A laboratory experiment was used as a model to investigate interactions within a hyper-arid and hyper-saline weathering environment based on conditions observed in the Atacama Desert, northern Chile. The experimental design produced a matrix of 18 different weathering environments with six levels of salt (sodium nitrate) availability and three levels of fog precipitation. The experiment took place over an 11-week period. Over this time, 10 weekly fog applications modified the experimental conditions. From a theoretical perspective, the weekly fog applications generate an open rather than a closed system, which may be more representative of conditions in the field. The breakdown of the chalk blocks used was assessed in two ways. Firstly, qualitative observations were made at weekly intervals of the nature of morphological changes. Secondly, at the end of the experiment, breakdown was assessed by counting the number of discrete pieces greater than 1 g liberated by each block. The electrical conductivity at the base of each block was also measured. Collectively, the results indicate that fog appears to control the rate of breakdown and that salt availability controls the timing of the response. The style of breakdown was similar irrespective of the fog and salt levels. The morphological observations show the results of a sequence of events, with the blocks evolving as the experiment progresses; as such, they provide an experimental example of the role of inheritance in weathering processes.  相似文献

Leishmaniases are endemic in many countries, mainly in rural areas. In Brazil, Leishmania infection is responsible for many cases of Leishmaniases, including recent reports in urban regions. Despite their sensitivity, traditional serological and parasitological methods for detecting Leishmaniases have proven inadequate for species discrimination. This study aimed to identify Leishmania species in biological samples by a fast methodology, avoiding "in vitro" cultivation. Knowledge of the Leishmania species is an important tool in regions where both New World visceral leishmaniasis (VL) and cutaneous leishmaniasis (CL) are prevalent. As these new foci appear in areas not traditionally endemic for VL, the main problem is to distinguish between true autochthonous infections and infections acquired in other well-known endemic areas. Since, domestic dogs are known to be the main VL and CL reservoir, they are regularly investigated in endemic areas to prevent, principally, severe and often fatal VL in humans. However, several infected dogs present no clinical signs or clinical signs similar to other canine diseases. Here, we evaluated the ability of PCR to diagnose VL and distinguish L. (L.) chagasi from other Leishmania species in domestic dogs. Samples from 114 dogs from 30 cities (Sao Paulo, Brazil) were divided into two groups: 44 symptomatic and 70 asymptomatic. They were assayed by parasitological methods (culture and microscopic examination) and PCR to determine L. (L.) chagasi, L. (V.) braziliensis; and in some cases, Leishmania spp. Parasitological tests and PCR-L. chagasi were concordant in 105 samples (92%). VL was confirmed in 49 dogs, while 56 had negative results. Of the 114 samples, 9 had discordant results, but were further tested by PCR-Leishmania spp. with positive results. VL was also confirmed in 4 dogs having negative parasitological tests and positive PCR-L. chagasi. Consequently, this PCR was positive for 100% (53/49) of dogs with parasites detected in parasitological tests. Also, PCR demonstrated high specificity detecting 61 dogs negative for VL. Leishmania infection was negative in 56 dogs, and 5 with positive culture and PCR-Leishmania spp. had CL since they were positive in PCR-L. braziliensis. This study shows the importance of including PCR in diagnosis of Leishmaniases by differential diagnosis contributing to the surveillance and control of VL programs.  相似文献

Bovine rabies remains endemic in Brazil and despite control efforts, the disease still spreads insidiously. The main vector is the hematophagous bat, Desmodus rotundus. The present work aimed to create a predictive qualitative model of the occurrence of bovine rabies in each municipality in 25 of the 27 Brazilian States. The risk of rabies transmission from bats to bovine was estimated using decision-tree models of receptivity and vulnerability. Questionnaires, which covered a number of questions related to the surveillance of possible risk factors, such as bovine rabies outbreaks in the previous year, the presence of bat roosts, bat rabies positivity and environmental changes, were sent to the local veterinary units of each State. The bovine density and geomorphologic features were obtained from national databases and geographic information systems. Of the 433 municipalities presenting bovine rabies outbreaks in 2010, 178 (41.1%) were classified by the model as high risk, 212 (49.0%) were classified as moderate risk, 25 (5.8%) were classified as low risk, whereas the risk was undetermined in 18 municipalities (4.1%). An ROC curve was built to determine if the risk evaluated by the model could adequately discriminate between municipalities with and without rabies occurrence in future years. The risk estimator for the year 2011 was classified as moderately accurate. In the future, these models could allow the targeting of rabies control efforts, with the adoption of control measures directed to the higher risk locations and the optimization of the field veterinary staff deployment throughout the country. Additionally, efforts must be made to encourage continuous surveillance of risk factors.  相似文献

The herbicide glyphosate was sprayed aerially on a section of conifer forest in Atlantic Canada that had been previously clearcut and reforested. Glyphosate was then tested for effects on ammonification, nitrification, and denitrification for a period of 8 months by comparing microbial activity in treated and untreated zones of the clay loam forest soil and the overlying decomposing litter, both with a pH of 3.8. With ammonification, there was generally a stimulation of activity in both the forest litter (FL) and forest soil (FS) that had been exposed to glyphosate during spraying. Nitrification rates in FL and FS were very low and glyphosate had no appreciable stimulatory or inhibitory effect on nitrification. Although glyphosate stimulated denitrification in a few instances, it generally had no significant effect on denitrification activity in FL and FS exposed during spraying. With all processes, microbial activity in FL was significantly greater than that in FS. Laboratory bioassays were also performed with FL and FS, as well as two silt loam (pH 5.8 and 6.4) and one sandy loam (pH 6.8) agricultural soils, using glyphosate concentrations up to 200 times higher than field application rates. With ammonification and denitrification, glyphosate generally stimulated activity at all levels tested and in all soil used. Glyphosate stimulated ammonification by 50% at concentrations ranging from 140 to 550 μg g?1 for the soils and >4000 μg g?1 for FL. With denitrification, the corresponding herbicide levels were approximately 2250 μg g?1 for FS, > 10,000 for FL, and 450 for an agricultural soil. With nitrification, it was estimated that glyphosate concentrations greater than 1000 to 2000 μg g?1 would be required to cause a 50% inhibition of activity. The careful use of glyphosate in forestry should have no toxic effects on N cycling in soils.  相似文献

