J) region genes. Recently, the entire canine T cell receptor γ (TRG) locus was described. It is arranged in 8 cassettes with up to 3 V genes, 2 J genes and 1 C gene each. However, no data is available beyond the level of sequence analysis. The objective of this study was to identify rearranged genes of the canine TRG locus through experimental analysis and to assess gene usage and patterns of rearrangement in a series of canine T cell lymphomas. Rearranged genes were identified through computational analysis of recombination signal sequences (RSSs), a gene's potential to generate a polyclonal smear, and through sequencing of clonal rearrangements in a series of T cell lymphomas. Out of a total of 32 Vγ and Jγ genes, 21 genes were found to rearrange, 8 genes were considered not rearranged and 3 genes were suspected to rearrange but their status could not be determined definitely. Rearrangements of the canine TRG locus were assessed in a group of canine T cell lymphomas as well as 3 neoplastic T cell lines. An average of 4.6 rearrangements per lymphoma was found suggesting that canine T cells routinely rearrange multiple cassettes per allele. The most commonly rearranged Vγ genes belonged to subgroups Vγ2, Vγ3, and Vγ7. Genes in cassettes 2 and 3 preferentially rearranged within their respective cassettes, while Vγ genes in cassette 7 rearranged to a Jγ gene in cassette 8. There was a strong preference for Vγ2 genes to rearrange to a 3' Jγ gene and for Vγ3 and Vγ7 genes to rearrange to a 5' Jγ gene. This rearrangement pattern coincided with the conservation of the spacer sequence between V and J gene subgroups rather than the topologic location of genes. These data show that highly divergent spacer sequences allow for equally efficient recombination and suggest that spacer sequences can mediate compatibility between V and J genes.  相似文献

A stochastic simulation model was used to assess the additional costs related to segregated transport to slaughter. This control measure was assumed to be implemented within a producers' association to decrease Salmonella prevalence in pork. Calculations were based on the additional shipments caused by the separate transport of low- and high-prevalence herds and on the additional transport distance caused by changed routing. The results showed that there is not necessarily a considerable increase in the number of shipments due to herd status separation for transport. The percentage of shipments changed due to segregated transport varied between 43% and 69% depending on the threshold prevalence. The additional costs per slaughtered pig varied between 0.07€/pig and 0.58€/pig under the given assumptions. Costs were governed by the percentage of changed shipments and the additional distance of a changed shipment. Due to the fact that the percentage of changed shipments is related to the distribution of herd prevalence within the producers' association, there is no cost-effective threshold in general. Different producers' associations incur different costs caused by segregated transport to slaughter at the same threshold prevalence. The current study supports producers' associations in evaluating the additional costs of segregated transport for their members.  相似文献

Hypersensitivity dermatitides (HD) are commonly seen in cats, and they are usually caused by environmental, food and/or flea allergens. Affected cats normally present with one of the following clinical reaction patterns: head and neck excoriations, usually symmetrical self-induced alopecia, eosinophilic skin lesions or miliary dermatitis. Importantly, none of these clinical presentations is considered to be pathognomonic for HD skin diseases, and the diagnosis of HD is usually based on the exclusion of other pruritic diseases and on a positive response to therapy. The objectives of this study were to propose sets of criteria for the diagnosis of nonflea-induced HD (NFHD). We recruited 501 cats with pruritus and skin lesions and compared clinical parameters between cats with NFHD (encompassing those with nonflea, nonfood HD and those with food HD), flea HD and other pruritic conditions. Using simulated annealing techniques, we established two sets of proposed criteria for the following two different clinical situations: (i) the diagnosis of NFHD in a population of pruritic cats; and (ii) the diagnosis of NFHD after exclusion of cats with flea HD. These criteria sets were associated with good sensitivity and specificity and may be useful for homogeneity of enrolment in clinical trials and to evaluate the probability of diagnosis of NFHD in clinical practice. Finally, these criteria were not useful to differentiate cats with NFHD from those with food HD.  相似文献

