Fluorescence spectroscopy has emerged in recent years as an effective way to detect cervical cancer. Investigation of the data preprocessing stage uncovered a need for a robust smoothing to extract the signal from the noise. Various robust smoothing methods for estimating fluorescence emission spectra are compared and data driven methods for the selection of smoothing parameter are suggested. The methods currently implemented in R for smoothing parameter selection proved to be unsatisfactory, and a computationally efficient procedure that approximates robust leave-one-out cross validation is presented.  相似文献

Three salt marshes on a 50-km transect along the north bank of the Westerschelde Estuary were investigated to determine whether salt marshes in the estuary had responded to shipping channel modifications in recent decades. Marsh accretion rates were estimated mainly from 137Cs profiles with further evidence from 241Am because changes in both rate of deposition and nature of the accreting material precluded use of standard 210Pb(excess) dating models. The 137Cs profiles usually show peaks corresponding to atmospheric deposition from the 1963 fallout maximum and sometimes from the Chernobyl accident, although intervening enhanced 137Cs activities derived from the nuclear reprocessing marine discharges of Sellafield and La Hague are clearly discernible. In all three marshes (Ritthem at the mouth of the estuary and Zuidgors and Waarde at 20 and 45 km upstream), a marked, near-coincident change in the rate of accumulation and in the grain size of material deposited occurred around 1980. This may be related to a combination of channel deepening and straightening operations undertaken in the mid-1970s and/or natural changes in winter wave climate.  相似文献

Fusion with a fertilizing spermatozoon induces the mammalian oocyte to undergo a remarkable series of oscillations in cytosolic Ca(2+), leading to oocyte activation and development of the embryo. The exact molecular mechanism for generating Ca(2+) oscillations has not been established. A sperm-specific zeta isoform of phospholipase C (PLCzeta) has been identified in mice. Mouse PLCzeta triggers Ca(2+) oscillations in mouse oocytes and exhibits properties synonymous with the 'sperm factor' that has been proposed to diffuse into the oocyte after gamete fusion. The present study isolated the PLCzeta homologue from human and cynomolgus monkey testes. Comparison with mouse and monkey PLCzeta protein sequences indicates a shorter X-Y linker region in human PLCzeta and predicts a distinctly different isoelectric point. Microinjection of complementary RNA for both human and cynomolgus monkey PLCzeta elicits Ca(2+) oscillations in mouse oocytes equivalent to those seen during fertilization in mice. Moreover, human PLCzeta elicits mouse egg activation and early embryonic development up to the blastocyst stage, and exhibits greater potency than PLCzeta from monkeys and mice. These results are consistent with the proposal that sperm PLCzeta is the molecular trigger for egg activation during fertilization and that the role and activity of PLCzeta is highly conserved across mammalian species.  相似文献

The report of work-related musculoskeletal pain may be related to worker evaluations of the design and management of work through two mechanisms: one biomechanical and the other stress-related. This study of engineering workers (n = 204) explored the validity of these mechanisms using sequential logistic regression. Analyses suggested that workers' ratings of the adequacy of the design and management of their work were related to their report of work-related musculoskeletal pain. However, the mechanisms appeared to be activated in certain conditions. The reporting of pain in the upper body was both biomechanically- and stress-related, whereas that in the lower body was only biomechanically-related. It is argued that the mechanism activated appeared to be determined by the anatomical location of the pain, and probably the variance shared between the different aspects of work design and management, on the one hand, and the mechanical load of the job, on the other.  相似文献

Inferior temporal cortex of squirrel monkeys consists of caudal (ITC), intermediate (ITI), and rostral (ITR) subdivisions, possibly homologous to TEO, posterior TE, and anterior TE of macaque monkeys. The present study compared visual learning in squirrel monkeys with ablations of ITC; ITI and ITR (group ITRd); or ITI, ITR, and more ventral cortex, including perirhinal cortex (group ITR+), with visual learning in unoperated controls. The ITC monkeys had significant impairments on pattern discriminations and milder deficits on delayed nonmatching to sample (DNMS) of objects. The ITRd monkeys had deficits on some pattern discriminations but not on DNMS. The ITRd monkeys were significantly impaired on DNMS and some pattern discriminations. Leachate levels within a landfill must often be controlled for environmental and/or regulatory reasons by means of pumping from wells. Conventional vertical wells are usually used for this purpose, but there is a perception that they are inefficient. In this paper, the feasibility of using directional drilling to install horizontal wells for leachate control in landfills is investigated with reference to pilot and full scale field trials at Rainham, U.K. The difficulties of well-screen design and installation in a landfilled waste are discussed; the insights gained during trial installation are described; and the effectiveness of three trial wells is assessed with reference to the leachate flow rates and drawdowns achieved, in comparison with conventional vertical wells. It is concluded that the drilling rig used must be sufficiently powerful to cope with the likelihood of at least partial borehole collapse around the well-screen during installation; that the screen slot size can be based on at least the D30 particle size of the waste and a natural filter allowed to develop around the well (provided that the resulting well screen is strong enough); and that as experience with the technology grows, directionally drilled horizontal wells could represent a viable, cost effective alternative to conventional vertical wells for leachate control in landfills.  相似文献
