Nonylphenol is present in surface water and aquatic sediments and because of its lipophilic characteristics shows a considerable potential to bioaccumulate in aquatic organisms. Nonylphenol inhibits testicular growth and induces vitellogenin synthesis in male rainbow trout. In order to better understand the effects of nonylphenol on fish and its impact in the aquatic environment, it is essential to elucidate the metabolic fate of this compound. A single oral dose (5 mg, 1850 KBq) of [3H]4-n-nonylphenol resulted in 1.1% and 3.0% of the ingested radioactivity eliminated in urine after 24 and 48 h, respectively. Four metabolites were separated by radio-HPLC and tentatively identified by mass spectrometry. Urinary metabolites likely resulted from the initial omega-oxidation of 4-n-nonylphenol to the putative 9-(4-hydroxyphenyl)-nonanoic acid which subsequent beta-oxidation led to 4-hydroxybenzoic acid as major metabolite. Intermediary metabolites, namely 3-(4-hydroxyphenyl) propionic acid and 3-(4-hydroxyphenyl)-2-propenoic acid confirmed the occurrence of this beta-oxidative pathway. Urinary metabolites identified in this study were quite different from biotransformation products previously described in bile of trout treated with 4-n-nonylphenol.  相似文献

Dihydroxyacetone kinase (DAK) is essential for methanol assimilation in methylotrophic yeasts. We have cloned the DAK gene from Pichia pastoris by functional complementation of a mutant that was unable to grow on methanol. An open reading frame of 1824 bp was identified that encodes a 65·3 kDa protein with high homology to DAK from Saccharomyces cerevisiae. Although DAK from P. pastoris contained a C-terminal tripeptide, TKL, which we showed can act as a peroxisomal targeting signal when fused to the green fluorescent protein, the enzyme was primarily cytosolic. The TKL tripeptide was not required for the biochemical function of DAK because a deletion construct lacking the DNA encoding this tripeptide was able to complement the P. pastoris dakΔ mutant. Peroxisomes, which are essential for growth of P. pastoris on methanol, were present in the dakΔ mutant and the import of peroxisomal proteins was not disturbed. The dakΔ mutant grew at normal rates on glycerol and oleate media. However, unlike the wild-type cells, the dakΔ mutant was unable to grow on methanol as the sole carbon source but was able to grow on dihydroxyacetone at a much slower rate. The metabolic pathway explaining the reduced growth rate of the dakΔ mutant on dihydroxyacetone is discussed. This work is devoted to a presentation of the ECOCLIMAP-II database for Western Africa, which is an upgrade for this region of the former initiative, ECOCLIMAP-I, implemented at global scale. ECOCLIMAP-II is a dual database at 1-km resolution that comprises an ecosystem classification and a coherent set of land surface parameters. This new physiographic information (e.g. leaf area index, fractional vegetation cover, albedo and land cover classification), was especially developed in the framework of the African Monsoon Multidisciplinary Analysis (AMMA) programme in order to support the modelling of land-atmosphere interactions, which stresses the importance of the present study. Criteria for coherence between prevalent land cover classifications and the analysis of time series of the satellite leaf area index (LAI) between 2000 and 2007 constitute the analysis tools for setting up ECOCLIMAP-II. The LAI and inferred fraction of vegetation cover are spatially distributed per land cover unit. The fraction of vegetation cover is handled to split the land surface albedo into vegetation and bare soil albedo components, as is required for a large number of applications. The new ECOCLIMAP-II land cover product is improved with regard to the spatial coherence compared to former version. The reliability of the physiographic details is also confirmed through verification with land cover products at higher resolution.  相似文献

The goal of this study is to propose a new classification of African ecosystems based on an 8-year analysis of Normalized Difference Vegetation Index (NDVI) data sets from SPOT/VEGETATION. We develop two methods of classification. The first method is obtained from a k-nearest neighbour (k-NN) classifier, which represents a simple machine learning algorithm in pattern recognition. The second method is hybrid in that it combines k-NN clustering, hierarchical principles and the Fast Fourier Transform (FFT). The nomenclature of the two classifications relies on three levels of vegetation structural categories based on the Land Cover Classification System (LCCS). The two main outcomes are: (i) The delineation of the spatial distribution of ecosystems into five bioclimatic ecoregions at the African continental scale; (ii) Two ecosystem maps were made sequentially: an initial map with 92 ecosystems from the k-NN, plus a deduced hybrid classification with 73 classes, which better reflects the bio-geographical patterns. The inclusion of bioclimatic information and successive k-NN clustering elements helps to enhance the discrimination of ecosystems. Adopting this hybrid approach makes the ecosystem identification and labelling more flexible and more accurate in comparison to straightforward methods of classification. The validation of the hybrid classification, conducted by crossing-comparisons with validated continental maps, displayed a mapping accuracy of 54% to 61%.  相似文献

The conductivity of CoPc (cobalt phthalocyanine) and Co[(SO3Na)2,3Pc] was measured under a flow of two different gases (NH3 and O3), during exposure/recovery cycles. It appears that the relative responses are linearly related to the concentration, in the 20-200 ppb range for O3 and in the 20-200 ppm range for NH3. Observed during time, the sensing parameters allow a qualitative understanding of the kinetics. The comparative study of those products under both different gases gave interesting results for sensor applications. Whereas CoPc is sensitive to both gases, its sulfonated counterpart is only sensitive to NH3.  相似文献

Blocking the adherence of bacteria to cells is an attractive complementary approach to current antibiotic treatments, which are faced with increasing resistance. This strategy has been particularly studied in the context of urinary tract infections (UTIs), in which the adhesion of pathogenic Escherichia coli strains to uroepithelial cells is prevented by blocking the FimH adhesin expressed at the tips of bacteria organelles called fimbriae. Recently, we extended the antiadhesive concept, showing that potent FimH antagonists can block the attachment of adherent‐invasive E. coli (AIEC) colonizing the intestinal mucosa of patients with Crohn′s disease (CD). In this work, we designed a small library of analogues of heptyl mannoside (HM), a previously identified nanomolar FimH inhibitor, but one that displays poor antiadhesive effects in vivo. The anomeric oxygen atom was replaced by a sulfur or a methylene group to prevent hydrolysis by intestinal glycosidases, and chemical groups were attached at the end of the alkyl tail. Importantly, a lead compound was shown to reduce AIEC levels in the feces and in the colonic and ileal mucosa after oral administration (10 mg kg?1) in a transgenic mouse model of CD. The compound showed a low bioavailability, preferable in this instance, thus suggesting the possibility of setting up an innovative antiadhesive therapy, based on the water‐soluble and non‐cytotoxic FimH antagonists developed here, for the CD subpopulation in which AIEC plays a key role.  相似文献