ObjectiveTo investigate the influence of a dexmedetomidine constant rate infusion (CRI) in horses anaesthetized with isoflurane.Study designProspective, randomized, blinded, clinical study.AnimalsForty adult healthy horses (weight mean 491 ± SD 102 kg) undergoing elective surgery.MethodsAfter sedation [dexmedetomidine, 3.5 μg kg?1 intravenously (IV)] and induction IV (midazolam 0.06 mg kg?1, ketamine 2.2 mg kg?1), anaesthesia was maintained with isoflurane in oxygen/air (FiO2 55–60%). Horses were ventilated and dobutamine was administered when hypoventilation [arterial partial pressure of CO2 > 8.00 kPa (60 mmHg)] and hypotension [arterial pressure 70 mmHg] occurred respectively. During anaesthesia, horses were randomly allocated to receive a CRI of dexmedetomidine (1.75 μg kg?1 hour?1) (D) or saline (S). Monitoring included end-tidal isoflurane concentration, cardiopulmonary parameters, and need for dobutamine and additional ketamine. All horses received 0.875 μg kg?1 dexmedetomidine IV for the recovery period. Age and weight of the horses, duration of anaesthesia, additional ketamine and dobutamine, cardiopulmonary data (anova), recovery scores (Wilcoxon Rank Sum Test), duration of recovery (t-test) and attempts to stand (Mann–Whitney test) were compared between groups. Significance was set at p < 0.05.ResultsHeart rate and arterial partial pressure of oxygen were significantly lower in group D compared to group S. An interaction between treatment and time was present for cardiac index, oxygen delivery index and systemic vascular resistance. End-tidal isoflurane concentration and heart rate significantly increased over time. Packed cell volume, systolic, diastolic and mean arterial pressure, arterial oxygen content, stroke volume index and systemic vascular resistance significantly decreased over time. Recovery scores were significantly better in group D, with fewer attempts to stand and significantly longer times to sternal position and first attempt to stand.Conclusions and clinical relevance A dexmedetomidine CRI produced limited cardiopulmonary effects, but significantly improved recovery quality.  相似文献

Stem rust of wheat, caused by Puccinia graminis f. sp. tritici, was under control worldwide for over 30 years by utilizing genetic resistance. The emergence of stem rust in 1998 in eastern Africa in form of race Ug99 and its evolving variants with virulence to many resistance genes were recognized as potential threats to wheat production. In this study we identified genomic regions contributing to putatively durable, adult plant resistance (APR) to wheat stem rust. A recombinant inbred line (RIL) population of 298 lines was previously developed at CIMMYT from a cross between ‘Avocet S’ and ‘Pavon 76’. Pavon 76 has been described to carry APR to stem rust. Avocet S carries the race-specific resistance gene Sr26. A subset of RILs without Sr26 segregated for APR to stem rust race Ug99 when evaluated in Kenya for three years. Single year and joint year analysis by inclusive composite interval mapping using 450 DArT markers identified five quantitative trait loci (QTL) that contributed to the resistance of wheat to stem rust race Ug99. Three of these, including QSr.cim-3B, which probably represents the Sr2 gene, were contributed by Pavon 76 whereas the remaining two QTL were contributed by Avocet S. QSr.cim-3B, or putatively Sr2, on chromosome arm 3BS explained 32 % of the phenotypic variation while the additional QTL in Pavon contributed 24 and 20 %, respectively. Two QTL from Avocet S explained 8 and 6 % of phenotypic variance, respectively. A combination of APR QTL from the two parents resulted in transgressive segregants expressing higher levels of resistance than Pavon 76. Our results indicate that it is possible to accumulate several minor resistance genes each with a small to intermediate effect resulting in a variety that exhibits negligible disease levels even under high stem rust pressure.  相似文献

A 3‐year‐old male New Caledonian giant gecko, or Leach's gecko (Rhacodactylus leachianus) presented with acute lethargy and coelomic distention. Findings from survey radiographs and an upper gastrointestinal tract contrast study were consistent with severe aerophagia, a collapsed left lung, and hyperinflation of the right lung due to suspected bronchial obstruction. The gecko was treated with conservative medical management, but was found dead 5 days after presentation. Necropsy findings showed intussusception of the proximal left lung into the left mainstem bronchus and trachea.  相似文献

