Niobium(V) oxide (Nb2O5): application to phosphoproteomics   总被引:1,自引:0,他引:1  
Proteomics-based analysis of signaling cascades relies on a growing suite of affinity resins and methods aimed at efficient enrichment of phosphorylated peptides from complex biological mixtures. Given the heterogeneity of phosphopeptides and the overlap in chemical properties between phospho- and unmodified peptides, it is likely that the use of multiple resins will provide the best combination of specificity, yield, and coverage for large-scale proteomics studies. Recently titanium and zirconium dioxides have been used successfully for enrichment of phosphopeptides. Here we report the first demonstration that niobium pentoxide (Nb 2O 5) provides for efficient enrichment and recovery ( approximately 50-100%) of phosphopeptides from simple mixtures and facilitates identification of several hundred putative sites of phosphorylation from cell lysate. Comparison of phosphorylated peptides identified from Nb 2O 5 and TiO 2 with sequences in the PhosphoELM database suggests a useful degree of divergence in the selectivity of these metal oxide resins. Collectively our data indicate that Nb 2O 5 provides efficient enrichment for phosphopeptides and offers a complementary approach for large-scale phosphoproteomics studies.  相似文献

The effect of edible cassava starch–chitosan coatings incorporated with rosemary pepper (Lippia sidoides Cham.) essential oil and pomegranate peel extract on the shelf-life of tomatoes during storage at 25 °C for 12 days was investigated. Sixteen formulations, containing 10 g L?1 cassava starch and various concentrations of chitosan (5, 10, 20, 30 g L?1), essential oil (0, 2.5, 5, 10 mL L?1) and pomegranate peel extract (0, 5, 10, 20 mL L?1) were prepared and applied to tomatoes. Physical–chemical and microbiological analyses were performed on days 1, 4, 8 and 12. Most of the coatings delayed the ripening of tomatoes, lowering the total soluble solids (38?44 g sucrose kg?1) and weight loss (93?128 g kg?1) and maintaining constant firmness compared to the uncoated tomatoes (45 g sucrose kg?1, 175 g kg?1) at 12 days of storage. Conversely, except red intensity (a*), which was higher for the uncoated samples, the colour parameters (L*, b*) of the coated and control tomatoes were similar at the end of storage. Uncoated and coated tomatoes showed no contamination during storage. The coatings showed potential to maintain the quality of tomatoes during storage at 25 °C for 12 days. In this context, tomatoes coated with the formulation comprising 10 g L?1 cassava starch, 10 g L?1 chitosan, 10 mL L?1 essential oil and 20 mL L?1 pomegranate peel extract showed the lowest weight loss and reduced total soluble solids content compared with uncoated ones.  相似文献

Unlike other animals, human children will copy all of an adult's goal-directed actions, including ones that are clearly unnecessary for achieving the demonstrated goal. Here we highlight how social affiliation is key to this species-specific behavior. Preschoolers watched 2 adults retrieve a toy from a novel apparatus. One adult included irrelevant actions in her demonstration; the other only used actions causally related to opening the apparatus. After both adults took turns demonstrating, 1 left the test room, and the remaining adult gave the apparatus to the child. Children reproduced the irrelevant actions only when given the apparatus by the adult who had demonstrated them, even though the departed adult's actions emphasized how unnecessary these redundant actions were. Our results highlight the specialized skills for participating in cultural groups that have evolved in humans and provide insight into why finding such high fidelity copying in other animals has proven elusive. Increasing the growth performance of broiler chickens by supplementing their diets with exogenous enzymes can also contribute to positive changes in gut health. In this respect the growth of various bacteria normally associated with the gastrointestinal tract of poultry was assessed in vitro using a medium containing arabinoxylan, β‐glucan, guar gum and raffinose and their corresponding enzymes. Overall, enzymes releasing the largest amounts of free sugars yielded the largest increase in bacterial numbers. Accordingly, β‐glucan and raffinose treated with their respective enzymes promoted the largest number of bacterial types, reaching a minimum of 1.0 log10 population within 6 h at 40 °C. A broiler chicken growth trial was also conducted using wheat‐, barley‐ and corn‐based diets with and without enzyme and probiotic addition. Escherichia coli, coliforms, enterococci and aerobic and anaerobic sporeformers were monitored for growth in both the caecum and ileum. Enzyme supplementation reduced E. coli levels in the caecum of broilers fed wheat‐ or corn‐based diets. A further reduction in E. coli numbers was observed in broilers fed the same diets supplemented with a combination of enzyme and probiotic. Enzyme supplementation had much less of an effect on microbial populations in the ileum. Inclusion of probiotics reduced E. coli levels in the caecum and ileum but only in broilers fed wheat‐ and corn‐based diets. Anaerobic spore levels in the ileum increased in all diets containing probiotic. Overall, inclusion of enzymes or probiotics exhibited mixed effects on gut bacteria, depending on the nature of the carbohydrate source and enzyme. We report the results of the study of the polycrystalline powder of the diamond-lonsdaleite system by X-ray diffractometry, transmission electron microscopy and UV Raman spectroscopy. The measured data of structural parameters are in good agreement with ab initio calculations. We show that the Raman spectrum is proportional to the phonon density of states of the diamond-lonsdaleite system.  相似文献

The physical mechanism of the current flow in Pd-(heavily doped p-AlxGa1?x N) ohmic contact is studied. Chloride-hydride epitaxy was used to grow the p-Al0.06Ga0.94N solid solution with uncompensated acceptor concentration N a N d ranging from 3×1018 up to 1019 cm?3. Thermal vacuum deposition and subsequent thermal treatment were used to form an ohmic Pd contact. It is shown that, after the thermal treatment, the Pd-p-Al0.06Ga0.94N barrier contact with a potential barrier height of about 2.3 V becomes ohmic and the barrier height decreases to approximately 0.05 V. For uncompensated acceptor concentration N a N d =3×1018 cm?3, thermionic emission is found to be the main mechanism of the current through the Pd-p-Al0.06Ga0.94N ohmic contact. An increase in N a N d to approximately 1019 cm?3 in the solid solution leads to a transition from thermionic emission (at high temperatures) to tunneling (at low temperatures).  相似文献

For decades, it has been known that the tumor microenvironment is significant for glioma progression, namely the infiltration of myeloid cells like microglia and macrophages. Hence, these cell types and their specific tasks in tumor progression are subject to ongoing research. However, the distribution of the brain resident microglia and the peripheral macrophages within the tumor tissue and their functional activity are highly debated. Results depend on the method used to discriminate between microglia and macrophages, whereby this specification is already difficult due to limited options to distinguish between these both cell populations that show mostly the same surface markers and morphology. Moreover, there are indications about various functions of microglia and macrophages but again varying on the method of discrimination. In our review, we summarize the current literature to determine which methods have been applied to differentiate the brain resident microglia from tumor-infiltrated macrophages. Furthermore, we compiled data about the proportion of microglia and macrophages in glioma tissues and ascertained if pro- or anti-tumoral effects could be allocated to one or the other myeloid cell population. Recent research made tremendous efforts to distinguish microglia from recruited macrophages. For future studies, it could be essential to verify which role these cells play in brain tumor pathology to proceed with novel immunotherapeutic strategies.  相似文献

Linear optoelectronic amplification is investigated as an economic option for the provision of sufficient optical gain for fibre transmission systems of several hundred km lengths. Experimental results are presented demonstrating a system span capability in excess of 100 dB at 420 Mbit/s and 1.55 mu m wavelength with two intermediate linear repeaters.