Flavonoid consumption via tea drinking has been attributed a number of potential health benefits including cancer prevention, anti-inflammatory action, and cardioprotectant activity. Although the predominant flavonoids in fresh leaf and green tea are known to be flavan-3-ols and flavan-3-O-gallates ("the catechins"), the biochemical effects of tea polyphenol consumption on living systems are generally poorly understood. Metabonomic methods utilizing (1)H NMR spectroscopy of biofluids and principal component analysis (PCA) have been applied to investigate the bioavailability and metabolic responses of rats to a single dose of 22 mg of epicatechin (EC) dissolved in water. Urine samples were collected twice daily (0-8 and 8-24 h) from male Sprague-Dawley rats (n = 10) prior to dosing and for 2 days after dosing. A series of subtle urinary biochemical effects were evident from the (1)H NMR spectra showing that EC was both bioavailable and biochemically active. The identifiable biochemical effects associated with EC dosing included decreased urinary concentrations of taurine, citrate, dimethylamine, and 2-oxoglutarate. These effects were predominately seen within the first 8 h after dosing. EC metabolites were also observed in the urine during this time period. PCA of later time points after dosing (24-32 and 32-48 h) showed that the effects of EC were reversible. This is the first in vivo study demonstrating the overall endogenous metabolic effects of EC consumption and shows the bioavailability of EC via metabolic effects and excretion of EC metabolites.  相似文献

An investigation of the ethanolic extract from stems of Caesalpinia echinata Lam (Leguminosae-Caesalpinioideae) led to the isolation of five new cassane diterpenes along with known lambertianic acid. Their structures were determined based on spectroscopic methods. A preliminary study on leishmanicidal activity demonstrated that compounds 1, 2 and 6 were found to inhibit the growth of amastigote-like forms of Leishmania amazonensis without affecting mononuclear cells obtained from human peripheral blood.  相似文献

OBJECTIVE: To develop a standard method for measuring tear production in large Psittaciformes using the phenol red thread (PRT) tear test. To establish mean PRT tear test values in clinically normal pet birds and to assess the reproducibility of the test. ANIMALS: Two geographically distinct populations of healthy, large Psittaciformes (pilot study and study population). Species groups include: cockatoo (17), macaw (22), Amazon (10), African gray (1), eclectus (1) and pionus (1). PROCEDURE: The PRT was placed under the upper eyelid and both eyelids were held closed throughout the test. The PRT tear test was repeated after 2 months in the pilot study and after approximately 5 min in the study population. RESULTS: The mean PRT tear test values for the pilot study were OD=28.2 mm/15 s (+/- 6.3 mm), OS=24.1 mm/15 s (+/- 6.6 mm) and OD=25.4 mm/15 s (+/- 3.3 mm), OS=25.5 mm/15 s (+/- 5.2 mm), for the first and second visits respectively. The mean PRT tear test value for the study population was: OD=19.8 mm/15 s (+/- 4.3 mm), OS=20.1 (+/- 3.9 mm) and OD=20.0 mm/15 s (+/- 4.5 mm), OS=19.1 mm/15 s (+/- 3.3 mm), for the first and second tests respectively. There was poor repeatability between tests. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the PRT tear test is a practical method for measuring tear production in large Psittaciformes, although adaptations to the established diagnostic method are necessary to overcome some anatomical differences present in birds. Good reproducibility of the PRT tear test could not be demonstrated in a clinically normal population of birds but geographic location appears to significantly influence results.  相似文献

Summary Field trials were carried out in Ecuador with two indigenous communities, Ninín Cachipata and La Esperanza, to determine farmers’ preferences for quinoa (Chenopodium quinoa Willd.) cultivars and to improve PPB processes. More women than men participated, reflecting that quinoa, a primarily subsistence crop, is mainly managed by women. Farmers’ field selection criteria for quinoa in the field were mostly based on yield, earliness and plant colour; however only breeders’ measurements of yield and panicle height significantly correlated to farmer selection scores. Older women gave higher scores than younger women or men, apparently due to a concept of no cultivar being without value. Working in same gender pairs improved evaluation richness. INIAP technicians were more discriminating in their evaluations than farmers. They also used additional selection criteria of disease resistance and uniformity. At seed selection, farmers from Ninín Cachipata, where food security is not assured, chose lines based on yield, while farmers from La Esperanza, where resources are less limiting, also considered seed size, colour, saponin content and marketability. Field characteristics were not taken into consideration at seed selection, signifying that farmers are less interested in those characteristics, or that it was difficult for them to correlate field data when presented in tabular form with seed characteristics. Future trials with small farmers should have fewer lines or replications to avoid farmer fatigue during evaluation. Farmers who grow primarily for subsistence in semi-arid environments have more interest in growing quinoa, and more to gain from having improved cultivars; therefore future participatory efforts should focus on them.  相似文献