Reducing greenhouse gas emissions from arable soil while maintaining productivity is a major challenge for agriculture. Biochar is known to reduce N2O emissions from soil, but the underlying mechanisms are unclear. This study examined the impact of green waste biochar (20 Mg ha?1) and lime (CaCO3; 2 Mg ha?1) application on soil gas transport properties and related changes in these to soil N2O and CO2 emissions measured using automated chambers in a field experiment cropped with maize. In situ soil water content monitoring was combined with laboratory measurements of relative soil gas diffusion coefficient (Dp/D0) at different matric potentials, to determine changes in Dp/D0 over time. Cumulative N2O emissions were similar in the control and lime treatment, but much lower in the biochar treatment. Cumulative CO2 emissions decreased in the order: lime treatment > biochar treatment > control soil. When N2O emissions were not driven by excess N supply shortly after fertilisation, they were associated with Dp/D0 changes, whereby decreases in Dp/D0 corresponded to N2O emissions peaks. No distinct pattern was observed between CO2 emissions and Dp/D0. Cumulative N2O emissions were positively related to number of days with Dp/D0 < 0.02, a critical limit for soil aeration. These results indicate that improved soil gas diffusivity, and hence improved soil aeration, may explain the effect of biochar in reducing N2O emissions. They also suggest that knowledge of Dp/D0 changes may be key to explaining N2O emissions.  相似文献

OBJECTIVE: Food-based dietary guidelines (FBDGs) are globally promoted as an important part of national food and nutrition policies. They are presented within policy as key features of the strategy to educate the public and guide policy-makers and other stakeholders about a healthy diet. This paper examines the implementation of FBDGs in four countries: Chile, Germany, New Zealand and South Africa - diverse countries chosen to explore the realities of the FBDG within policy on public health nutrition. DESIGN: A literature review was carried out, followed by interviews with representatives from the governmental, academic and private sector in all four countries. RESULTS: In all four countries the FBDG is mainly implemented via written/electronic information provided to the public through the health and/or education sector. Data about the impact of FBDGs on policy and consumers' food choice or dietary habits are incomplete; nutrition surveys do not enable assessment of how effective FBDGs are as a factor in dietary or behavioural change. Despite limitations, FBDGs are seen as being valuable by key stakeholders. CONCLUSION: FBDGs are being implemented and there is experience which should be built upon. The policy focus needs to move beyond merely disseminating FBDGs. They should be part of a wider public health nutrition strategy involving multiple sectors and policy levels. Improvements in the implementation of FBDGs are crucial given the present epidemic of chronic, non-communicable diseases.  相似文献

Prolonged summer droughts due to climate change are expected for this century, but little is known about the effects of drying and wetting on biogenic trace‐gas fluxes of forest soils. Here, the response of CO2, N2O, NO, and CH4 fluxes from temperate forest soils towards drying–wetting events has been investigated, using undisturbed soil columns from a Norway spruce forest in the “Fichtelgebirge”, Germany. Two different types of soil columns have been used for this study to quantify the contribution of organic and mineral horizons to the total fluxes: (1) organic horizons (O) and (2) organic and mineral soil horizons (O+M). Three drying–wetting treatments with different rewetting intensities (8, 20, and 50 mm of irrigation d–1) have been compared to a constantly moist control to estimate the influence of rainfall intensity under identical drying conditions and constant temperature (+15°C). Drought significantly reduced CO2, N2O, and NO fluxes in most cycles. Following rewetting, CO2 fluxes quickly recovered back to control level in the O columns but remained significantly reduced in the O+M columns with total CO2 fluxes from the drying–wetting treatment ranging approx. 80% of control fluxes. Fluxes of N2O and NO remained significantly reduced in both O and O+M columns even after rewetting, with cumulative fluxes from drying–wetting treatments ranging between 20% and 90% of the control fluxes, depending on gas and cycle. Fluxes of CH4 were small in all treatments and seem to play no significant role in this soil. No evidence for the release of additional gas fluxes due to drying–wetting was found. The intensity of rewetting had no significant effect on the CO2, N2O, NO, and CH4 fluxes, suggesting that the length of the drought period is more important for the emission of these gases. We can therefore not confirm earlier findings that fluxes of CO2, N2O, and NO during wetting of dry soil exceed the fluxes of constantly moist soil.  相似文献
